Experiments were made on the nature of fungal competition on sal leaf-litter with emphasis on the antibiotic action of culture filtrates. The culture filtrates of Mortierella subtilissima, Aspergillus candidus, A. flavus, A. niger, Penicillium rubrum, Papulaspora sp., Staphylococcus aureus and Bacillus subtilis were found most effective in suppressing the growth of various leaf-litter fungi. Dominant fungi which displayed best competitive tolerance to the staling products of culture filtrates were Aspergillus flavus, A. niger, A. sclerotiorum, A. terreus and Trichoderma harzianum.  相似文献

The effects of cryoprotectants and their concentrations, prefreezing temperature, equilibration time, thawing temperature, and the regenerative efficacy of cryopreserved vegetative thalli (apical tips) of Gracilaria corticata J. Agardh., Ulva lobata Duetzing, and Hypnea musiformis (Wulfar) Lamouroux were evaluated. The apical tips were suspended in various cryoprotective solutions and slowly cooled to ?40?C over a period of 4 h. After the slow cooling step, the suspensions were immediately immersed in liquid nitrogen (LN), which were thawed after 2 days. Both U. lobata and H. musiformis survived maximally in 10% DMSO, whereas 10% glycerol was found most suitable for G. corticata. A slow cooling temperature of ?40?C and thawing temperature of 40?C were found most suitable irrespective of the species. The equilibration time of 60 min was found most suitable for G. corticata, while 45 min was sufficient for both H. musiformi and U. lobata. Cryopreserved vegetative thalli were then successfully regenerated, though the survival was considerably reduced up to a period of 28 days after which it was stabilized.  相似文献

ABSTRACT

Soil and plant samples were collected from on-going long-term experiment at Indian Agricultural Research Institute, New Delhi farm to study the distribution of various forms of copper (Cu) and their contribution to availability and plant uptake in maize (Zea mays L.)—wheat (Triticum aestivum L.) sequence. The optimum dose-based treatments selected for the study were nitrogen-phosphorus-potassium (NPK), NPK + Farmyard manure (FYM), NPK+ zinc (Zn) and control (no fertilizer or manure). Uptake of Cu by maize and wheat varied from 17.0 to 37.5 and 60.8 to 149.3 g ha?1, respectively, under different treatments. Copper uptake by wheat was significantly higher under 100% NPK + FYM than that with 100% NPK. There was no significant difference among the treatments with respect to diethylenetriaminepentaacetic acid (DTPA)-extractable Cu in 0–15, 15–30, 30–45, and 45–60 cm soil layers. However, with increasing depth of soil, it showed declining trend under all the treatments. Mean value of total Cu was 28, 32, 25, and 21 mg kg?1 in 0–15, 15–30, 30–45, and 45–60 cm depths, respectively. Major part of the total Cu was present as residual form. Sorbed copper (SORB–Cu) contributed directly towards its availability both in pre-sowing maize and post-harvest wheat soil samples. SORB–Cu and organic matter bound Cu (OM–Cu) contributed directly towards the uptake by the component crops. Copper associated with easily reducible manganese, carbonate, and iron and aluminum oxides were most recalcitrant forms present in soil and their effects on availability and crop uptake were adverse.  相似文献

Sarcotriol (ST) has been shown to be chemopreventive on 7,12-dimethyl-benz(a)anthracene (DMBA) initiated and 12-O-tetradecanoylphorbol-13-acetate (TPA)-promoted skin tumor development in CD-1 mice in recent studies from our laboratory. The objective of this study was to determine the chemopreventive effects of ST on ultraviolet B (UVB)-induced skin tumor development in female SKH-1 hairless mice, an experimental model relevant to human skin cancer development, and its possible mechanisms of action. Female SKH-1 mice were divided into two groups: Control and ST treated. Control was topically treated with 100 μL acetone and ST treated group administered with 30 μg ST in 100 μL acetone one hour before UVB exposure. For UVB-induced tumorigenesis, carcinogenesis was initiated and promoted by UVB (180 mJ/cm2). Group weights and tumor counts were taken once every week. After 30 weeks, mice were sacrificed and dorsal skin samples were collected. The proteins from the skin sample were further used for SDS-PAGE and Western blotting using specific antibodies against caspase-3, caspase-8, caspase-9 and p53. Tumor multiplicity was found 19.6, 5.2 in the control and ST treated groups respectively. Caspase-3, -8, -9 and p53 were significantly (P < 0.05) upregulated in ST treated group compared to Control group. Together, this study for the first time identifies the chemopreventive effects of ST in UVB-induced carcinogenesis possibly by inducing apoptosis and upregulating p53.  相似文献

Arsenic (As) contamination of the environment has emerged as a concerning issue recently for which phytoremediation has been suggested as a viable solution. Hydrilla verticillata (L.f.) Royle is a widely distributed rapidly growing aquatic weed possessing significant potential to accumulate As and is thus a potential candidate for the purpose of As phytoremediation. In the present study, an investigation of thiol metabolism was conducted in H. verticillata, which revealed differential effects upon exposure to arsenite [As(III)] and arsenate [As(V)]. The accumulation of arsenic was found to be higher upon exposure to As(III) than to As(V). Besides, As(III) was found to induce the activities of enzymes, such as cysteine synthase and γ-glutamylcysteine synthetase and the amounts of cysteine and glutathione (GSH) to higher levels than that observed with As(V). The activity of glutathione-S-transferase was, however, stimulated to a higher level upon exposure to As(V) than As(III). The activity of arsenate reductase was found to increase upon As(V) exposure at all concentrations and durations. In addition, a significant stimulation in the activity of phytochelatin synthase was noticed in vitro with an increase in As/GSH concentration and time of incubation. Arsenic detoxification in H. verticillata thus appeared to involve an induction of thiol synthesis and consumption in a coordinated manner, though differentially upon exposure to As(III) and As(V). The information gained through this study would help in better designing of the pilot experiment at the field level depending on the chemical composition of the contaminated water.  相似文献

A stratified random sampling approach was employed to quantify total biomass across prevalent non-commercial forest understory species found in six counties of northwest Florida, USA. The moisture content (wet basis) and calorific values of these species were also measured. Total green biomass from forest understory species was estimated to be around 12 million metric tons, mostly comprised of Cliftonia monophylla (titi, buckwheat tree) and Cyrilla racemiflora (white titi, swamp titi). This understory forest biomass would be sufficient to generate about 28.8 million GJ of electricity or 1589.25 million liters of ethanol. A need was identified to determine the inventory of forest understory biomass at the state level and assess the overall sustainability of utilizing forest understory biomass for bioenergy.  相似文献

Genetic relationships were examined among thirty germplasm accessions belonging to five Ocimum species using RAPD markers. A very high degree of polymorphism (98.20%) was observed. UPGMA cluster analysis of genetic similarity indices grouped all the accessions into two major clusters corresponding to previously reported botanical sections. Intra-clustering within the two clusters precisely grouped the accessions belonging to one species in one sub-cluster as expected from their genetic background. Our results show that RAPD technique is a sensitive, precise and efficient tool for genomic analysis in Ocimum species, that may be useful in future studies, by assigning new unclassified germplasm accessions to specific taxonomic groups and reclassifying previously classified accessions of other Ocimum species by traditional criteria on a more objective basis. Grain legumes contribute significantly to total world food production. Legumes are the primary source of dietary proteins in many developing countries, where protein hunger and malnutrition are widespread. Grain legumes germplasm constitute ~15% of the 7.4 M accessions preserved globally. Nearly, 78% of the CGIAR??s, 0.217 M accessions, have been characterized, compared to 34% of national genebank collections. Interestingly, limited data on grain quality are available as the primary focus has been on morpho-agronomic traits. Clearly, more resources should be targeted on biochemical evaluation to identify nutritionally rich and genetically diverse germplasm. The formation of core and mini core collections has provided crop breeders with a systematic yet manageable entry point into global germplasm resources. These subsets have been reported for most legumes and have proved useful in identifying new sources of variation. They may however not eliminate the need to evaluate entire collections, particularly for very rare traits. Molecular characterization and association mapping will further aid to insights into the structure of legume diversity and facilitate greater use of collections. The use of high resolution elevational climate models has greatly improved our capacity to characterize plant habitats and species?? adaptive responses to stresses. Evidence suggests that there has been increased use of wild relatives as well as new resources resulting from mutagenesis to enhance the genetic base of legume cultigens.  相似文献
