When chickens were vaccinated with a recombinant fowlpox virus (FPV) containing the Newcastle disease virus (NDV) hemagglutinin-neuraminidase (HN) cDNA under the control of the thymidine kinase (TK) promoter and inserted into the FPV TK gene, the FPV antibody response to the recombinant virus was similar to the response to vaccination with standard FPV, and the recombinant virus protected chickens against challenge with virulent FPV. While the presence of the NDV HN cDNA was demonstrated in the recombinant virus, which was stable on serial passage, expression of HN was not detected by hemagglutination, Western blot analysis or immunoprecipitation of infected cell lysate. Chickens vaccinated with the recombinant virus failed to mount an NDV hemagglutination-inhibition antibody response, and they did not resist challenge with velogenic NDV. It was concluded that the TK promoter was too weak to drive the HN gene, but that the insertion into the FPV TK gene did not reduce the immunogenicity of the virus.  相似文献

The diagnostic value of intravenously administered johnin purified protein derivative (PPD) was studied in 45 cattle of different age, coming from herds infected by, or free from, Mycobacterium paratuberculosis. In addition to observing the clinical symptoms, the animals' sera were assayed for specific antibodies by the complement fixation (CFT) and immunodiffusion (AGID) tests. The blastogenic transformation of peripheral lymphocytes was determined on the basis of 3HTdR incorporation. Changes in the neutrophilic leucocyte/lymphocyte ratio of the blood were also monitored. Detection of the pathogen in the faeces was attempted by microscopic examination and by culturing. Combined evaluation of responses elicited by intravenously administered johnin PPD can be a valuable aid in recognizing infected animals, particularly those among the heifer progeny of infected cows.  相似文献

The aim of the present study was to clarify the effects of hypothalamic dopamine (DA) on salsolinol (SAL)‐induced prolactin (PRL) release in goats. The PRL‐releasing response to an intravenous (i.v.) injection of SAL was examined after treatment with augmentation of central DA using carbidopa (carbi) and L‐dopa in male goats under 8‐h (8 h light, 16 h dark) or 16‐h (16 h light, 8 h dark) photoperiod conditions. The carbi and L‐dopa treatments reduced basal PRL concentrations in the 16‐h photoperiod group (P < 0.05), while a reduction was not observed in the 8‐h photoperiod group. The mean basal plasma PRL concentration in the control group for the 8‐h photoperiod was lower than that for the 16‐h photoperiod (P < 0.05). SAL significantly stimulated the release of PRL promptly after the injection in both the 8‐ and 16‐h photoperiod groups (P < 0.05). PRL‐releasing responses for the 16‐h photoperiod were greater than those for the 8‐h photoperiod (P < 0.05). The carbi and L‐dopa treatments blunted SAL‐induced PRL release in both the 8‐ and 16‐h photoperiods (P < 0.05). These results indicate that hypothalamic DA blunts the SAL‐induced release of PRL in male goats, regardless of the photoperiod, which suggests that both SAL and DA are involved in regulating the secretion of PRL in goats.  相似文献

Titanium is the tenth most common element in the Earth's crust. In soils we find it in form of TiO2, some titanates and Ti-silicates. Therefore, in most cases Ti is not taken up by plants and therefore, it is not a member of the food chain. Water-soluble and pH-stable chelate form, Ti-ascorbate was applied in plant and animal nutrition. It was demonstrated on a very large scale that Ti-ascorbate has a very avorable, beneficial role in various physiological processes. The results show that Ti should be a member of the family of demonstrated beneficial elements.  相似文献

OBJECTIVE: To determine whether strength of serologic recognition of bovine leukosis virus (BLV) by use of ELISA is associated with blood lymphocyte counts. DESIGN: Prospective study. ANIMALS: 161 cows with positive results of ELISA for BLV. PROCEDURE: Sample-to-positive ratio (S:P), which is the ratio between the test sample and a positive control sample, was compared among lymphocytotic and nonlymphocytotic cows. A regression model was constructed to evaluate the association between blood lymphocyte concentration and S:P, age, and the interaction of these terms. RESULTS: Mean S:P differed significantly between lymphocytotic (2.58 +/- 0.36) and nonlymphocytotic (2.38 +/- 0.39) cows. Age and S:P were significantly associated with lymphocyte count. CONCLUSIONS AND CLINICAL RELEVANCE: Sample-to-positive ratio and lymphocyte count were related; however, cows with high S:P were not always lymphocytotic. Culling cows on the basis of S:P will reduce the herd load of infectious virus faster than random culling of ELISA-positive cows; however, culling on the basis of lymphocyte count will eliminate a greater proportion of the reservoir of infection.  相似文献

To optimize their growth and survival, plants perceive and respond to ultraviolet-B (UV-B) radiation. However, neither the molecular identity of the UV-B photoreceptor nor the photoperception mechanism is known. Here we show that dimers of the UVR8 protein perceive UV-B, probably by a tryptophan-based mechanism. Absorption of UV-B induces instant monomerization of the photoreceptor and interaction with COP1, the central regulator of light signaling. Thereby this signaling cascade controlled by UVR8 mediates UV-B photomorphogenic responses securing plant acclimation and thus promotes survival in sunlight.  相似文献

Sera of pigs immunized with parts of the transmissible gastroenteritis virus (TGEV) spike (S) protein expressed by recombinant baculoviruses were tested, together with a TGEV hyperimmune antiserum, for their abilities to protect three-day-old piglets against TGEV infection. The piglets were infected with virulent TGEV and the sera were given orally 3 h before infection, together with the virus, and every 6 h postinfection during the 30 h of the experiment. Virus shedding was monitored by TGEV isolation from rectal swab samples. The sera containing antibodies induced by the complete S protein or the amino terminal half of the S protein showed protective properties, indicated by delayed onset of clinical signs and virus shedding, similar to the TGEV hyperimmune serum. Those immune sera containing antibodies induced by shorter recombinant proteins were not protective.  相似文献

ABSTRACT Producers of champignon (Agaricus bisporus) and oyster mushroom (Pleurotus ostreatus) are facing recent incidents of green mold epidemics in Hungary. We examined 66 Trichoderma strains isolated from Agaricus compost and Pleurotus substrate samples from three Hungarian mushroom producing companies by a polymerase chain reaction-based diagnostic test for T. aggressivum, sequence analysis of the internal transcribed spacer region 1 (ITS1) and ITS2 and (selectively) of the fourth and fifth intron of translation elongation factor 1alpha (tef1alpha), and restriction fragment length polymorphism of mitochondrial DNA. Seven Trichoderma species were identified: T. aggressivum f. europaeum (17 isolates), T. harzianum (three isolates), T. longibrachiatum (four isolates), T. ghanense (one isolate), T. asperellum (four isolates), T. atroviride (nine isolates), and a still undescribed phylogenetic species, Trichoderma sp. DAOM 175924 (28 isolates). T. aggressivum f. europaeum was exclusively derived from A. bisporus compost, whereas Trichoderma sp. DAOM 175924 exclusively occurred in the substrate for Pleurotus cultivation. Sequences of the latter strains were co-specific with those for Trichoderma pathogens of P. ostreatus in Korea. The widespread occurrence of this new species raises questions as to why infections by it have just only recently been observed. Our data document that (i) green mold disease by T. aggressivum f. europaeum has geographically expanded to Central Europe; (ii) the green mold disease of P. ostreatus in Hungary is due to the same Trichoderma species as in Korea and the worldwide distribution of the new species indicates the possibility of spreading epidemics; and (iii) on mushroom farms, the two species are specialized on their different substrates.  相似文献
