The availability of safe, commercially prepared stock feed for production animals is an important step in ensuring animal health and welfare and the safety of food animal products for human consumption. Animal feed quality assurance programs include microbiological monitoring of raw materials, mill equipment and finished feed. Over a period of 16 years, 23,963 samples for Salmonella culture and serotyping were collected from 22 stock feed mills. A multivariable generalized linear mixed model (GLMM) was used to identify mill and sample type factors that increase the odds of detecting Salmonella. The odds of detecting a Salmonella positive sample was greatest in samples from raw materials and in mills that processed restricted animal material (RAM). The percentage of positive samples ranged from 7.2% in 2003 to 2.8% in 2017. Of the 1,069 positive samples, 976 were serotyped with 61 different Salmonella serotypes isolated. The serotype most frequently isolated from raw materials was S. Agona, (n = 108) whilst S. Anatum was the serotype most frequently isolated from equipment and finished feed (n = 156). The diversity of Salmonella serotypes differed between mills and different stages of the production line. Microbiological monitoring in the commercial preparation of animal feed in Australian stock feed mills guides the implementation of quality control measures and risk mitigation strategies thereby reducing the prevalence and diversity of potentially zoonotic bacteria such as Salmonella, enhancing food safety for both animal and consumer.  相似文献

The interaction between the sporocysts of Sarcocystis muris and mouse peritoneal macrophages was studied both in vitro and in vivo in an attempt to determine whether or not resident peritoneal macrophages might effect the excystation of S. muris sporozoites from sporocysts injected intraperitoneally. Sporocysts of S. muris were phagocytosed by peritoneal macrophages both in vitro and in vivo. The addition of either unheated mouse serum or fetal calf serum did not significantly alter the level of phagocytosis. The percentage of phagocytosis in vivo and by thioglycolate-, proteose peptone- and BCG-elicited macrophages in vitro was greater than that shown by unstimulated macrophages in vitro. After 8 h incubation in vivo and in vitro a small proportion of sporocysts (less than 5%) was seen to have collapsed walls and up to 5% to have stained sporozoites, suggesting increased permeability of the sporocyst wall. The significance of increased permeability of the cyst wall in the process of sporozoite excystation is discussed.  相似文献

Unpurified DNA derived from cultures of equine fetal kidney cells infected with either equine herpesvirus type 1 or equine herpesvirus type 4 was amplified by the polymerase chain reaction using one pair of oligonucleotide primers. Restriction endonuclease digestion of the amplified segments with PvuII, followed by electrophoresis, revealed restriction fragment length polymorphisms which enabled the two virus types to be differentiated.  相似文献

Cattle are known reservoirs and asymptomatic excretors of Cryptosporidium, a protozoan parasite that causes severe and protracted diarrhoea in people. The incidence of Cryptosporidium was investigated in 288 matched samples taken from beef carcases of 1 g samples of faeces retrieved immediately after de-legging, 25 cm2 samples of beef excised from the rump of uneviscerated carcases, and 25 cm2 samples of beef excised from the brisket area of eviscerated carcases. Cryptosporidium species were detected in 21 of the faecal samples after salt flotation and immunofluorescent microscopy. The species isolated from the positive samples were identified by restriction fragment length polymorphism and PCR as Cryptosporidium andersoni (54.5 per cent) and Cryptosporidium parvum genotype 2 (45.5 per cent). In the faecal samples, there was a significantly higher prevalence of the parasite in samples taken in summer (May to July) and winter (November to January) than in spring or autumn. No Cryptosporidium species were recovered from any of the beef samples.  相似文献

Abstract

AIMS: To determine the concentration of Campylobacter spp. as well as faecal indicator bacteria; faecal coliforms, Escherichia coli and enterococci in the faeces of healthy adult horses in a sample of properties in the Canterbury region of New Zealand.

METHODS: The faeces of healthy adult horses (n=59), including ponies, pleasure horses and Thoroughbreds, were collected from eight properties around Christchurch, New Zealand. The faeces were analysed for concentrations of Campylobacter spp and faecal indicator bacteria; faecal coliforms, Escherichia coli and enterococci. The presence of other animals on the properties sampled as well as the age, feed and health of the horses at the time of sampling was recorded.

RESULTS: Enterococci and faecal coliforms were isolated from all samples, and E. coli was isolated from 58/59 samples. Mean concentrations of faecal coliforms and E. coli did not differ between properties, but there was a significant difference in mean concentration of enterococci between properties. Campylobacter spp. were detected in two faecal samples with one isolate being determined by PCR analysis to be a thermotolerant Campylobacter species, the other C. jejuni.

CONCLUSIONS: This is the first known report quantifying the concentration of Campylobacter spp. present in healthy adult horses in New Zealand. The presence of equine faecal material in water could elevate concentrations of faecal bacteria and therefore needs to be considered as a source of water contamination. The access of horses to waterways and coastal environments may also need to be restricted to prevent transmission of faecal indicator bacteria and potentially zoonotic agents.  相似文献

Fibreglass pools with sediment were used as model farming ponds to investigate the interactive effects of pond preparation and feeding rate on prawn production, water quality, bacterial dynamics, abundance of benthos and prawn feeding behaviour. Pools were either fertilised 1 month (prepared) or 2 days (unprepared) prior to stocking and either ‘high’ or ‘low’ feeding rates were used. The ‘high’ rate was 5.0% (range 4–8%) wet prawn biomass/day and was similar to that recommended for commercial farms. The ‘low’ rate was 2.5% (range 2–4%) wet prawn biomass/day. Juvenile Penaeus monodon (2.0–7.5 g) were stocked at 15 prawns/m2 and were cultured for 71 days. With the exception of one prepared, high feeding-rate pool where mass mortality (> 80%) of prawns occurred following an interruption to aeration, prawn survival was high (> 86%) and was unaffected by preparation, feeding rate or their interaction. Pond preparation improved growth and biomass gain by about 20%. Growth was 4% higher with the higher feeding rate but biomass gain was not affected and, as food conversion ratio was much worse, use of the lower feeding rate offers considerable scope to reduce production costs, especially during cooler periods. There was no interaction in relation to growth between pond preparation and feeding rate. Meiofauna were more abundant, and prawns grew faster, in prepared pools than unprepared pools at the start of the experiment. However, changes in bacterial dynamics or meiofauna abundance over time did not explain reductions in prawn growth over time. In general, water quality was reduced in pools receiving the high feeding rate compared with low feeding rate pools. Other interactive effects of pond preparation and feeding rate on water quality, bacteria, benthos and prawn feeding behaviour are discussed.  相似文献
