  1. Monitoring the abundance of mobile and wide‐ranging cetacean populations for conservation management is challenging, especially when the management is focused on static protected areas. Where abundance estimates are derived from mark–recapture data, such as photo‐identification of naturally marked individuals, unpredictable movements of animals in and out of the survey area can reduce ‘capture’ probabilities and affect the precision and accuracy of resulting estimates.

A Bayesian hierarchical log–linear model was applied to photo‐identification data collected in summer 2014 to derive a multi‐site abundance estimate for a population of bottlenose dolphins, Tursiops truncatus, ranging widely throughout the coastal waters of western Ireland. In addition, the effects of varying levels of sampling effort on the minimum detectable decrease in population size were examined.
  3. The abundance (median) of dolphins was estimated as 189 (coefficient of variation (standard deviation/mean), 0.11; 95% highest‐posterior density interval, 162–232). Over 50% of the well‐marked dolphins encountered throughout the study were sighted in more than one distinct coastal area, thus displaying high mobility. In addition, it was found that it would require biennial surveys to detect a 25% decline in abundance within the six‐year reporting period of the EU’s Habitats Directive.

Given that the Special Area of Conservation designated for these dolphins consists of two separate areas covering a substantial portion of the west coast of Ireland, the multisite approach is appropriate for monitoring this population. It produces a more precise estimate and is well suited for sparse recapture data collected opportunistically at multiple sites, when the lack of resources prevents large‐scale surveys or when concentrating surveys on smaller localized areas fails to capture the broad range and unpredictable occurrence of the animals. The Bayesian multi‐site approach could be applied to the management of other wide‐ranging marine or terrestrial taxa.

Background

Failure of passive transfer of maternal immunity via colostrum can occur in the bovine, and a number of blood tests have been developed to test calves for this failure. It is not clear which test is most suitable for this purpose. The objective was to examine the most commonly used tests for failure of passive transfer and to decide which is most suitable for routine laboratory use. 126 serum samples were taken from calves of dairy cows after birth but prior to colostrum feeding, and at 48 h of age. Five different tests were compared against radial immunodiffusion which is considered the appropriate reference method. These tests were serum gamma-glutamyltransferase levels, serum protein levels, serum globulin levels, an enzyme linked immunosorbent assay and the zinc sulphate turbidity test.

Results

The tests examined displayed high sensitivity but widely varying specificity. Examination of the use of different cut-off points allowed some improvement in specificity at the expense of sensitivity, but the tests which had performed best at the original cut-off points still displayed the best performance. Gamma-glutamyltransferase levels as a measure of colostrum absorption returned, in this study, the best balance between sensitivity and specificity. The ELISA used in this study and serum globulin levels displayed performance similar to the gamma-glutamyltransferase levels. Serum total protein was less successful than others examined at providing both sensitivity and specificity but may, when performed via refractometer, be useful for on-farm testing. As currently performed the poor sensitivity for which the zinc sulphate turbidity test is most often criticized is evident. Modification of the cut-off point to increase specificity is less successful at balancing these parameters than the ELISA, gamma-glutamyltransferase levels, and globulin levels.

Conclusions

Gamma-glutamyltransferase levels, ELISA testing and circulating globulin levels performed best in detecting failure of passive transfer in serum samples, although all three had some practical considerations.  相似文献

Scientists feel discomfort when they are asked to create certainty, where none exists, for use as an alibi in policy‐making. Recently, the scientific literature has drawn attention to some pitfalls of simulation‐based fisheries management‐strategy evaluation (MSE). For example, while estimates concerning central tendencies of distributions of simulation outcomes are usually fairly robust because they are conditioned on ample data, estimates concerning the tails of distributions (such as the probability of falling below a critical biomass) are usually conditional on few data and thus often rely on assumptions that have no strong knowledge base. The clients of scientific advice, such as the European Commission, are embracing the mechanization of the evaluation of proposed Harvest Control Rules against the precautionary principle and management objectives. Where the fisheries management institutions aim for simple answers from the scientists, giving ‘green/red light’ to a proposed management strategy, the scientists are forced into a split position between satisfying the demands of their advisory role and living up to the standards of scientific rigour. We argue against the mechanization of scientific advice that aims to incorporate all relevant processes into one big model algorithm that, after construction, can be run without circumspection. We rather encourage that fisheries advice should be a dynamic process of expert judgement, incorporating separate parallel concurrent, lines of scientific evidence, from quantitative and qualitative modelling exercises and factual knowledge of the biology and the fishery dynamics. This process can be formalized to a certain degree and can easily accommodate stakeholder viewpoints.  相似文献

The daily dynamics (during 33 days) of the number (colony-forming units (CFU)) of nitrogen-fixing bacteria and of the nitrogen-fixing activity (the acetylene method) were determined in a gray forest soil under a fallow land and under an intensely cultivated field. The daily dynamics of the CFUs determined on the nitrogen-free medium in the samples from both plots had wavelike patterns. The daily values of the actual and potential activities of nitrogen fixation in the samples from the fallow land plot and of the actual activity of nitrogen fixation in the samples from the intensely cultivated soil were low and close to the detection limit. The potential activity of nitrogen fixation in the intensely cultivated soil was significant and also had a wavelike pattern. The harmonic analysis of the daily dynamics of the CFUs and nitrogen fixation showed the statistically significant harmonics of these biological characteristics pointing to the objective and regular character of the wavelike dynamics. The revealed dynamics of the biological characteristics of the soils and the methods of their analysis are important in terms of the comparative study of the biological properties of different soils.  相似文献

Macroalgae, or seaweeds, are a rich source of components which may exert beneficial effects on the mammalian gut microbiota through the enhancement of bacterial diversity and abundance. An imbalance of gut bacteria has been linked to the development of disorders such as inflammatory bowel disease, immunodeficiency, hypertension, type-2-diabetes, obesity, and cancer. This review outlines current knowledge from in vitro and in vivo studies concerning the potential therapeutic application of seaweed-derived polysaccharides, polyphenols and peptides to modulate the gut microbiota through diet. Polysaccharides such as fucoidan, laminarin, alginate, ulvan and porphyran are unique to seaweeds. Several studies have shown their potential to act as prebiotics and to positively modulate the gut microbiota. Prebiotics enhance bacterial populations and often their production of short chain fatty acids, which are the energy source for gastrointestinal epithelial cells, provide protection against pathogens, influence immunomodulation, and induce apoptosis of colon cancer cells. The oral bioaccessibility and bioavailability of seaweed components is also discussed, including the advantages and limitations of static and dynamic in vitro gastrointestinal models versus ex vivo and in vivo methods. Seaweed bioactives show potential for use in prevention and, in some instances, treatment of human disease. However, it is also necessary to confirm these potential, therapeutic effects in large-scale clinical trials. Where possible, we have cited information concerning these trials.  相似文献
