Conventional methods of measuring labile chemical species of trace metals in soil solutions, such as chemical competition following centrifuging, are inadequate if the speciation changes during sampling and extraction. A new technique, diffusive gradients in thin films (DGT), measures labile species of trace metals in natural waters and sediments in situ. A well-defined diffusive gel layer distinguishes it from other resin-based techniques. It perturbs the soil in a controlled way by introducing an in situ local sink for metal ions. Resulting fluxes to the device are quantitatively measured, allowing assessment of re-supply kinetics and in some cases measurement of in situ soil solution concentrations. We used DGT to measure fluxes of Cd, Co, Cu, Ni, Pb and Zn in a sludge-treated soil at various moisture contents (27–106%). Replicate measurements showed that the precision of DGT-measured fluxes was within 10%. For moisture contents exceeding the field capacity (42%), the DGT response reflected soil water concentrations. At smaller moisture contents, changes related to tortuosity and dilution were reflected in the measurements. This technique has the potential for in situ measurements in the field where it should provide quantitative flux data on individual soils and provide a good surrogate for bioavailable metal.  相似文献

Monoglycerides are widely used in the baking industry because of their antistaling effects, mainly suppressing crumb firming. Commercial monoglycerides are normally prepared from hydrogenated fats, with stearate being the most common fatty acid. In a previous study, monoglycerides such as monopalmitate (C16) and monostearate (C18) had positive effects on Canadian short process bread but no improvements on sponge‐and‐dough process (SDP) bread. The objective of this study was to investigate the effects of saturated monoglycerides of varying fatty acid chain length (C14–C22) on SDP breadmaking quality by using volume judgment, crumb image analysis, and texture measurements. Higher levels (1.00–1.50%) of all monoglycerides (C14, C16, and C18) significantly (P < 0.05) increased loaf volume and cell diameter. The larger cell diameter with increasing levels of these monoglycerides may have resulted from softer, more extensible dough handling properties and greater gas cell stability during baking. Addition of C16 and C18 caused the largest increase in crumb softness with increasing monoglyceride levels but showed relatively low resilience, which might be related to larger loaf volume (i.e., lower density of bread). However, addition of blended monoglycerides C14+C16 increased crumb softness and loaf volume while partially retaining resilience. Each monoglyceride had a different function in breadmaking quality and somewhat positive effects on SDP.  相似文献

Classical swine fever virus, like many enveloped viruses, may be regarded as moderately fragile. It shows a short but variable survival time in the environment, depending on physical conditions, but may remain viable for prolonged periods in favourable circumstances as found, for example, in stored meat. Published information is reviewed on the survival time of the virus under a variety of physical and chemical conditions, including the variable influence of environmental factors. The principles of virus inactivation by heat, pH or chemicals are discussed, with a consideration of the principles of containment for laboratories working with the virus.  相似文献

The objectives of this study were (1) to determine the prevalence of pathological abdominal adhesion formation following exploratory laparotomy; (2) to establish the site of adhesion formation and its relationship to the initial lesion; (3) to ascertain whether the development of intra-abdominal adhesions decreases long-term survival and (4) to identify risk factors for adhesion formation. Of 1014 horses treated surgically for acute gastrointestinal disease, 113 (10.1%) were subjected to repeat laparotomy, with surgical records available for 99 of these cases. Pathological adhesions were the most common diagnosis at repeat laparotomy (28%), followed by complications associated with the anastomosis (16%). Adhesions were not associated with the site of the primary lesion, resection, or endotoxaemia, consistent with the hypothesis that surgical trauma is the most important stimulus in adhesion formation. Together these findings strongly support the need for pan-abdominal, rather than site-specific adhesion prevention measures in all horses undergoing exploratory laparotomy.  相似文献

The utilization of glucose and citrate either with or without being mixed with one of four commonly-occurring soil mineral phases was assessed using both bacterial cultures and soil incubations. Citrate and glucose, both -labelled, were added separately to bacterial cultures. In each case they were rapidly degraded with >60% recovered as CO2. In the presence of soil the respired proportion declined especially for glucose. The presence of mineral phases (illite–mica, kaolinite, mixed soil clay or ferric hydroxide) had little observable affect on the utilization of glucose in either bacterial cultures or soil. However, ferric hydroxide did induce an alteration in the way was partitioned within the microbial cells. In contrast, citrate degradation was greatly reduced over short periods (6–22 h) in the presence of the three clay materials and almost completely inhibited by the presence of ferric hydroxide. The importance of substrate sorption in the context of carbon utilization in the rhizosphere is discussed.  相似文献

Nitrate () leaching can cause elevated concentrations of ‐N in water, which can have adverse impacts on water quality and human health. In grazed pasture systems, most of the ‐N leaching occurs beneath animal urine‐N deposits. The objective of this study was to investigate the effect of four different pasture species compositions [perennial ryegrass/white clover (P. ryegrass WC), tall fescue/white clover (T. fescue WC), Italian ryegrass/white clover (It. ryegrass WC) and perennial ryegrass/Italian ryegrass/white clover/red clover/chicory/plantain (Diverse)] on ‐N leaching losses from animal urine patches, and to examine the relative importance of root system architecture and seasonal activity to reduce ‐N leaching losses. The results show that ‐N leaching losses were 24–54% lower beneath It. ryegrass WC than other pasture species. Total dry matter (DM) yield in the season following establishment was 11–58% greater in the It. ryegrass WC pasture, while average winter daily N uptake rate of It. ryegrass WC over the two seasons was on average 58% greater than P. ryegrass WC and T. fescue WC. In the second season, the P. ryegrass WC and T. fescue WC pastures had up to 140 and 82% more roots between 0 and 40 cm depth, respectively, than the other pasture species compositions. These results suggest that in grazed pasture systems, high plant winter activity (plant growth/root metabolic activity) is more important than specific root architecture (e.g. deep roots) to reduce ‐N leaching losses.  相似文献

Abstract

A cool season perennial grass with a root system capable of penetrating hardpans and which can accumulate adequate Mg to prevent deficiencies in forage is needed in the Coastal Plain region. A greenhouse experiment was conducted to determine the effects of magnesium (Mg) concentration in nutrient solution and root diameter on Mg uptake and growth of tall fescue (Festuca arundinacea Schreb.). Propagules of two fescue clones with large root diameter (LDR >1 mm), two clones with small root diameter (SDR <0.8 mm), and a single clone from ‘Kentucky 31’ (Ky‐31) were transferred into 12‐liter tanks containing Mg concentrations of 3, 21, 42, 125, 250, and 500 μM as MgSO4 and grown for 39 or 70 days. Leaf Mg concentration was increased linearly with Mg solution concentration in LDR clones for a 39‐day growth period (Harvest 1), but increased according to a cubic equation in the SDR clones and the Ky‐31. Predicted leaf Mg concentration as a function of solution Mg followed a cubic equation for a 70‐day growth period (Harvest 2) in all clones. Predicted root Mg concentration was linearly related to Mg solution concentration for the LDR clones and the Ky‐31, but followed a cubic equation for the SDR clones for the first growth period. For the second growth period, the root Mg concentration of the SDR clones and the Ky‐31 was increased linearly, while the LDR clones followed a quadratic equation. Magnesium uptake followed a cubic equation with Mg solution concentration for both growth periods on all tall fescue clones. This nonlinear variation of Mg uptake and plant Mg concentration with respect to solution Mg concentration strongly suggests that a dual uptake mechanism might have been present in tall fescue clones. Root volume was greater in the SDR than LDR clones or Ky‐31 for both growth periods. The Ky‐31 had the greatest leaf and root dry weight for both growth periods, while the LDR clones had the lowest.  相似文献

The biochemical changes in fresh cow manure caused by the earthworm Eisenia andrei (Bouché) were measured over a period of four months, under controlled laboratory conditions. Earthworms were introduced into each of four plastic containers (0.4 × 0.27 × 0.15 m) containing fresh cow manure (2500 g), and four containers containing manure but without earthworms served as controls. Earthworms reduced the pH and decreased the moisture content in the manure. The C:N ratio of the manure with or without earthworms decreased progressively from 36 to 21. The ash and total nitrogen contents increased greatly for a few weeks after the introduction of earthworms, reflecting a rapid breakdown of carbon compounds and mineralization of nitrogen by the earthworms. CO2 evolution decreased rapidly (44%) one week after the introduction of earthworms, and continued at a lower rate throughout the 17 weeks (51% reduction as compared to 22% without earthworms), indicating increasing stability of the organic matter. Earthworms reduced microbial biomass early in the process, but enhanced nitrogen mineralization and increased the rates of conversion of ammonium-nitrogen into nitrate. The major general effect of earthworms on the organic wastes was to accelerate the maturation of the organic wastes as demonstrated by enhanced growth of lettuce and tomato seedlings.  相似文献
