In this study, lipid classes and fatty acid composition were determinedin eggs of P. vannamei as a function of survival to zoeaIII stage. Spawns were reared individually to zoea III and grouped, accordingtotheir final survival to this larval stage, into spawns of high and lowsurvival.Eggs of individual spawns were analyzed for lipid and fatty acid composition ofneutral and polar lipids and the results were then grouped according tosurvivalto zoea III. The lipids within each group (high and low survival to zoea III)were pooled for the separation of phospholipids and the analysis of their fattyacid composition.Higher levels of triglycerides, carotenoids, and linoleic (18:2n-6) acid ineggs were associated with improved survival to zoea III. Linoleic acid washigher in spawns from the high survival group in both neutral and polarfractions and in most of the phospholipid classes analyzed. Docosahexanoic acid(DHA) was not related to survival to zoea, probably because its content washighenough (> 15% of total fatty acids) to satisfy embryo and early larvaldevelopment needs. A high content of eicosapentaenoic acid (EPA) was found inphosphatidylethanolamine, lysophosphatidylethanolamine, phosphatidylserine, andphosphatidylinositol compared to other phospholipid classes, suggesting aspecific role of EPA in these lipids. These results describe the specificphospholipid composition of penaeid eggs and could (potentially) be used aspredictive indicators of larval quality for research and production purposes.  相似文献

Neurones located 200 to 300 microns from the surface of the auricular lobe of the frog cerebellar cortex, and identified as Purkinje cells, were activated antidromically from the eighth cranial nerve. A parallel anatomical study confirmed the existence of this projection. On the basis of these findings the existence of a cerebello-vestibular efferent system is postulated, the precise significance of which is as yet unclear. However, since Purkinje cells in other species have an inhibitory action on their target cells, the Purkinje efferent system to the vestibular organ may have an action similar to that ascribed to the olivo-cochlear bundle upon the cochlea, that is, to serve as an inhibitory control system.  相似文献

The aim of this paper was to study how maturity and maceration length affect color, phenolic compounds, polysaccharides, and sensorial quality of Cabernet Sauvignon and Tempranillo wines at three stages of grape ripening. Ripeness increased color extractability, phenolic compounds, and polysaccharide concentrations. Moreover, the proanthocyanidin mean degree of polymerization (mDP) and the percentage of prodelphinidins also increased with maturity, whereas the percentage of galloylation decreased. In general, wines from riper grapes contain higher proportions of skin proanthocyanidins. Color and anthocyanin concentration decreased when the maceration was longer, whereas polysaccharide and proanthocyanidin concentrations did the opposite. It was also detected that the mDP and the percentage of prodelphinidins decreased when the maceration was extended, whereas the percentage of galloylation increased. These data seem to indicate that proanthocyanidin extraction from seeds is clearly increased throughout the maceration time.  相似文献

The usefulness of the solvent mixture dichloromethane/methanol for lipid extraction and the determination of lipid classes and fatty acids in samples of different natures was conducted. Two different extraction methods were compared, one containing chloroform/methanol, another containing dichloromethane/methanol. Total lipid extraction showed some minor differences but no variation in the lipid classes. Regarding the fatty acid profile, in Echium virescens seeds, 17 major fatty acids could be identified and quantified, and all were equally extracted when either solvent system was employed. In Echium acanthocarpum hairy roots, 17 major fatty acids were quantified, showing some statistical differences for one cell line in favor of chloroform. The data obtained from the liquid nutrient medium were also comparable. The cod roe sample showed 31 major fatty acids, showing no statistical differences between the two solvent systems. Contrarily, the CH 2Cl 2 method was able to extract 31 main fatty acids found in European seabass dorsal muscle more efficiently than the CHCl 3 method. The results indicate that, for lipid extraction and fatty acid assessment, dichloromethane/methanol can readily replace the commonly employed chloroform/methanol, thus avoiding the major health, security, and regulatory problems associated with the use of chloroform.  相似文献

An experimental design has been used to optimize the extraction of volatile compounds from summer truffle aroma (Tuber aestivum) by using headspace solid phase microextraction. The extracted compounds have been analyzed by gas chromatography with a flame ionization detector and by gas chromatography-mass spectrometry (GC-MS). In an attempt to develop an objective method to fully characterize truffle aroma, a fiber of medium polarity (for flavors) was used to avoid discrimination toward very nonpolar and polar volatile compounds. To optimize the extraction conditions, a response surface experimental design was applied considering three factors such as extraction temperature, equilibrium time, and extraction time. From the statistical analysis of the experimental design, it was possible to determine that the most important factor influencing the abundance of aroma compounds was the extraction temperature. Optimal extraction temperature was established at approximately 50 degrees C. By using GC-MS, it was possible to identify 37 compounds, most of them previously described as responsible for truffle aroma.  相似文献

Background

In practical farming, there is often a need for short-term availability of information on the soil nutrient status.

Aims

To develop a new express method for the extraction of major plant-available nutrients and measurement of soil nutrients. In future, this method shall serve for in-field measurements of soil samples with an ion-sensitive field-effect transistor (ISFET).

Methods

Various extraction conditions such as type of extractant, soil-to-solution ratio, time, and intensity were investigated on a broad selection of dried soil samples in the laboratory. Based on 83 field-moist soil samples with varying clay contents, these conditions were compared to standard laboratory methods.

Results

With increasing extraction time, the nutrient concentrations increased. When the soil-to-solution ratio was reduced, a greater share of nutrients was extracted, independent of soil type. H2O and 0.01 M CaCl2 and standard calcium-acetate-lactate (CAL) solution proved to be too weak in the short period to reach the ISFET sensor measurement range. Higher concentrated CAL solutions performed much better. Finally, a 5-min CaCl2 extraction followed by the removal of an aliquot for the determination of soil pH and NO3 was found to be effective. The remaining solution was then mixed with 0.20 M CAL solution for the analysis of H2PO4 and K+ at 10 min of extra extraction time. This extraction method showed very good correlations with the values based on the German laboratory reference methods for pH (R2 = 0.91) and for nitrate (R2 = 0.95). For phosphorus and potassium, we obtained an R2 of 0.70 and 0.81, respectively, for all soils. When soils were grouped according to clay content higher correlations were found.

Conclusions

A new express method based on a wet-chemical approach with a soil preparation procedure was successfully developed and validated. This seems to be a valuable basis for future in-field measurements via ISFET.  相似文献

The classification and phylogeny of the species belonging to Solanum section Lycopersicon is a complex issue that has not yet reached a widely accepted consensus. These species diverged recently, are still closely related and, in some cases, are still even capable of interspecific hybridization, thereby blurring the difference between intra- and interspecific variation. To help resolve these issues, in the present study, several accessions covering the natural range for each species were used. In addition, to avoid biases due to the molecular method employed, both AFLP markers and two nuclear-gene sequences, CT179 and CT66, were used to characterize the plant materials. The data obtained suggest a classification similar to those previously proposed by other authors, although with some significant changes. Twelve species were recognized as distinct based on this dataset. According to the data presented, the recently proposed species, S. corneliomulleri, is indistinguishable from S. peruvianum s.str. In addition, both the sequence and the AFLP trees suggest that S. arcanum could represent a complex of populations composed of two cryptic species. With regard to phylogenetic relationships among these species, some clear groups were found: the Lycopersicon group formed by S. pimpinellifolium, S. lycopersicum, S. cheesmaniae and S. galapagense; the Arcanum group constituted by S. chmielewskii, S. neorickii, S. arcanum and S. huaylasense; and the Eriopersicon group made up of S. peruvianum and S. chilense. Solanum pennellii and S. habrochaites are not included in any group, but are the closest to the S. lycopersicoides outgroup. We present here an indicator of soil quality that evaluates soil ecosystem services through a set of 5 subindicators, and further combines them into a single general Indicator of Soil Quality (GISQ). We used information derived from 54 properties commonly used to describe the multifaceted aspects of soil quality. The design and calculation of the indicators were based on sequences of multivariate analyses. Subindicators evaluated the physical quality, chemical fertility, organic matter stocks, aggregation and morphology of the upper 5 cm of soil and the biodiversity of soil macrofauna. A GISQ combined the different subindicators providing a global assessment of soil quality.Research was conducted in two hillside regions of Colombia and Nicaragua, with similar types of land use and socio-economic context. However, soil and climatic conditions differed significantly. In Nicaragua, soil quality was assessed at 61 points regularly distributed 200 m apart on a regular grid across the landscape. In Colombia, 8 plots representing different types of land use were arbitrarily chosen in the landscape and intensively sampled. Indicators that were designed in the Nicaragua site were further applied to the Colombian site to test for their applicability.In Nicaragua, coffee plantations, fallows, pastures and forest had the highest values of GISQ (1.00; 0.80; 0.78 and 0.77, respectively) while maize crops and eroded soils (0.19 and 0.10) had the lowest values. Examination of subindicator values allowed the separate evaluation of different aspects of soil quality: subindicators of organic matter, aggregation and morphology and biodiversity of macrofauna had the maximum values in coffee plantations (0.89; 0.72 and 0.56, respectively on average) while eroded soils had the lowest values for these indicators (0.10; 0.31 and 0.33, respectively).Indicator formulae derived from information gained at the Nicaraguan sites were not applicable to the Colombian situation and site-specific constants were calculated.This indicator allows the evaluation of soil quality and facilitates the identification of problem areas through the individual values of each subindicator. It allows monitoring of change through time and can guide the implementation of soil restoration technologies. Although GISQ formulae computed on a set of data were only valid at a regional scale, the methodology used to create these indices can be applied everywhere.  相似文献
