Four natural products were isolated from the fungus Botryosphaeria rhodina, and their effects on photosynthesis were tested. Only lasiodiplodin (1) inhibited ATP synthesis and electron flow from water to methylviologen; therefore, it acts as a Hill reaction inhibitor in freshly lysed spinach thylakoids. Photosystem I and II and partial reactions as well as ATPase were measured in the presence of 1. Three new different sites of 1 interaction and inhibition were found: one at CF1, the second in the water-splitting enzyme, and the third at the electron-transfer path between P680 and QA; these targets are different from that of the synthetic herbicides present. Electron transport chain inhibition by 1 was corroborated by fluorescence induction kinetics studies.  相似文献

Two rapid tests evaluated in dogs considered to be of high risk of infection with the Chagas parasite Trypanosoma cruzi using two immunochromatographic assays: Trypanosoma Detect™ for canine, InBios, Seattle, WA and CHAGAS STAT-PAK™ assay, Chembio Diagnostic Systems, Medford, NY, in south central Louisiana. For this purpose a serological survey was carried out in a total of 122 dogs and a serum bank was created. These 122 animals were first tested by IFAT that was used as the standard test. From the serum bank 50 samples were tested using the two rapid Chagas assays and results compared to the standard test IFAT. The serological survey using IFAT showed a prevalence of T. cruzi infection in 22.1% of the tested dogs. In the immunochromatographic assays, 13 and 11 animals were positive on rapid assay: Trypanosoma Detect™ for canine, InBios and CHAGAS STAT-PAK™, Chembio Diagnostic Systems, respectively compared to 11 positive by IFAT. These two immunochromatographic tests have shown high susceptibility and specificity compared to our standard method IFAT. The rapid, easy and accurate screening assays used in conjunction with confirmatory tests, would be an excellent tool for veterinarians to diagnose T. cruzi infection. Early detection of T. cruzi infection may prevent complications through an effective treatment. Greater awareness by veterinarians of the risk, clinical findings, history along with diagnostic methods will contribute greatly to an understanding of the true prevalence of Chagas disease in dogs in Louisiana.  相似文献

Precocious lines of Eimeria acervulina "Cu" and "I" strains were obtained after 25 passages of oocysts in chickens that showed a shortening of the prepatent period for first oocyst output from 96 h to 81 and 82 h, respectively. Both precocious lines were evaluated for pathogenicity using as criteria weight gain, lesion score and total oocyst production. Infection of the "Cu" precocious line in chickens showed a high weight gain, low lesion score and low oocyst production, when compared to parent strain infected chickens. However, the results did not show a significant difference in relation to the criteria used above for the E. acervulina "I" precocious line when compared to its parent strain. This suggests a low degree of attenuation for the "I" strain but good attenuation for the precocious "Cu" line. The histopathological observations of chickens infected with the E. acervulina "Cu" parent strain and precocious line, comparing life cycle and intestinal lesions, showed: (1) parasite stages only in the border cells of infected chicken intestinal villi, for the precocious line; (2) parasite stages in the border cells of the intestinal villi and submucosa cells near the Lieberkühn glands of the intestine; and (3) high degree of inflammatory cells around the parasites in chickens infected with the parent strain. The "Cu" strain was also characterized for sensitivity against eight anticoccidial drugs. Sensitivity was observed for four anticoccidial drugs and partial resistance for four other drugs, although the strain had never had previous contact with anticoccidial drugs, suggesting the presence of a natural resistance factor. This Brazilian E. acervulina "Cu" precocious line showed attenuation for pathogenicity in chickens, suggesting that it could be a suitable strain for use as a live vaccine in Brazil.  相似文献

REASONS FOR PERFORMING STUDY: This study reports a neurological disease unrecognised until now in ponies in southern Brazil. HYPOTHESIS: Epidemiological data strongly suggests that the ingestion of Sida carpinifolia is involved in the aetiology. We tested the hypothesis that it is an acquired lyosomal storage disease. METHODS: Following the death of 3 ponies, all ponies from the premises were closely monitored; epidemiological data and clinical findings carefully recorded. Fragments of several organs, including CNS, were fixed in neutral formalin and embedded in paraffin-wax. Sections were stained with haematoxylin and eosin. Representative sections of the cerebellum and trigeminal ganglia were submitted to lectin histochemical procedures. RESULTS: The neurological disorder, characterised by stiff gait, muscle tremors, abdominal pain and death, was observed on a farm with 3 hectares of pasture. Three of 11 ponies died 15-20 days after they had been introduced into a new paddock heavily infested by the plant Sida carpinifolia. No significant gross lesions were observed. The main histological findings included multiple cytoplasmatic vacuoles in swollen neurones in the brain, cerebellum, spinal cord, autonomic ganglia (trigeminal and celiac ganglia), and submucosal and myenteric plexus of the intestines. In the kidneys, there was marked vacuolation of the proximal convoluted tubular cells. Sections of cerebellum and trigeminal ganglion were submitted to lectin histochemistry. The vacuoles in different cerebellar and ganglion cells reacted strongly to the following lectins: Concanavalia ensiformis, Triticum vulgaris and succinylated-Triticum vulgaris. CONCLUSIONS: The pattern of staining coincides with that of both swainsonine toxicosis and inherited mannosidosis reports. The histopathological changes were similar to those described in S. carpinifolia spontaneous and experimental poisoning in goats. This disease seems to be similar to Swainsona, Oxytropis and Astragalus toxicosis. POTENTIAL RELEVANCE: S. carpinifolia should be evaluated as a possible cause in the diagnosis of equine neuropathies.  相似文献

Congenital Neospora caninum infection was diagnosed in a Saanen goat from a farm in southern Brazil. The kid was unable to nurse and had difficulty rising, ataxia, and opistothotonos. The neurologic signs became more severe 3 days after birth, when it was euthanized. No gross lesions were observed at necropsy. Multifocal infiltrates primarily of mononuclear cells, nodular microgliosis, and perivascular cuffs of lymphocytes, plasma cells, and few neutrophils were observed in the brain, mostly in the cortex and adjacent to ventricles. Rare multinucleate giant cells were observed adjacent to inflammatory foci. Several tissue cysts with a thick wall that reacted strongly with polyclonal antiserum to N. caninum were in the cerebral cortex and medulla oblongata. Lesions were also present in heart, lungs, and liver, but N. caninum tachyzoites were not found. Distinguishing features in this goat kid included neurologic impairment resulting from congenital infection with N. caninum and the presence of granulomatous inflammation with rare giant cells associated with degeneration of tissue cysts.  相似文献

The silverleaf whitefly, Bemisia tabaci (Gennadius) biotype B (Hemiptera: Aleyrodidae), is considered one of the most important pests of cowpea (Vigna unguiculata L. Walp.), limiting the productivity of this crop worldwide. Chemical control is still the main strategy for the management of populations of this insect. However, due to the harmful effects of pesticides on the environment and to humans, less injurious alternatives have been investigated. Along this line, the use of resistant genotypes can be a valuable tool for the control of the silverleaf whitefly. In this paper, we investigate some biological aspects of B. tabaci biotype B confined on 14 genotypes of cowpea. We evaluated the incubation period, egg viability, duration of nymphal stages, total duration of the juvenile phase, instar mortality and total mortality of the immature stage. The genotype MNC 99-541 F21 exhibited antibiosis against the whitefly, prolonging the lifecycle of the insect. The genotypes Canapu, BRS-Urubuquara and TE97-304 G-4 also exhibited antibiosis, causing high nymphal mortality. These results may help in breeding programmes to develop cowpea lines with resistance to B. tabaci biotype B.  相似文献

Lectins are proteins or glycoproteins with the ability to link to carbohydrates at the cell surface in a specific and reversible manner. Studies have showed that lectins are demonstrate immunomodulatory and antitumor agents. This study aims to evaluate the effect of lectin extracted from the seeds of Canavalia brasiliensis (ConBr) on murine melanoma B16F10 cells by analyzing cell viability, apoptosis index, cell migration, production of cytokines and nitric oxide (NO). Results showed that ConBr was able to reduce cell viability and thwart apoptosis, which could be observed by decrease in cell migration. ConBr also induced NO and IL-12 synthesis. Altogether, these data demonstrate the potential of ConBr as a therapeutic agent for melanoma.  相似文献

This work evaluated the preference of Ascia monuste orseis among 29 cultivars of collard greens through free- and no-choice assays using female adults (attractiveness and oviposition) and third-instar larvae (feeding preference), in both laboratory and greenhouse conditions. In free-choice tests with female adults, the L, V, Y, H, A, and Z cultivars produced oviposition non-preference in butterflies. Cultivars R, P, C, and D produced feeding non-preference in third-instar larvae of A. monuste orseis in free- and no-choice tests. Our findings may be useful for genetic improvement focusing on the development of B. oleracea var. acephala cultivars resistant to A. monuste orseis.  相似文献
