The association of lactic acid bacteria with mucosal surfaces plays important roles in the beneficial effects of these bacteria on human health, such as colonization of the gastrointestinal tract for pathogen antagonism. Previously, we found that the adhesion of Lactococcus lactis 7‐1 to porcine gastric mucin was higher with fructose than with lactose, galactose or xylose as the carbon source. In this study, we examined the effect of growth conditions on the adhesion of strain 7‐1 grown on fructose. Medium components affect the adhesion: the adhesion of strain 7‐1 grown with sodium acetate was higher than that without it. The enhancement of adhesion by sodium acetate was not observed under aerobic conditions. Cellular properties grown with or without sodium acetate were characterized: strain 7‐1 grown with sodium acetate had similar sugar contents, and different fatty acid composition to those grown without it. Strain 7‐1 grown with sodium acetate showed significantly lower cell yield and significantly higher hydrophobicity than those grown without it, which is associated with higher adhesion. Fructose and sodium acetate are frequently used in the food industry; this study may reveal a simple way to enhance the adhesion of lactic acid bacteria by growing them with these substances. 相似文献

SummaryTo control the bolting of Japanese bunching onion (Allium fistulosum L.) photoperiodically, the effect of photoperiods before, during and after vernalization on flower initiation and development and the varietal differences were investigated using the two mid-season flowering cvs Kincho and Asagi-kujo, and a late-season flowering cv. Cho-etsu. A long-day photoperiod (LD, 16 h) given before vernalization inhibited flower initiation. Especially, the bolting rate of ‘Asagi-kujo’ decreased by about a half, compared with the short-day photoperiod (SD, 8 h). The interaction between the effect of night temperature (3°C, 7°C, 11°C or 15°C) and the effect of the photoperiod (SD and LD) during vernalization was also investigated. In ‘Kincho’, LD did not affect flower initiation at 3°C, but inhibited flower initiation at 7°C, 11°C and 15°C. In ‘Asagi-kujo’, flower initiation was significantly inhibited by LD under all temperature conditions. This inhibitory effect was stronger at 11°C and 15°C than at 3°C and 7°C. In ‘Cho- etsu’, LD significantly inhibited flower initiation at 3°C and 7°C, and flower initiation rarely occurred at 11°C and 15°C. In this study, generally, LD during vernalization inhibited flower initiation in all cultivars. Thus Japanese bunching onion required a short-day photoperiod in flower initiation, which was stronger in ‘Asagi-kujo’ and ‘Cho-etsu’ than in ‘Kincho’. From these results, we conclude that low temperature and a short-day photoperiod complementarily induce flower initiation in Japanese bunching onion. Varietal differences exist in the requirement of low temperature and a short-day photoperiod: the primary requirement in ‘Kincho’ is low temperature and that in ‘Asagi-kujo’ is a short-day. After flower initiation, the early stage of flower development is day-neutral, and after the floret formation stage, a long-day photoperiod promotes flower development and elongation of the seedstalk. 相似文献

The thermoplastic flow behavior of cedar flour steamed at different temperatures in the range of 160–220°C was measured using a rheometer and compared with that of beech flour. The temperature at which the cedar flour starts to flow was approximately 70°C higher than that of beech flour, and the cedar flour exhibited low flowability. Furthermore, thermoplastic moldings were prepared from cedar and beech flours, and their physical properties were examined. Similar to the case of beech, a resin-like molding with a density of approximately 1.45?g/cm3 was obtained from the cedar flour steamed at 180°C or higher, and it was revealed by SEM observation that in these moldings wood flour particles adhere to each other. - The presence of threatened or endangered species often strongly influences management and conservation decisions. Within the Murray–Darling Basin (MDB), Australia, the presence of threatened native fish affects the management and allocation of water resources. In New South Wales, these decisions are currently based on traditional fisheries data and a predictive MaxEnt model. However, it is important to verify the model's predictive power given the implication it may have, but this requires methods with a high detection sensitivity for rare species.
- Although the use of environmental DNA (eDNA) monitoring, in particular eDNA metabarcoding, achieves a higher detection sensitivity compared with traditional methods, earlier surveys in the MDB have shown that the highly abundant and invasive common carp (Cyprinus carpio) can reduce detection probabilities for rare species. Consequently, a polymerase chain reaction (PCR) blocking primer designed to block the amplification of carp eDNA could increase the detection probabilities for rare native species while simultaneously reducing the required sampling effort and survey costs. Although PCR blocking primers are often used in ancient DNA and dietary studies, no aquatic eDNA metabarcoding study to date has evaluated the potential benefits of using PCR blocking primers.
- A laboratory and field-based pilot study was used to address this knowledge gap and assess the impact of a blocking primer, targeting cyprinid fishes (including carp), on the detection probabilities of native species and the minimum sampling effort required.
- Results showed that the inclusion of the blocking primer increased the detection probabilities for native species by 10–20% and reduced the minimum required sampling effort by 25–50%. These findings provide important insights into possible methods for optimizing eDNA metabarcoding surveys for the detection of rare aquatic species.

Objective To evaluate the anaesthetic death risk for dogs and cats in a French private practice. Study design Observational cohort study. Animal population All small animals anesthetized at the Centre Hospitalier Vétérinaire des Cordeliers between April 15th, 2008 and April 15th, 2010. Methods General anaesthesia was defined as a drug‐induced unconsciousness characterised by a controlled and reversible depression of the central nervous system and analgesia, sufficient to allow endotracheal intubation. Patient outcome (alive or dead) was assessed at the end of anaesthesia defined as the meeting point of the return of consciousness, rectal temperature >36 °C and ability to maintain sternal recumbency. Death occurring during anaesthesia was recorded. Relationship between anaesthetic death and ASA status, species, age, nature of the procedure, anaesthetic protocol and occurrence of epidural administration of a combination of morphine and bupivacaine were analysed. Results During the study period 3546 animals underwent general anaesthesia. The overall death rate in the present study was 1.35% (48 in 3546, 95% CI 0.96–1.75). The death rate of healthy animals (ASA 1 and 2) was 0.12% (3 in 2602 95% CI 0.02–0.34). For sick animals (ASA status 3 and over), the overall death rate was 4.77% (45 in 944 95% CI 3.36–6.18). The death rates in the ASA 3, 4 and 5 categories were 2.90%, 7.58% and 17.33%, respectively. The main factor associated with increased odds of anaesthetic death in ASA categories 3 and over was poor health status (ASA physical status classification). The nature of the procedure the patient underwent and epidural administration of a combination of morphine and bupivacaine were not correlated with the occurrence of death during anaesthesia. Neither species nor age effects were detected. Conclusion and clinical relevance Specific factors were associated with increased odds of anaesthetic death, especially poor health status. Efforts must be directed towards thorough preoperative patient evaluation and improvement of clinical conditions if possible. Identification of risk factors before anaesthesia should lead to increased surveillance by trained staff. This could result in better outcomes. 相似文献

Tritrichomonas suis (=T. foetus) has recently been reported to be a causative agent of chronic large-bowel diarrhea in cats. While the disease was previously attributed to Pentatrichomonas hominis, the etiologic agent for feline trichomonal diarrhea was identified as T. suis. Although feline trichomonosis due to T. suis has been reported at prevalences ranging from 14 to 31% in Europe and the U.S., no reports of the pathogen have been published to date in Japan. In 2008, however, we encountered a case of feline trichomonosis at the Veterinary Teaching Hospital of Hokkaido University. The parasite was identified as T. suis by nested PCR amplification of partial internal transcribed spacer region 1 and 5.8S ribosomal RNA gene sequences with T. suis-specific primers and DNA sequencing of the amplified products. We then conducted surveys for feline trichomonosis in three different animal hospitals using either cultivation and/or PCR-based assays. The results revealed that 13 of 147 samples (8.8%) were positive for T. suis, and that 5 of the 13 infected cats, which ranged between 1 month and 7.5 years-old, showed chronic diarrhea. Seven of the infected cats were purebred and 6 were mixed breed. These findings suggested that feline trichomonosis is prevalent in Japan, and that T. suis may play a role as a causative agent of feline chronic diarrhea. 相似文献

A common assumption is that ecosystem services respond linearly to changes in habitat size. This assumption leads frequently to an "all or none" choice of either preserving coastal habitats or converting them to human use. However, our survey of wave attenuation data from field studies of mangroves, salt marshes, seagrass beds, nearshore coral reefs, and sand dunes reveals that these relationships are rarely linear. By incorporating nonlinear wave attenuation in estimating coastal protection values of mangroves in Thailand, we show that the optimal land use option may instead be the integration of development and conservation consistent with ecosystem-based management goals. This result suggests that reconciling competing demands on coastal habitats should not always result in stark preservation-versus-conversion choices. 相似文献

We examined the cerebellum and cerebrum of 4 vaccinated dogs, 3–60-mo-old, that displayed clinical signs of canine distemper virus (CDV) infection, and died 7–40 d after developing neurologic signs. The main histologic lesions were demyelination, gliosis, meningitis, perivascular lymphocytic cuffing, and inclusion bodies. These lesions were similar in all 4 cases regardless of the time since vaccination, except that meningoencephalitis and gliosis were subacute in 3 dogs and chronic in 1 dog. However, these differences did not appear to be related to their vaccination status. Immunohistologically, a CDV-positive immunoreaction was seen mainly in astrocytes, neurons and their axons, lymphocytes around and in the blood vessels of the pia mater and choroid plexus, ependymal cells of each ventricle, and the cells of the choroid plexus. The histologic and immunohistologic changes were similar in the cerebellum and cerebrum. The genetic characterization of the virus strains in 2 of these naturally occurring canine distemper cases confirmed that they were South American wild-type strains (Kiki and Uy251) belonging to the EU1/SA1 lineage. Successful prairie restoration will depend in part on convincing private landowners with agricultural and recreational use goals to implement appropriate rangeland management practices, such as prescribed burning and cattle grazing, to control invasive species and encroachment of woody plants. However, landowners have been slow to adopt appropriate practices in the US Midwest. The purpose of this study was to explore attitudes and behaviors of private landowners toward prescribed burning and moderate stocking as rangeland management tools. A survey was mailed to 193 landowners (response rate 51%) in the Grand River Grasslands region of southern Iowa and northern Missouri. While 68% of landowners viewed grazing as a legitimate land management tool, only half of landowners thought of fire as a legitimate tool. Over 75% of respondents believed that the increase in eastern redcedar and other trees in grasslands was a problem, with 44% considering it a major problem. Although 84% of landowners said that they had taken action to control eastern redcedar, only 25% had participated in a prescribed burn. Income from agriculture and recreational goals were negatively and significantly correlated (-0.252, P = 0.035). While holding recreational goals constant in the analysis, landowners reporting income from agriculture goals as very or extremely important were negatively and significantly associated with reporting environment and grassland factors as very or extremely important. Adoption of prescribed burning by private landowners might be more widespread if proponents focus on the effectiveness of fire for controlling eastern redcedar, which is viewed as a problem by most landowners in the region. Intervention efforts must include landowners with different goals as part of the promotion and educational process. 相似文献
