Rubber mats covering concrete slatted flooring are a developing market in dairy barns but remain rare in beef cattle facilities. The aim of this study was to investigate the effects of covering slatted concrete floor with perforated rubber mats on behaviour and occurrence of skin and claw lesions in fattening bulls. The groups of six bulls each with a total average age of 9.8 months were kept over 1 year on either slatted concrete (CONCRETE PEN) or on slatted concrete covered completely (RUBBER PEN) or partially (CHOICE PEN) with perforated rubber mats. Every quarter-year, behaviour (preference of flooring, lying, aggression, mounting) was recorded. In two-weekly intervals the incidence of skin lesions was examined. At 12 and 18 months of age the rising time of the bulls was measured. At the beginning of the study and after slaughter claw dimensions were recorded. Bulls in the CHOICE PEN preferred (P<0.01) the rubber coated area throughout the experiment. Animals in the RUBBER and the CHOICE PENS showed more lying periods (P<0.01) and had a lesser incidence of skin lesions (P<0.01) compared to bulls in the CONCRETE PEN. Bulls in the CHOICE PEN needed less time for rising (2.7+/-0.3s) than bulls in the CONCRETE PEN (4.4+/-0.5s, P<0.01). Net claw growth differed significantly between all pens (RUBBER>CHOICE>CONCRETE; P<0.05). In conclusion, the results of this study indicate that rubber coated slatted flooring has a positive influence on the housing conditions of beef cattle. 相似文献
The spliceosome, a ribonucleoprotein complex that includes proteins and small nuclear RNAs (snRNAs), catalyzes RNA splicing through intron excision and exon ligation to produce mature messenger RNAs, which, in turn serve as templates for protein translation. We identified four point mutations in the U4atac snRNA component of the minor spliceosome in patients with brain and bone malformations and unexplained postnatal death [microcephalic osteodysplastic primordial dwarfism type 1 (MOPD 1) or Taybi-Linder syndrome (TALS); Mendelian Inheritance in Man ID no. 210710]. Expression of a subgroup of genes, possibly linked to the disease phenotype, and minor intron splicing were affected in cell lines derived from TALS patients. To gain a better understanding of the mechanisms underlying spatial heterogeneity in ichthyoplankton communities in the SW Atlantic, we examined for the first time the latitudinal and continental shelf–slope gradients in ichthyoplankton structure and oceanographic features in the Campos Basin during the relaxation phase of coastal upwellings (late autumn to early winter). This region, located on SE Brazil's continental margin, is the most productive offshore oil basin in the country and has ecological relevance owing to the existence of areas that experiences mesoscale (tens to hundreds of kilometers) processes (eddies, filaments, and upwelling) caused by the interaction of continental shelf and slope circulation with deep water masses. The present study collected a total of 3892 fish eggs and 10,030 larvae from 36 sampling stations, averaging 22 eggs per 100 m3 and 56 larvae per 100 m3. A total of 250 taxa (5 for fish eggs and 248 for larvae) were identified, encompassing 80 families and 145 genera. Species distribution exhibited a considerable degree of spatial variability, which was related mostly to hydrological characteristics. In general, greater densities were associated with higher nutrient concentrations areas. Through distance-based Redundancy Analysis, some discriminating species were found to associate with certain areas of the continental shelf characterized by colder temperatures. The ichthyoplankton distribution patterns suggested a potential influence from mesoscale oceanographic fronts, specifically those that induce upwelling of the cold and nutrient-rich South Atlantic Central Water. Over the past decades, the general trend towards shade reduction and intensification of cacao management has led to biodiversity losses. In the Peruvian Amazon, the regional government is heavily promoting crop conversion to shift from regionally marketed foods towards cacao (Theobroma cacao) and copoazu (Theobroma grandiflorum). While this shift is already visibly impacting the farming landscape and the lives of many smallholder farmers, little is known about the reasons that drive farmers to choose certain types of Theobroma species or cacao varieties over others. In this paper, we addressed how cacao farmers perceive and manage specific and varietal Theobroma diversity. We interviewed cacao farmers (n?=?20) during a seed-exchange fair and adapted a version of the four-square analysis to explore which Theobromas are currently adopted by farmers and why. The native cacao variety (cacao chuncho) was the one cultivated by most farmers, followed by the more industrial clonal varieties. The source of seeds and seedlings for the most cultivated varieties was a mix of donations by public institutions (for clonal varieties) and informal exchange among farmers (mainly for native and criollo varieties, and species i.e. macambo (Theobroma bicolor) and cacauillo (Theobroma speciosum)). The cacao varieties incentivized by public institutions were the least desired for future investment. The motivations for farmers to plant Theobromas was mainly based on the perceived current and potential market, but their desire to invest in a given species or variety was also based on pest resistance, traditional use, farm diversification and taste. Copoazu and CCN cacao varieties are produced by many households in large areas, while macambo, cacauillo and the cacao varieties TSH and porcelana are produced by few households in small areas. Our study suggests that agroforestry systems that include alternative Theobroma species are multi-strata and more diverse than cacao-based systems. It highlights the risks of agrobiodiversity loss associated with the promotion of industrial cacao varieties, and the importance of seed and seedling access for the development of diverse farming systems. We recommend the expansion of local seed-sharing networks and the extension of public cacao-donation campaigns to neglected and underutilized Theobromas with socio-economic and environmental benefits, in order to make the local farming systems more diverse and resilient.
After cryopreservation for 13.3-15.8 years, the viability of the infective larvae (L3) of Trichostrongylus axei, T. colubriformis, Oesophagostomum columbianum, Haemonchus contortus, Ostertagia circumcincta, T. falculatus, Nematodirus spathiger, Chabertia ovina and Dictyocaulus filaria was assessed in sheep, by being deposited at their predilection sites. D. filaria was, however, an exception, in that the L3 were injected into the jugular vein. The mean development of all the species was 22.8%, but if three species (O. columbianum, C. ovina and D. filaria), that developed poorly are disregarded, then the mean development was 33.4%, similar to previous tests after shorter periods of cryopreservation. The L3 of some of the species appeared sluggish when examined 10-15 min after being thawed, and in the case of H. contortus practically all the larvae of the original batch tested in the previous trials of the series appeared dead when thawed for use in the present trial, and were replaced by another batch of L3 of the same species. When re-examined after about 8 h, however, a high percentage of the L3 of the original batch appeared to have become revitalised, and their viability was tested in a trial reported elsewhere. The intestinal cells of the majority of the L3 of N. spathiger, O. circumcincta and C. ovina were vesiculated when they were thawed. Nevertheless, the degree of development of the former two species was of the highest in the trial, and it can be concluded that this phenomenon does not necessarily impede the viability of larvae. 相似文献
A 12-year-old female, neutered German shepherd dog developed progressive hindlimb followed by forelimb ataxia leading to tetraplegia. Neurological examination suggested lower motor dysfunction. Biochemical evaluation revealed a monoclonal hypergammaglobulinaemia, hypoalbuminaemia and hypercalcaemia. Multiple lytic lesions were identified radiographically in numerous bones. A bone marrow aspirate confirmed the diagnosis of multiple myeloma, with large numbers of plasma cells seen in clusters. An electromyogram revealed positive sharp waves and fibrillation potentials in the skeletal muscles of the limbs, suggesting a polyneuropathy. The dog was treated with chemotherapy using melphalan and prednisolone. Both the hypergammaglobulinaemia and the polyneuropathy resolved and the dog had normal motor function four weeks after commencing treatment. Polyneuropathy may occur as a paraneoplastic syndrome secondary to myeloma, and in this case was reversible following treatment of the underlying disease. 相似文献
The Kunitz-type protease inhibitors are the best-characterized family of serine protease inhibitors, probably due to their abundance in several organisms. These inhibitors consist of a chain of ~60 amino acid residues stabilized by three disulfide bridges, and was first observed in the bovine pancreatic trypsin inhibitor (BPTI)-like protease inhibitors, which strongly inhibit trypsin and chymotrypsin. In this review we present the protease inhibitors (PIs) described to date from marine venomous animals, such as from sea anemone extracts and Conus venom, as well as their counterparts in terrestrial venomous animals, such as snakes, scorpions, spiders, Anurans, and Hymenopterans. More emphasis was given to the Kunitz-type inhibitors, once they are found in all these organisms. Their biological sources, specificity against different proteases, and other molecular blanks (being also K+ channel blockers) are presented, followed by their molecular diversity. Whereas sea anemone, snakes and other venomous animals present mainly Kunitz-type inhibitors, PIs from Anurans present the major variety in structure length and number of Cys residues, with at least six distinguishable classes. A representative alignment of PIs from these venomous animals shows that, despite eventual differences in Cys assignment, the key-residues for the protease inhibitory activity in all of them occupy similar positions in primary sequence. The key-residues for the K+ channel blocking activity was also compared. 相似文献

At least three haemotropic mycoplasmas have been recognized in cats: Mycoplasma haemofelis (Mhf), 'Candidatus Mycoplasma haemominutum' (CMhm) and 'Candidatus M. turicensis' (CMt). The latter was originally identified in a Swiss pet cat with haemolytic anaemia and shown to be prevalent in domestic cats and wild felids worldwide using molecular methods. So far, there has been no confirmatory morphological evidence of the existence of CMt presumably due to low blood loads during infection while CMhm has only been characterized by light microscopy with discrepant results. This study aimed to provide for the first time electron microscopic characteristics of CMt and CMhm and to compare them to Mhf. Blood samples from cats experimentally infected with CMt, CMhm and Mhf were used to determine copy numbers in blood by real-time PCR and for transmission and scanning electron microscopy. High resolution scanning electron microscopy revealed CMt and CMhm to be discoid-shaped organisms of 0.3 μm in diameter attached to red blood cells (RBCs). In transmission electron microscopy of CMt, an oval organism of about 0.25 μm with several intracellular electron dense structures was identified close to the surface of a RBC. CMhm and CMt exhibited similar morphology to Mhf but had a smaller diameter. This is the first study to provide morphological evidence of CMt thereby confirming its status as a distinct haemoplasma species, and to present electron microscopic features of CMhm. 相似文献
