The role of dermonecrotic toxin (DNT) of Bordetella bronchiseptica and Pasteurella multocida, purified by repeated chromatography in Sephacryl S-200 gel, in the pathogenesis of atrophic rhinitis (AR) of swine was studied bacteriologically, clinically and pathologically. Two-week-old specific pathogen-free (SPF) piglets were parenterally treated with 30 micrograms of DNT 3 times at 2-day interval and 7-week-old piglets were treated with 15 micrograms of DNT twice a week for 5 weeks. In 2- to 3-week-old piglets, both B. bronchiseptica DNT and P. multocida DNT produced nasal turbinate lesions with similar severity, characterized by damage of the cilia, epithelial metaplasia, intensive proliferation of osteoblasts, regressive changes, and diffuse osteocytic osteolysis. In 7- to 12-week-old piglets, treatment with B. bronchiseptica DNT failed to produce progressive changes in the nasal turbinates. Histopathological examination revealed osteogenic processes and osteoid synthesis besides the proliferation of osteoblasts and mild osteocytic osteolysis. Moreover, severe gross pathological lesions developed in the stomach, liver, kidneys, and lymphoid organs. The piglets' appetite and body weight gain gradually decreased during the DNT treatment and in the last week when the toxic signs appeared. Treatment of 7- to 12-week-old piglets with P. multocida DNT resulted in progressive AR. Histopathologically, diffuse osteocytic osteolysis was observed in the nasal turbinates. Neither clinical signs nor pathological lesions of the visceral organs developed in these piglets. The authors emphasize that the DNT of B. bronchiseptica basically differs from that of P. multocida in biological properties, though there are certain similarities between the DNTs. 相似文献

Laboratory incubations of sieved (<2mm) forest humus were used to study the response of C and N mineralization to perturbation. Considerable variation in the ratio of mineralized C to mineralized N was observed. This ratio widened with increasing temperature. At constant temperature, addition of P stimulated CO2-C evolution and reduced NH4+-N production, also widening the C:N ratio of substrate mineralized. Addition of weak base stimulated mineralization of N more than C, reducing the C:N ratio of substrate mineralized. Addition of weak acid, mineral-N, or excessive amounts of water inhibited CO2-C evolution while stimulating production of NH4+-N, resulting in a “negative correlation” between the two, and reducing the C:N ratio of substrate mineralized still further.Results were interpreted in terms of effects on microbial biomass. A relatively benign treatment (P addition) may promote microbial growth and respiration, reducing net N availability. A moderate perturbation (addition of weak base) favors new organisms growing partly at the expense of microbial necromass. These organisms will mineralize some necromass-N, increase net N mineralization, and reduce the C:N ratio of substrate mineralized. Under severe conditions (addition of acid) the C:N ratio of substrate mineralized approaches that of the microbial biomass itself, suggesting that the biomass is the primary substrate mineralized. Microbial mortality is likely to be a significant factor affecting the supply of N in field situations, and should be included in any general model of soil N mineralization processes. 相似文献

Specific contrast ultrasound is widely applied in diagnostic procedures on humans but remains underused in veterinary medicine. The objective of this study was to evaluate the use of microbubble-based contrast for rapid ultrasonographic diagnosis of thrombosis in small animals, using male New Zealand white rabbits (average weight about 3.5 kg) as a model. It was hypothesized that the use of microbubble-based contrast agents will result in a faster and more precise diagnosis in our model of thrombosis. A pro-coagulant environment had been previously established by combining endothelial denudation and external vessel wall damage. Visualization of thrombi was achieved by application of contrast microbubbles [sterically stabilized, phospholipid-based microbubbles filled with sulfur hexafluoride (SF6) gas] and ultrasonography. As a result, rapid and clear diagnosis of thrombi in aorta abdominalis was achieved within 10 to 30 s (mean: 17.3 s) by applying microbubbles as an ultrasound contrast medium. In the control group, diagnosis was not possible or took 90 to 180 s. Therefore, sterically stabilized microbubbles were found to be a suitable contrast agent for the rapid diagnosis of thrombi in an experimental model in rabbits. Arid areas are highly sensitive to climate change and are ideal model systems to study the potential impact of climate change on species' community structure. Biological soil crust (BSC) formation plays an ecological role in a number of key processes in the development of dry ecosystems. It was hypothesized that BSC succession and function are affected by aridity level and limited by rainfall. Furthermore, it is possible to infer the direction of the BSC succession based on aridity level, and the latter can imitate future climate change scenarios. The objectives of this study were to investigate the microbial biomass and diversity of the BSC structure in three sites differing in aridity level (semiarid, arid and hyper-arid), by combining physical and biophysiological measurements with 16S rRNA gene fragment and phospholipid fatty acid (PLFA) analyses. Physical and biophysiological parameters of the BSC were significantly influenced by aridity level. Total protein and polysaccharide contents were strongly correlated with total PLFA-based microbial biomass. Gram-positive biomarkers and microbial biomass were significantly higher in the wettest (semiarid) site than in the driest (hyper-arid) one. Multivariate-analysis based ordination of the PLFA data segregated the cluster of semiarid data from that of the hyper-arid site, while data from the arid site were dispersed between the two. The phylogenetic distribution of prominent 16S rRNA bacterial gene sequences along the aridity levels was in agreement with the PLFA analysis: the hyper-arid site was dominated by the cyanobacterium Microcoleus vaginatus, while diverse populations of cyanobacteria and soil bacteria were found in the other sites. These complementary tools allowed a simple and sensitive measurement of the influence of aridity levels on BSC successional stage. The effectiveness of Nitrapyrin (NI) as a nitrification inhibitor is largely determined by the rate at which it is hydrolyzed. A procedure was developed in which Cl? liberated during NI hydrolysis was used to follow NI degradation in laboratory studies. The procedure allowed determination of Cl? from as little as 0.5 mg·l?1 Nitrapyrin and permitted analysis of up to 20 samples h?1. The method must be restricted to experiments where background Cl?1 can be either removed by leaching or corrected for by use of control samples.Temperature was the most important factor affecting NI hydrolysis and the process was not biologically-mediated. The rate of hydrolysis was not affected by pH in the range 2.7–11.9, or by sorption on colloidal surfaces providing the surfaces remained in contact with the solution. Hydrolysis was slower in a soil at field moisture capacity than at saturation. The findings also showed that experiments conducted in containers with lipophilic components (e.g. rubber or polyethylene) may be suspect due to sorption of NI. 相似文献
