The use of a column containing 60 g of silica gel for cleanup and the use of isobutane as a reactant reagent for chemical ionization-mass spectrometric analysis of the saturated and monounsaturated alkyl side-chain 2-alkylcyclobutanones (2-ACBs; specifically induced by irradiation from fat in foods until the proof of contrary) has improved both the sensibility and the selectivity of the method when applied for the detection of irradiated foods. The quality of the chromatograms obtained was improved, allowing the detection of food samples (avocados) irradiated at low doses (0.1 kGy) or irradiated ingredients included in low proportions (less than 5%, wt/wt) in nonirradiated culinary foods. These analytical modifications for the detection of 2-ACBs on the official EN 1785 method enable an extension of its current field of application using common equipment of food quality control laboratories.  相似文献

Variants of sperm whale myoglobin (Mb) were used to assess the mechanism of heme protein-mediated lipid oxidation in washed cod muscle. A myoglobin variant with high hemin affinity (V68T) was an exceptionally poor promoter of lipid oxidation, while a Mb variant with low hemin affinity (H97A) was a potent promoter of lipid oxidation. V68T releases hemin slowly due to the ability of threonine to hydrogen bond with coordinated water and the distal histidine within the heme crevice. H97A rapidly releases hemin because the relatively small alanine residue creates a channel for water to easily enter the heme crevice which weakens the covalent linkage of hemin to the proximal histidine. A variant sensitive to heme degradation (L29F/H64Q) was a weaker promoter of lipid oxidation compared to wild-type Mb. This suggests that degrading the heme ring and releasing iron decreased the ability of Mb to promote lipid oxidation. Free radicals resulting from hemin-mediated decomposition of lipid hydroperoxides have the capacity to propagate lipid oxidation and degrade hemin catalyst. This may explain why heme proteins behave as reactants rather than "catalysts" of lipid oxidation in washed cod. Collectively these studies strongly suggest that released hemin is the critical entity that drives heme protein-mediated lipid oxidation in washed fish muscle.  相似文献

Plants link atmospheric and soil carbon pools through CO2 fixation, carbon translocation, respiration and rhizodeposition. Within soil, microbial communities both mediate carbon-sequestration and return to the atmosphere through respiration. The balance of microbial use of plant-derived and soil organic matter (SOM) carbon sources and the influence of plant-derived inputs on microbial activity are key determinants of soil carbon-balance, but are difficult to quantify. In this study we applied continuous 13C-labelling to soil-grown Lolium perenne, imposing atmospheric CO2 concentrations and nutrient additions as experimental treatments. The relative use of plant- and SOM-carbon by microbial communities was quantified by compound-specific 13C-analysis of phospholipid fatty acids (PLFAs). An isotopic mass-balance approach was applied to partition the substrate sources to soil respiration (i.e. plant- and SOM-derived), allowing direct quantification of SOM-mineralisation. Increased CO2 concentration and nutrient amendment each increased plant growth and rhizodeposition, but did not greatly alter microbial substrate use in soil. However, the increased root growth and rhizosphere volume with elevated CO2 and nutrient amendment resulted in increased rates of SOM-mineralisation per experimental unit. As rhizosphere microbial communities utilise both plant- and SOM C-sources, the results demonstrate that plant-induced priming of SOM-mineralisation can be driven by factors increasing plant growth. That the balance of microbial C-use was not affected on a specific basis may suggest that the treatments did not affect soil C-balance in this study.  相似文献

Oil-in-water emulsions containing cationic droplets stabilized by lecithin-chitosan membranes were produced using a two-stage process. A primary emulsion was prepared by homogenizing 5 wt % corn oil with 95 wt % aqueous solution (1 wt % lecithin, 100 mM acetic acid, pH 3.0) using a high-pressure valve homogenizer. This emulsion was diluted with aqueous chitosan solutions to form secondary emulsions with varying compositions: 1 wt % corn oil, 0.2 wt % lecithin, 100 mM acetic acid, and 0-0.04 wt % chitosan (pH 3.0). The particle size distribution, particle charge, and creaming stability of the primary and secondary emulsions were measured. The electrical charge on the droplets increased from -49 to +54 mV as the chitosan concentration was increased from 0 to 0.04 wt %, which indicated that chitosan adsorbed to the droplet surfaces. The mean particle diameter of the emulsions increased dramatically and the emulsions became unstable to creaming when the chitosan concentration exceeded 0.008 wt %, which was attributed to charge neutralization and bridging flocculation effects. Sonication, blending, or homogenization could be used to disrupt flocs formed in secondary emulsions containing droplets with high positive charges, leading to the production of emulsions with relatively small particle diameters (approximately 1 microm). These emulsions had good stability to droplet aggregation at low pH (< or =5) and ionic strengths (<500 mM). The interfacial engineering technology utilized in this study could lead to the creation of food emulsions with improved stability to environmental stresses.  相似文献

The objective of this study is to examine the influence different zooplankon grazing rate relationships have on water quality eutrophication model results in a tidal estuary. The water quality model being used includes phytoplankton and both dissolved and particulate constituents as state variables. It includes zooplankton grazing of the phytoplankton, where excretions from the latter increase the particulate constituents. The phytoplankton and particulate constituents also undergo settling. When applied to an existing estuary, the model results are different for a first order zooplankton grazing rate formulation than for a second order zooplankton grazing rate formulation.Constituents enter the estuary as wastewater discharges, freshwater river inflows, exchanges between the sediment and water interface, and exchange across the tidal boundary. The water quality model is coupled with a three-dimensional time varying hydrodynamic and transport model that allows accurate computation of the tidal velocity field transporting the constituents from their sources throughout the estuary. Comparisons of model results between the first order and the second order density dependent zooplankton grazing relationships allow determining which of the two processes best describe water quality behavior for the estuary being examined. The second order density dependent zooplankton grazing relationship performs best in application.  相似文献

Blue honeysuckle (Lonicera caerulea L.) is a northern-adapted crop species with extremely early phenology and cold hardiness. Restricted adaptation to temperate climates is a current limitation to the crop’s mainstream potential for large-scale production. Based on the broad germplasm base at the University of Saskatchewan, vegetative growth cessation and leaf drop phenology were characterized to complement analysis of spring phenological adaptation in a temperate climate. A multi-trial site of three foundation groups and a single-site trial of three improved groups compared with their parental foundation genotypes were conducted in the Fraser Valley, British Columbia, Canada in 2012 and 2013. The current study shows that blue honeysuckle germplasm contains considerable phenological variation that will permit adaptation to temperate climates. It also demonstrates that breeding for improved adaptation is possible using the existing germplasm resources. For Russian germplasm with the earliest phenology, adaptation to northern latitudes and short growing seasons results in early floral initiation, which presents an indirect limitation to production in more southern latitudes due to secondary flowering. Early leaf drop indirectly limits production and is related to poor adaptation to high chill environments. Skew toward later leaf drop in improved groups, produced from hybridization with Japanese and Kuril germplasm with intermediate and late phenologies, respectively, shows that development of better adaptation to southern latitudes can be achieved.  相似文献

The greenhouse gases CO2 and N2O emissions were quantified in a long-term experiment in northern France, in which no-till (NT) and conventional tillage (CT) had been differentiated during 32 years in plots under a maize–wheat rotation. Continuous CO2 and periodical N2O soil emission measurements were performed during two periods: under maize cultivation (April 2003–July 2003) and during the fallow period after wheat harvest (August 2003–March 2004). In order to document the dynamics and importance of these emissions, soil organic C and mineral N, residue decomposition, soil potential for CO2 emission and climatic data were measured. CO2 emissions were significantly larger in NT on 53% and in CT on 6% of the days. From April to July 2003 and from November 2003 to March 2004, the cumulated CO2 emissions did not differ significantly between CT and NT. However, the cumulated CO2 emissions from August to November 2003 were considerably larger for NT than for CT. Over the entire 331 days of measurement, CT and NT emitted 3160 ± 269 and 4064 ± 138 kg CO2-C ha−1, respectively. The differences in CO2 emissions in the two tillage systems resulted from the soil climatic conditions and the amounts and location of crop residues and SOM. A large proportion of the CO2 emissions in NT over the entire measurement period was probably due to the decomposition of old weathered residues. NT tended to emit more N2O than CT over the entire measurement period. However differences were statistically significant in only half of the cases due to important variability. N2O emissions were generally less than 5 g N ha−1 day−1, except for a few dates where emission increased up to 21 g N ha−1 day−1. These N2O fluxes represented 0.80 ± 0.15 and 1.32 ± 0.52 kg N2O-N ha−1 year−1 for CT and NT, respectively. Depending on the periods, a large part of the N2O emissions occurred was probably induced by nitrification, since soil conditions were not favorable for denitrification. Finally, for the period of measurement after 32 years of tillage treatments, the NT system emitted more greenhouses gases (CO2 and N2O) to the atmosphere on an annual basis than the CT system.  相似文献

The 2-alkylcyclobutanones (2-ACBs) are formed from triglycerides by irradiation treatment and may be used as markers for this type of food processing. This paper describes a detection method for the analysis of monounsaturated alkyl side chain 2-ACBs, which is formed upon irradiation from monounsaturated fatty acids which frequently are the most abundant fatty acids in foods. The estimated radioproduction yields of the cis-2-(dodec-5'-enyl)-cyclobutanones (cis-2-dDeCB) and the cis-2-(tetradec-5'-enyl)-cyclobutanones (cis-2-tDeCB) were 1.0 +/- 0.5 and 0.9 +/- 0.2 nmol.mmol(-1) precursor fatty acid.kGy(-1), respectively, being similar to that of saturated 2-ACBs. The stability study of the s- and mu-2-ACBs in poultry meat samples irradiated at 10 kGy and stored for 3-4 weeks at 4 degrees C and 25 degrees C showed that these compounds undergo some transformation, their amounts being reduced by about 50%. These storage losses did not depend on the saturation state of the alkyl side chain. The EI-MS detection limit of 2-tDeCB is 3 times higher (0.6 pmol) than that of 2-dodecylcyclobutanone (0.2 pmol). Consequently, when the oleic acid content of the analyzed food exceeds the content of palmitic acid by a factor of 3, it would be of an advantage to apply 2-tDeCB as a marker for detection of the irradiation treatment.  相似文献

Augmentation of small rare species populations is sometimes suggested on genetic grounds. However, outbreeding depression via dilution of local adaptation or break-up of genomic coadaptation may occur. These effects depend on the causes of population divergence. Here, we compare genetic measures of population divergence in Liatris scariosa var. novae-angliae, a rare New England perennial. We measured GST, neutral marker subdivision, and QST, quantitative subdivision of propagule and juvenile plant traits. GST was relatively high. QST for leaf shape exceeded GST, indicating local adaptation, while QST for other traits fell within or below the GST range. Local adaptation appears low for juvenile traits, although the high GST cautions against translocation because of potential coadaptation. If translocation is still required, however, donor populations should contain high quantitative genetic diversity. We assess population size and allozyme diversity as predictors of quantitative genetic variation, but find these poor proxies for direct measurement.  相似文献
