Live frozen vaccines containing Babesia bovis, Babesia bigemina or Anaplasma centrale were prepared using glycerol as cryoprotectant and stored in liquid nitrogen. The viability of the vaccines was tested inoculating calves 1 h (n = 12), 2 h (n = 12), 12 h (n = 6) and 24 h (n = 6) after thawing. Babesia bovis and A. centrale were detected in thin and/or thick blood smears in all vaccinated calves; however, 1 of 12 calves inoculated 1 h after thawing and 3 of 6 calves inoculated 24 h after thawing did not develop a B. bigemina parasitaemia. The longer post-thawing durability of frozen vaccines cryoprotected with glycerol compared with those cryoprotected with dimethyl sulfoxide, presented by other authors, will extend their use under field conditions. 相似文献

A 1-year-old Australian Shepherd (AS) was presented for a routine hereditary eye examination. During the examination multiple raised, brown to orange lesions were noted in the fundus, which could not be attributed to a known retinal disease in this breed. As they clinically most closely resembled canine multifocal retinopathy (cmr) and no indication of an acquired condition was found, genetic tests for BEST1 gene mutations were performed. These showed the dog to be homozygous for the cmr1 (C73T/R25X) gene defect. Furthermore, ultrasound (US), electroretinography (ERG), and optical coherence tomography were performed, confirming changes typical for cmr. Subsequently, the AS pedigree members were genetically and clinically tested, demonstrating autosomal recessive inheritance with no clinical symptoms in carrier animals, as was previously described for cmr. To our knowledge, this is the first reported case of canine multifocal retinopathy in the AS breed. Further investigations are under way. 相似文献

The interaction of Tritrichomonas foetus with its host is a complex process that involves colonisation, attachment and persistence. The goal of the present study was to describe the interaction of T. foetus with the genital tract using a model of non-oestrogenised female BALB/c mice which had been intravaginally infected with a suspension of T. foetus during oestrus. Animals were sacrificed after 10 weeks and the uteri fixed and processed for light and electron microscopy. Ultrastructural analysis showed that the attached protozoa interacted with the mucosa through a somal projection. With an amorphous secretion at the protozoa-host cell interface. There was no direct contact between the protozoal plasma membrane and the epithelial cell membrane. Our results demonstrated the participation of an active phagocytosis and the destruction of T. foetus by eosinophils. 相似文献

Gravitational lensing is a powerful astrophysical and cosmological probe and is particularly valuable at submillimeter wavelengths for the study of the statistical and individual properties of dusty star-forming galaxies. However, the identification of gravitational lenses is often time-intensive, involving the sifting of large volumes of imaging or spectroscopic data to find few candidates. We used early data from the Herschel Astrophysical Terahertz Large Area Survey to demonstrate that wide-area submillimeter surveys can simply and easily detect strong gravitational lensing events, with close to 100% efficiency. 相似文献

A feeding trial was conducted to determine the minimum dietary vitamin C requirement of juvenile red drum and characterize signs of vitamin C deficiency. Semipurified diets containing 400 g crude protein kg–1 from lyophilized red drum muscle and crystalline amino acids were used in the feeding trial. The basal diet without supplemental vitamin C contained ≈ 6 mg vitamin C kg–1 and was fed for a 1-week conditioning period. Red drum, initially averaging ≈ 3.6 g each, were fed diets supplemented with 0, 10, 20, 30, 45, 60, 75, or 150 mg vitamin C kg–1 as ascorbate polyphosphate for a period of 10 weeks. Fish fed the basal diet began to exhibit overt signs of vitamin C deficiency, including reduced weight gain, lordosis, scoliosis and loss of equilibrium after 8 weeks. Total ascorbate was undetectable in liver of fish fed the basal diet while liver ascorbate levels in fish fed the other diets generally reflected dietary supplementation. Regression analysis of weight gain data using the broken-line model resulted in a minimum vitamin C requirement ( ± SE) of 15 ± 3 mg kg–1 diet. 相似文献

Gonadic conditioning and maturation of queen clam Dosinia ponderosa fed three microalgae (Isochrysis galbana, Tetraselmis suecica and Chaetoceros calcitrans) at three temperatures (20, 25 and 30°C) were assessed. Histological analysis showed six gonadic stages, indicators of maturity. The evaluation included sex proportion, oocyte diameter and frequency of gonadic stages after 22 (middle of trial) and 44 days (end of trial). At day 22, around 60% of females conditioned at 20°C, reached stage III (advances grade of maturation) when fed T. suecica. At day 44, around 50% of females were on stage III, conditioned at 20 and 25°C and fed T. suecica and C. calcitrans. For males, the best combination was C. calcitrans at 20 and 25°C. The temperature of 30°C resulted inadequate for maturation, especially when fed the diatom Isochrysis galbana. High mortalities occurred at that temperature with the three microalgal diets, especially in females. The results permits conclude that T. suecica and C. calcitrans are good sources of feed for D. ponderosa and that the best range of temperature for gonadic development is 22–25°C.
