This study investigates the effect of NaOH and NaOCl treatments on chemical composition, morphology and crystalline structure of Agave americana L. fibers. These fibers have been subjected to NaOCl and NaOH alkali treatments at different concentrations.The percentages of lignin and hemicellulose show a decrease with alkaline treatments which, in turn, induces a modification of both morphological and crystalline structures.Unit cell dimensions and crystallite size were more affected with NaOH treatment than NaOCl one. This may result from the mercerisation process which occurs with caustic soda treatment.The observed defibrillization on the treated fiber surface proves the dissolution of the non-cellulosic components present in the fiber cell wall by NaOH and NaOCl treatments. These morphological changes may improve the interaction between matrix and fiber in composites. 相似文献

Chemical investigation of the EtOAc extract of the endophytic fungus Bionectria ochroleuca, isolated from the inner leaf tissues of the plant Sonneratia caseolaris (Sonneratiaceae) from Hainan island (China), yielded two new peptides, pullularins E and F (1 and 2) together with three known compounds (3–5). The structures of the new compounds were unambiguously determined on the basis of one- and two-dimensional NMR spectroscopy as well as by high-resolution mass spectrometry. The absolute configurations of amino acids were determined by HPLC analysis of acid hydrolysates using Marfey’s method. The isolated compounds exhibited pronounced to moderate cytotoxic activity against the mouse lymphoma cells (L5178Y) with EC50 values ranging between 0.1 and 6.7 µg/mL. 相似文献

The main problem facing farmers in the Nile Delta is water shortages at the ends of irrigation networks and canals. These problems have worsened as water demands have increased. Egypt’s Ministry of Water Resources and Irrigation is currently trying to avoid water deficits by returning agricultural drainage water to the irrigation canals. In the Kafr El-Sheikh Governorate, some canals have an oversupply in some months and deficits in others. Ministry officials started a project by constructing culverts connected canal ends with the main drain (Bhr Nashrat) to provide supplemental agricultural drainage backflows (SADB) channeled through these culverts. However, this return is not controlled, and flows are based only on differences in the hydraulic head. In this study, we evaluated the effectiveness of SADB to counteract water shortages when the water supply from head regulators (WSHR) is insufficient. Our analysis considered the adequacy of the water supply and indicators of dependability and equity of supply. We tested two water supply conditions: (1) WSHR only and (2) WSHR plus SADB. During the summer (May–September) of 2008, SADB significantly improved the system’s ability to meet the irrigation water requirements (IWR) in some months. Adequacy and dependability, therefore, improved from “fair” to “good.” During the following winter (October–April), SADB improved adequacy and equity only in March and April, since water availability was generally sufficient under WSHR. 相似文献

Receptor tyrosine kinases are key regulators of cellular growth and proliferation. Dysregulations of receptor tyrosine kinases in cancer cells may promote tumorigenesis by multiple mechanisms including enhanced cell survival and inhibition of cell death. Araguspongines represent a group of macrocyclic oxaquinolizidine alkaloids isolated from the marine sponge Xestospongia species. This study evaluated the anticancer activity of the known oxaquinolizidine alkaloids araguspongines A, C, K and L, and xestospongin B against breast cancer cells. Araguspongine C inhibited the proliferation of multiple breast cancer cell lines in vitro in a dose-dependent manner. Interestingly, araguspongine C-induced autophagic cell death in HER2-overexpressing BT-474 breast cancer cells was characterized by vacuole formation and upregulation of autophagy markers including LC3A/B, Atg3, Atg7, and Atg16L. Araguspongine C-induced autophagy was associated with suppression of c-Met and HER2 receptor tyrosine kinase activation. Further in-silico docking studies and cell-free Z-LYTE assays indicated the potential of direct interaction between araguspongine C and the receptor tyrosine kinases c-Met and HER2 at their kinase domains. Remarkably, araguspongine C treatment resulted in the suppression of PI3K/Akt/mTOR signaling cascade in breast cancer cells undergoing autophagy. Induction of autophagic death in BT-474 cells was also associated with decreased levels of inositol 1,4,5-trisphosphate receptor upon treatment with effective concentration of araguspongine C. In conclusion, results of this study are the first to reveal the potential of araguspongine C as an inhibitor to receptor tyrosine kinases resulting in the induction of autophagic cell death in breast cancer cells. 相似文献

Our previous studies showed that several sipholane triterpenes, sipholenol A, sipholenone E, sipholenol L and siphonellinol D, have potent reversal effect for multidrug resistance (MDR) in cancer cells that overexpressed P-glycoprotein (P-gp/ABCB1). Through comparison of cytotoxicity towards sensitive and multi-drug resistant cell lines, we identified that the semisynthetic esters sipholenol A-4-O-acetate and sipholenol A-4-O-isonicotinate potently reversed P-gp-mediated MDR but had no effect on MRP1/ABCC1 and BCRP/ABCG2-mediated MDR. The results from [3H]-paclitaxel accumulation and efflux studies suggested that these two triterpenoids were able to increase the intracellular accumulation of paclitaxel by inhibiting its active efflux. In addition, western blot analysis revealed that these two compounds did not alter the expression levels of P-gp when treated up to 72 h. These sipholenol derivatives also stimulated the ATPase activity of P-gp membranes, which suggested that they might be substrates of P-gp. Moreover, in silico molecular docking studies revealed the virtual binding modes of these two compounds into human homology model of P-gp. In conclusion, sipholenol A-4-O-acetate and sipholenol A-4-O-isonicotinate efficiently inhibit the P-gp and may represent potential reversal agents for the treatment of multidrug resistant cancers. 相似文献

The effects of bias (over‐ and underestimates) in estimates of disease severity on hypothesis testing using different assessment methods was explored. Nearest percentage estimates (NPE), the Horsfall–Barratt (H‐B) scale, and two linear category scales (10% increments, with and without additional grades at low severity) were compared using simulation modelling to assess effects of bias. Type I and type II error rates were used to compare two treatment differences. The power of the H‐B scale and the 10% scale were least for correctly testing a hypothesis compared with the other methods, and the effects of rater bias on type II errors were greater over specific severity ranges. Apart from NPEs, the amended 10% category scale was most often superior to other methods at all severities tested for reducing the risk of type II errors. It should thus be a preferred method for raters who must use a category scale for disease assessments. Rater bias and assessment method had little effect on type I error rates. The power of the hypothesis test using unbiased estimates was most often greater compared with biased estimates, regardless of assessment method. An unanticipated observation was the greater impact of rater bias compared with assessment method on type II errors. Knowledge of the effects of rater bias and scale type on hypothesis testing can be used to improve accuracy and reliability of disease severity estimates, and can provide a logical framework for improving aids to estimate severity visually, including standard area diagrams and rater training software. 相似文献

This study was conducted to evaluate the effects of supplemental taurine on reproductive performance of Nile tilapia (Oreochromis niloticus) broodstock fed soybean meal‐based diets. Four isonitrogenous (350 g kg?1 protein), isocaloric (18 MJ kg?1) diets were formulated to contain 0, 5, 10 and 15 g kg?1 taurine. The diets were fed to triplicate groups of juvenile Nile tilapia (10–15 g average body weight) at a female: male ratio of 3 : 1, to apparent satiation, three times per day for 130 days. The size at first maturation decreased with increasing dietary taurine to 10 g kg?1 and levelled off with further taurine supplementation. The time to first spawning was also significantly shorter at 10 g kg?1 taurine level. Spawning performances, including spawning frequencies, total number of spawnings per tank, number of spawnings per female and absolute fecundity, were all significantly improved with increasing dietary taurine up to 10 g kg?1. However, the quadratic regression analyses indicated that the maximum spawning performance occurred at 8 g kg?1 of supplemental taurine. Eggs produced from broodstock fed 10 g kg?1 taurine exhibited significantly higher hatchability and required shorter time for hatching and yolk‐sac absorption and also resulted in higher larval weight than at other dietary taurine levels. The highest egg protein, total amino acids and taurine were also obtained at 10 g kg?1 taurine. These results suggest that 8 g kg?1 dietary taurine is required for optimum reproductive outputs of Nile tilapia broodstock. 相似文献

Water as a habitat is highly variable in terms of the physico‐chemical properties. The aim of the studies was to analyze the quality of the technology by evaluating the non‐specific cellular defence potential of rainbow trout (Oncorhynchus mykiss) in diversified intensity culture systems and in different periods of the breeding cycle. The evaluation was conducted in order to establish the scientific basis for the development of principles for recognition of the culture of this species as ‘organic’. Six rainbow trout rearing farms were selected for the studies and divided into two equal groups according to the production technology: farms with a flow through system (with single water usage – OOH) and facilities with multiple water usage via its backflow (re‐circulating system – RAS). The samples for tests were taken from 20 individuals from each fish farm. In all fish that originated both from OOH‐type farms and RAS operations, an increase in the activity of non‐specific cellular immunity in the autumn was observed. Statistically significant reduction in the activity of cellular defensive mechanisms in small (300–500 g) and big (501–850 g) fish occurred on two 3‐OOH and 3‐RAS farms in spring and autumn in 2011. During these periods, the research has shown the presence of Infectious Pancreatic Necrosis Virus (IPNV), which provoked an immune suppression. Based on the obtained results it is concluded that there were no significant differences in the parameters of non‐specific cellular immunity in rainbow trout originating from different farming systems OOH and RAS. 相似文献
