Microsphaeropsis sp. strain P130A was evaluated for the control of tuber-borne inoculum of Rhizoctonia solani based on the viability of sclerotia produced in vitro and on both the viability and production of tuber-borne sclerotia. The interactions between the antagonist and the pathogen, as well as the effect of the toxins produced by the antagonist on mycelial growth of R. solani were studied using transmission electron microscopy. On sclerotia produced in vitro, for all incubation periods (1 to 42 days), Microsphaeropsis sp. significantly reduced germination. Percent germination of sclerotia treated with Microsphaeropsis sp. decreased with increasing incubation period from an average of 82.0% after 1 day to stabilize at an average of 5.8% after 35 days. Similarly, percent germination of tuber-borne sclerotia was significantly lower when tubers were treated with Microsphaeropsis sp. Both 2% formaldehyde and Microsphaeropsis sp. treatments significantly reduced sclerotia germination to approximately 10% after 42 days of incubation at 4 degrees C. Furthermore, on tubers treated with the antagonist, the number of sclerotia per square centimeter decreased from 1.6 to 0.5 during the 8 months of storage at 4 degrees C, whereas an increase from 1.2 to 7.8 sclerotia per square centimeter was observed on untreated tubers. Microsphaeropsis sp. (strain P130A) colonized hyphae of R. solani within 4 days after contact on culture media. Transmission electron microscopic observations showed that the antagonist induced a rupture of the pathogen plasma membrane and that a chitin-enriched matrix was deposited at sites of potential antagonist penetration. Host penetration was not associated with pathogen cell wall alterations, which occurred at the time of progress of the antagonist in the pathogen cytoplasm. In the presence of a crude extract of Microsphaeropsis sp., cells of R. solani showed cytoplasm disorganization and breakdown of plasma membranes. Antibiosis and mycoparasitism were involved in the antagonism of R. solani by Microsphaeropsis sp., but the sequence by which these events occur, as well as the significance of wall appositions produced by R. solani, is yet to be established.  相似文献

The resistance to insecticides of three Sudanese strains of A. gossypii (Glov.) collected from cotton fields in the Sudan Gezira Scheme over three seasons (1988, 1989, 1990) and that of two French strains was studied in the laboratory. When compared with a known susceptible strain, the aphids were found to be resistant-to the eight insecticides tested. Evolution of resistance in Sudanese strains during the three crop seasons was observed. Assay of aphid homogcnate for carboxytesterase activity towards the substrates α-naphthyl acetate and β-naphthyl acetate showed that there was no enhancement of this class of enzyme and thus it was not a cause of resistance in this species. A study of interaction between the acetylcholinesterase (AchE) and pirimicarb established the kinetics of the inhibition process. I50 values were found to be much higher for the Sudanese strains than for the susceptible strain. First-order inhibition kinetics revealed that resistance towards pirimicarb in Sudanese-strains was caused by a modified AchE which had a reduced affinity (higher Ka value) and poor carbamylation ability (lower K2 value) for pirimicarb. The resistance mechanisms for the other insecticides remain to be studied.  相似文献

Inoculation of leaves of resistantPlatanus occidentalis and susceptiblePlatanus acerifolia leaves withCeratocystis fimbriata f. sp.platani, the canker stain disease agent, induced foliar necrosis and biosynthesis of two phytoalexins, scopoletin and umbelliferone. Foliar symptoms keep localized and accumulation of coumarin phytoalexins was rapid for incompatible interactions. Necrosis spread widely and accumulation of these phenolic compounds was much later and lower for compatible interactions. This experiment was conducted to determine the effects of castration on some metabolic alterations concerning proteins, lipids and minerals metabolism in both immature and mature rabbits. Twenty male New Zealand White rabbits (10 immature and 10 mature) were used. Blood samples were collected from the immature and mature intact animals (control non castrated). Moreover, the castrated animal groups (immature and mature) were sampled at 2, 4, 6 and 8 weeks after castration. The obtained results revealed that, castration caused a significant decrease in serum total protein, albumin, alpha 1-globulins and alpha 2-globulins concentrations. Meanwhile, gamma-globulins level was decreased temporarily at 2 weeks after castration in mature rabbits. beta-globulins level revealed non significant decrease at 2 weeks after castration in immature rabbits followed by a non significant increase. In mature rabbits beta-globulins level non significantly increased. A significant decrease in serum total nucleic acid concentrations was observed after castration throughout the experimental periods, whereas the value of serum uric acid level markedly increased after castration. Serum lipids concentrations (total lipids, total cholesterol, phospholipids and non esterified fatty acids) were increased after castration. The value of serum trace elements including copper, iron, zinc and manganese concentrations showed a significant decrease after castration. The decrease in serum copper and zinc levels were markedly observed in mature castrated rabbits. Also, serum sodium and potassium concentrations were decreased after castration. Testosterone propionate administration in mature castrated rabbits restored most of the investigated serum blood parameters to the levels of intact non castrated mature rabbit. It could be concluded that castration (deficiency of testosterone) induced marked decrease in protein synthesis and increased protein degradation, but serum lipids markedly elevated after castration in both immature and mature animals. Also, castration caused a marked decrease in some serum minerals and trace elements.  相似文献

In situ and in vivo digestibility experiments were conducted to determine the acceptability, digestibility, and safety of a return chewing gum/packaging (G/P) material mixture when fed to steers. In the in situ experiment, both ruminal and intestinal disappearances were measured. Two ruminally and duodenally cannulated steers, which were given free access to alfalfa hay (AH), were used in this study. Duplicate Dacron bags containing the G/P were incubated in the rumen for 0, 3, 6, 12, 24, and 48 h. After ruminal incubation, the 12-, 24-, and 48-h bags were placed in the duodenum and collected in the feces to determine intestinal disappearance. In situ ruminal DM disappearance was greater than 70% for all substrates tested at 0 h, indicating high solubility of the substrates in water, and began to reach a plateau after 12 h of incubation. Intestinal in situ disappearance was not different (P>.25) from zero. In the digestion trial, four ruminally cannulated steers (337+/-21.3 kg BW; mean +/- SD) were used in a 4x4 Latin square design with the following treatments: 0) 50% corn (C), 50% AH; 10) 45% C, 45% AH, 10% G/P; 20) 40% C, 40% AH, 20% G/P; 30) 35% C, 35% AH, 30% G/P. Steers fed G/P-containing diets had greater (P<.01) DMI than the control steers. Increasing the G/P resulted in a linear (P<.05) increase in DMI. Apparent DM digestibility tended to be higher (P<.10) for the G/P-containing diets than for the control. A quadratic effect (P<.05) on digestible DMI was observed, with greater (P<.01) digestible DMI values for G/P-containing diets (4.8 vs. 5.8 kg/d). Digestible organic matter and total nonstructural carbohydrate intakes followed trends similar to those of DM. Apparent aluminum digestibility of G/P-containing diets was not different (P>.13) from zero. The level of G/P in the diet had no effect (P>.2) on total VFA concentration or ruminal pH. There was a linear decrease (P<.01) in the molar percentage of isobutyrate and isovalerate in addition to a linear increase (P<.01) in butyrate and valerate with increasing levels of G/P. There was a quadratic effect (P<.01) on molar proportions of acetate and propionate and on the acetate:propionate ratio. Results of both experiments suggest that G/P may be fed to safely replace up to 30% of corn-alfalfa hay diets for growing steers with advantages in improving DMI and digestibility.  相似文献

Haematological estimations and serum biochemical analyses were made on 100 samples collected from clinically healthy Hijin racing camels (Camelus dromedarius) in Kuwait. The red blood cell counts, packed cell volume, haemoglobin concentration, mean corpuscular volume, mean corpuscular haemoglobin, mean corpuscular haemoglobin concentration and total white blood cell counts were estimated. In the serum biochemical analyses, sodium, potassium, iron, calcium, phosphorus, magnesium, total bilirubin, blood urea nitrogen, creatinine, glucose, total protein and cholesterol concentrations were measured, as were the alanine aminotransferase, aspartate aminotransferase, -glutamyltransferase, creatine kinase and alkaline phosphatase activities. The results are discussed in relation to other findings reported in camels.  相似文献

Two promising selected land races of safflower ( Carthamns tinctorious L.) designated as line 11 and line 13, together with the local variety Giza 1 , were examined under three levels of moisture depletions, i.e. 40, 60 and 80 % ASMD at Fayoum, Middle Egypt during 1986/87 and 1987/88 seasons. In both growing seasons, the second irrigation treatment i.e. 60 % ASMD gave the highest means for growth characters as well as seed yield and most of its components. Seed oil content per cent exhibited an increased as soil moisture depletion increased with no significant difference between entries. Safflower entries showed clear differences in some yield components which compensate each other and resulted in no significant difference in seed yield per hectar.
Consumptive use of water by safflower plants increased as the available soil moisture around the root zone increased. No significant differences were found in water consumed by safflower entries used in all experiments.
In the two experimental seasons, the best water use efficiency (WUE) for seed production was obtained also from the second treatment (60 % ASMD). The line 11 gave the best WUE.  相似文献

Fructans are fermentable carbohydrates and include short-chain fructooligosaccharides (scFOS), inulin, and hydrolyzed inulin (oligofructose, OF). Two studies with dogs were designed to examine the effects of low concentrations of fructans on nutrient digestibilities, fecal microbial populations, and endproducts of protein fermentation, and fecal characteristics. In Exp. 1, 11 adult male beagles were fed corn-based, kibbled diets supplemented with or without OF to provide 1.9 +/- 0.6 g/d. Dietary inclusion of OF decreased (P < 0.05) nutrient digestibilities, but did not affect fecal characteristics. Increasing OF concentration tended (P < 0.06) to linearly decrease fecal ammonia concentrations, but not those of branched-chain fatty acids (BCFA), amines, indole, or phenols. Fecal concentrations of total short-chain fatty acids (SCFA) and butyrate tended to be higher in OF-supplemented dogs (P < 0.10), as was the ratio of bifidobacteria to total anaerobes (P = 0.15). In Exp. 2, ileally cannulated adult female hounds were fed a meat-based kibbled diet and were assigned to four scFOS treatments (0, 1, 2, or 3 g/d) in a 4 x 4 Latin square design. Ileal nutrient digestibilities tended to increase (P < 0.15) with increasing concentrations of scFOS. On a DMI basis, fecal output tended to decrease linearly (P < 0.10) in response to increasing scFOS supplementation, whereas fecal score tended to exhibit a quadratic response (P = 0.12). In general, fecal concentrations of SCFA, BCFA, ammonia, phenols, and indoles were not altered by supplemental scFOS. Supplementation of scFOS increased fecal concentrations of total aerobes (P < 0.05) and decreased concentrations of Clostridium perfringens (P < 0.05). From these data, it seems that low levels of supplemental fructans have divergent effects on nutrient digestibility and fermentative endproducts, but do not adversely affect nutrient digestibility or fecal characteristics and may improve colonic microbial ecology in dogs.  相似文献
