Glyphosate is the most used herbicide in Argentina, accounting for 62% of the commercialized pesticides on the market. It is used as a weed controller in no-till systems, and it is also applied to various genetically modified crops (e.g., soybean, corn, and cotton). Although it has a high solubility in water, it tends to adsorb and accumulate in agricultural soils. The main objectives of this work were to compare the dissipation of glyphosate and the accumulation of its metabolite aminomethylphosphonic acid (AMPA) over time in three soils from agricultural areas of Argentina under long-term management with no-till (NT) and conventional tillage (CT) practices. There were no differences in dissipation between NT and CT, indicating that the glyphosate-degrading microflora was not modified by the different tillage managements. Moreover, tillage practices did not alter the general soil properties; therefore, glyphosate bioavailability was not affected by NT or CT practice. Forty percent of the applied glyphosate was degraded within the first three days in all soils, indicating a fast initial dissipation rate. However, the dissipation rate considerably decreased over time, and the degradation kinetics followed a bi-exponential (or two-compartment) kinetic model. No differences were found between tillage practices. Dissipation was not related to the microbial activity measured as soil respiration. The fast decrease in the concentration of glyphosate at the beginning of the dissipation study was not reflected in an increase in the concentration of AMPA. The estimated half-lives for glyphosate ranged between 9 and 38 d. However, glyphosate bioavailability decreases over time, as it is strongly adsorbed to the soil matrix. BACKGROUND: Fifteen novel derivatives of D‐DIBOA, including aromatic ring modifications and the addition of side chains in positions C‐2 and N‐4, had previously been synthesised and their phytotoxicity on standard target species (STS) evaluated. This strategy combined steric, electronic, solubility and lipophilicity requirements to achieve the maximum phytotoxic activity. An evaluation of the bioactivity of these compounds on the systems Oryza sativa–Echinochloa crus‐galli and Triticum aestivum–Avena fatua is reported here. RESULTS: All compounds showed inhibition profiles on the two species Echinochloa crus‐galli (L.) Beauv. and Avena fatua L. The most marked effects were caused by 6F‐4Pr‐D‐DIBOA, 6F‐4Val‐D‐DIBOA, 6Cl‐4Pr‐D‐DIBOA and 6Cl‐4Val‐D‐DIBOA. The IC50 values for the systems Echinochloa crus‐galli–Oryza sativa and Avena fatua–Triticum aestivum for all compounds were compared. The compound that showed the greatest selectivity for the system Echinochloa crus‐galli–Oryza sativa was 8Cl‐4Pr‐D‐DIBOA, which was 15 times more selective than the commercial herbicide propanil (Cotanil‐35). With regard to the system Avena fatua–Triticum aestivum, the compounds that showed the highest selectivities were 8Cl‐4Val‐D‐DIBOA and 6F‐4Pr‐D‐DIBOA. The results obtained for 6F‐4Pr‐D‐DIBOA are of great interest because of the high phytotoxicity to Avena fatua (IC50 = 6 µM , r2 = 0.9616). CONCLUSION: The in vitro phytotoxicity profiles and selectivities shown by the compounds described here make them candidates for higher‐level studies. 8Cl‐4Pr‐D‐DIBOA for the system Echinochloa crus‐galli‐Oryza sativa and 6F‐4Pr‐D‐DIBOA for Avena fatua‐Triticum aestivum were the most interesting compounds. This paper discusses the possibility of including the culturing of microalgae within a conventional wastewater treatment sequence by growing them on the blackwater (BW) from biosolid dewatering to produce biomass to feed the anaerobic digester. Two photobioreactors were used: a 12 L plexiglas column for indoor, lab-scale tests and a 85 L plexiglas column for outdoor culturing. Microalgae (Chlorella sp. and Scenedesmus sp.) could easily grow on the tested blackwater. The average specific growth rate in indoor and outdoor batch tests was satisfactory, ranging between 0.14 and 0.16 day?1. During a continuous test performed under outdoor conditions from May to November, in which the off-gas from the combined heat and power unit was used as the CO2 source, an average biomass production of 50 mgTSS L?1 day?1 was obtained. However, statistical analyses confirmed that microalgal growth was affected by environmental conditions (temperature and season) and that it was negatively correlated with the occurrence of nitrification. Background: Analysis of body fluids includes an estimate of total nucleated cell count (TNCC). Automated methods may enhance the accuracy and timeliness of TNCC results. Objective: The purpose of this report was to assess the ability of the ADVIA 120 hematology analyzer to accurately count nucleated cells in pleural and peritoneal fluids from animals, compared with manual counts. Methods: Pleural and peritoneal fluids submitted in EDTA tubes to our laboratory over a 17‐month period were used in the study. TNCC/μL was determined by a manual method, using a hemocytometer, and by an automated method, using the ADVIA 120. Correlation of results was determined by Passing‐Bablok regression, Bland–Altman plots, and Pearson correlation analysis. Results: Samples from dogs (n=36), cats (n=36), horses (n=59), and alpacas (n=11) were analyzed. High correlation in TNCC between methods was found for peritoneal fluid (n=93, r=.959), pleural fluid (n=49, r=.966), and all fluids combined (n=142, r=.960) (P<.001). Variation between methods was greater in samples with TNCCs<1000/μL (r=.62, P<.001). The ADVIA systematically overestimated the number of cells in all fluid samples by 95 cells/μL (confidence interval=19.2–190.5/μL). Conclusion: The ADVIA 120 reliably determines TNCC in pleural and peritoneal effusions and can be recommended for routine veterinary laboratory analysis. 相似文献

Small ruminant lentiviruses infect sheep and goats worldwide, causing chronic progressive diseases and relevant economic losses. Disease eradication and prevention is mostly based on serological testing. The goal of this research was to investigate the presence of the small ruminant lentiviruses (SRLVs) in Jordan and to characterize the serological response in sheep and goat populations. A panel of sera were collected from flocks located in Northern Jordan and Jordan Valley. The samples were tested using three ELISA assays: a commercially available ELISA based on p25 recombinant protein and transmembrane peptide derived from British maedi–visna virus (MVV) EV1 strain, an ELISA based on P16-P25 recombinant protein derived from two Italian strains representative of MVV- and caprine arthritis encephalitis virus (CAEV)-like SRLVs, and an ELISA based on SU5 peptide from the same two Italian isolates. The results indicate that both MVV- and CAEV-like strains are present in Jordan and that the majority of the viruses circulating among sheep and goat populations belong to the MVV-like genotype. 相似文献

Background, aim, and scope In urban areas, storm water runoff often transports various pollutants, some of which settle and form sediments. In order
to have the comprehensive view of the ecological state of storm water runoff recipients, both water and sediments of the stream
must be assessed. In the Baltic Sea Area, the Water Framework Directive & HELCOM Recommendations aim to prevent or minimise
pollution caused by harmful substances arising from storm water runoff, in order to promote the ecological restoration of
the Baltic Sea—one of the most vulnerable seas. The aim of the study was to investigate the toxicity of bottom sediments of
a small storm water runoff recipient focusing on the potential impact of successive discharges of urban storm water. Some
storm water runoff quality parameters and the toxicity of bottom sediments of recipients was studied in this research.
Materials and methods During 9 years, at four discharge points, minimum four grab samples per year at each discharge point were taken for chemical
characterisation. General parameters (pH, SS, BOD7, CODCr and TPH) in liquid phase samples were analysed according to standard methods. Annual limit values were taken from the Lithuanian
EPA requirements for the management of storm water runoff with a focus on prevention and control of contamination. Eleven
composite samples of stream bottom sediments, each consisting of ten sub-samples, were collected in 2006. Toxicity screening
from sediments was performed using the plant Lepidium sativum according to modified I. Magone’s methodology (Magone I, Bioindication of phytotoxicity of transport emission. In: Kachalova
O-L, Zinatne (eds) Bioindication of toxicity of transport emissions in the impact of highway emissions on natural environment. Riga, pp 108–116, 1989). The level of toxic impact of Lepidium sativum (compared to control) was assessed according to the modified method of Wang (Rev Environ Contam Toxicol 126:88–127, 1992).
Results The mean pH of urban storm water runoff does not vary much from neutral, but range values are quite different, from 4.0 up
to 8.7. The highest concentration of SS reached 800 mg L−1, TPH—2.4 mg L−1, BOD7—300 mg O2 L−1 and CODCr—1,400 mg L−1. The SS was above the limit in 64% of total amount of grab samples, TPH—37%, BOD7—41% and CODCr—55%. The toxicity analysis of the bottom sediments showed varying toxicity of bottom sediments along the stream. From nine
analysed samples of bottom sediments, 30% had weak toxicity, 30% medium and 30% strong toxicity on the test organism plant
L. sativum. There was one single sample with no toxic effects, so that the results showed that urban storm water has an unacceptable
environmental impact on recipients. It was also indicated that storm water runoff discharge alone is not the potential source
of toxicity of bottom sediments. The litter demonstrated a weak toxicity of bottom sediments as well.
Discussion Most local authorities do not consider storm water runoff discharges to be a matter of great concern because they believe
that surface runoff arising from rainfall is still relatively clean. The study showed that the current method of monitoring
storm water runoff quality by chemical analyses is not the best tool for environmental impact assessment and must be combined
with toxicity tests of bottom sediments of recipients.
Recommendations and perspectives To avoid the environmental impact of storm water runoff more attention should be paid to the development and implementation
of storm water runoff pollution prevention measures. The study implies that future research concerning the relationships between
storm water runoff deposit characteristics and biological activities must be developed to evaluate the contamination potential
of stream sediment deposits for local aquatic ecosystems. Further studies should be developed to characterise the activities
of the microbial community of storm water runoff sediments, and to monitor bioremediation in situ. 相似文献

Two protein hydrolyzate–based fertilizers (PHFs), one from alfalfa (AH) and one from meat flour (MFH), were studied chemically and biologically. AH and MFH revealed a different degree of hydrolysis and a different amino acid composition. The biostimulant activity was investigated using two specific and sensitive bioassays of auxins and gibberellins. Extracts of AH and MFH elicited a gibberellin‐like activity and a weak auxin‐like one. To improve our understanding of the biostimulant activity, AH and MFH were supplied to maize plants and their effect on growth and nitrate metabolism was studied. Both PHFs increased root and leaf growth and induced morphological changes in root architecture. Besides, the treatments increased nitrate reductase (NR) and glutamine synthetase (GS) activities, suggesting a positive role of the two hydrolyzates in the induction of nitrate conversion into organic nitrogen. Moreover, treatments enhanced GS1 and GS2 isoforms in maize leaves. The latter isoform, amounting to 5‐ to 7‐fold the level of the former, appears to be a superior form in the assimilation of ammonia. The high NR and GS activities together with the high induction of GS isoforms indicate a stimulatory effect of the two PHFs on the assimilation of nitrate. In addition, a role of amino acids and small peptides of the two PHFs is suggested in the regulation of the hormone‐like activity and nitrogen pathway. 相似文献

Qualitative and quantitative analyses of betalain pigments in 10 cultivars/lines of prickly pear (Opuntia spp.) fruit grown in Mexico were conducted with reverse phase high-performance liquid chromatography-diode array detection (HPLC-DAD) coupled with electrospray mass spectrometry (ESI-MS). Betacyanins and betaxanthins were identified by comparison with the UV/vis and mass spectrometric characteristics as well as the retention times of semisynthesized reference betaxanthins. Data revealed that the ratio and concentration of betalain pigments are responsible for the color in the different cultivars, showing the highest betalains content in the fruit of purple colored Camuesa (O. robusta Wendl.) (8.1 mg/g dry fruit), which is comparable to that found in red beet Beta vulgaris L. ssp. Var. Pablo) (8.6 mg/g dry tissue). Yellow betalains were absent in Reyna (O. alba-carpa) prickly pear cultivar. A total of 24 known/unknown betalains were present in the prickly pear fruit samples studied, including 18 betaxanthins and 6 betacyanins. Our results indicate that prickly pear cultivars can be considered as a potential source of yellow and red natural colorants. 相似文献
