Purpose

Interestingly, soil is the component of the natural environment in which most hydrophobic organic pollution including polycyclic aromatic hydrocarbons (PAHs) gets accumulated. The aim of the present paper was to determine the effect of soil pollution with PAHs on the elemental composition, spectral properties, and hydrophobic and hydrophilic properties of humic acids. The research was performed on different types of soil samples that were artificially polluted with selected PAHs (anthracene, pyrene, fluorene and chrysene).

Materials and methods

The soil samples were polluted with selected PAHs in an amount corresponding to 10 mg PAHs kg?1. The PAHs-polluted soil samples were incubated for 180 and 360 days at a temperature of 20–25 °C and fixed moisture (50 % of field water capacity). Humic acids (HAs) were extracted from the soil samples prior to the incubation (additionally, soils not polluted with PAHs) and after 180 and 360 days of incubation. For isolated HAs, the following analyses were performed: elemental composition, UV–Vis and IR spectra, susceptibility to oxidation, and hydrophilic (HIL) and hydrophobic (HOB) properties were determined using high-performance liquid chromatography.

Results and discussion

The research demonstrated that introducing anthracene, fluorene, pyrene and chrysene to soil samples resulted in a change in some of the quality parameters of humic acids. However, the intensity and the direction of those changes were determined by soil properties. The changes of the parameters, once PAHs were introduced, that did not depend on the soil properties were ΔA 665u and ΔA 465u (susceptibility to oxidation at wavelengths of 465 and 665 nm) as well as HIL/ΣHOB. The same tendency in changes in the structure of humic acids, once PAHs were introduced, was also observed based on the Fourier transform infrared spectra pattern.

Conclusions

A single pollution of soils with PAHs that leads to changes in the quality parameters of humic acids shows that, as for the soils permanently exposed to pollution with those compounds, significant changes can occur in the properties of humic acids. As a result, it can lead to a change in the functions played by humic acids in the environment.  相似文献

Mercury is a neurotoxin, its main source in the human organism being fish and seafood. The first level in the marine food web is formed of planktonic and benthic photosynthetic microorganisms, which form a biofilm on the surface of the hard bottom (epilithon) or plants (epiphyton). They are carriers of nutritional as well as toxic substances and pass these on to subsequent levels of the trophic web. Their biomass is often dominated by diatoms. This was the basis for the presented study into Hg accumulation in epilithic, epiphytic and planktonic diatoms, which was carried out in 2012–2013 in the coastal zone of the Puck Lagoon and the Gulf of Gdańsk (southern Baltic). In this coastal area, both micro- and macroorganisms develop particularly intensively. The collected results indicate an increase in Hg concentration in the biofilm during the warm season which, with the lengthening of the vegetative period due to global warming in recent years, is of great significance. As a consequence, the annual mercury load entering the trophic web is larger in comparison with a year in which there is a long, cold winter. An important parameter influencing the accumulation of Hg was the function of those organisms from the biofilm-forming communities. In this case, the highest concentrations of Hg were measured in organisms forming high-profile guilds.  相似文献

The 14-3-3 protein is one of the best candidates for coordinating all plant metabolic pathways. To verify this suggestion transgenic potato plants with repression of one (J4 and J5 plants), two (G1 plants), and six (G3 plants) constitutive 14-3-3 protein isoforms as well as plants overexpressing the 14-3-3 protein were studied. Reduction in the 14-3-3 protein level in the J4 and J5 transformants, the G1 transformants, and the G3 transformants was close to 29, 41.5, 38, and 55%, respectively. In the case of the 14-3-3 overexpressing plants (J2), a 30% increase in protein content was detected. Changes in nitrate reductase (NR), sucrose phosphate synthase (SPS), and starch synthase (SS) activities in the transgenic plants perfectly reflect the overall 14-3-3 protein level. The highest increase in enzyme activities was observed for the G3 plants and the lowest for the J4 transformants. The same was detected for the measured metabolites. The highest increase in the protein, starch, and sucrose levels was detected in the tubers from the G3 transgenic plants. Because there was almost no change in the isoform ratio in the transgenic plants when compared to the control, it is suggested that it is the overall content of the 14-3-3 protein, rather than the content of particular isoforms, which plays a crucial role in the regulation of enzyme activities and thus in metabolite synthesis. The properties of the 14-3-3 overexpressing plants are very similar to those of the control ones, suggesting that the protein is in excess in the nontransformants and a further increase in its content is not recognized by cell metabolism. A considerable influence of the 14-3-3 protein level on potato plant metabolism was demonstrated. This effect was observed in key metabolic enzyme activities and metabolite content as well. A high variability between mean values, representing individual transgenes, with respect to nitrate reductase, sucrose phosphate synthase, and starch synthase activities in the examined genotypes was noted. These changes were closely correlated with metabolite levels, among them protein, starch, reducing sugars, and sucrose. The results obtained for the five types of transgenic potato plants in comparison with the control were statistically assessed using discriminate function and cluster analyses.  相似文献

Conservation biology often relies on the protection of (semi)natural habitat remnants. However, the ever increasing human population is taking over natural resources and habitats. Here, contrary to most other studies, we ask how human-associated severe changes in the environment can be used to enrich local biodiversity. We tested if industrial activity (gravel excavation) leads to the creation of habitats that support grassland butterflies and how these areas add to the richness of local species when compared to typical semi-natural habitats (grasslands). We also identified key factors affecting the richness, abundance, diversity and commonness of butterfly species to provide practical recommendations. Species richness, diversity index and the occurrence of rare species were higher in gravel-pit shores than in grasslands. The richness of butterfly species and their abundance were positively affected by the richness of plant species, shrub density and age of the gravel-pit but negatively by the cover of water reservoirs in the surrounding area and the isolation of gravel-pits from grasslands. Butterfly diversity was positively influenced by the richness of plant species and proximity of human settlement but negatively by area of the shore and isolation. Our study is the first one to show the high value of gravel-pits for the conservation of butterflies. We recommend the inclusion of gravel-pits in a system of ecological networks and management of their surroundings to improve the colonization rate of rare species. We suggests that directing interest to the possible positive effects of industrial development on biodiversity may support conservation efforts.  相似文献

Leaching of nitrogen from arable land can lead to pollution of groundwater and surface water. Various measures have been implemented in agriculture to reduce leaching, but there is still potential to do more. To find the best agricultural management regime to limit the problem, leaching from soils under different management must be measured. Different methods to estimate leaching are available, but they have not been thoroughly evaluated. This study compared (a) leaching of nitrogen from five different fertilizer treatments differing in amount and source (mineral and organic) of nitrogen and (b) two different methods for measuring leaching (tile-drained field plots and lysimeters). Nitrogen leaching from five different fertilizer treatments was studied for three years in a tile-drained field facility and for three of the treatments also for 16 months in a lysimeter facility. Leaching from organic and mineral nitrogen sources was similar in the three-year field study. Mineral nitrogen input above the economic optimum tended to give greater leaching (by 24%–43%) than nitrogen input at the expected optimum. Measurements in lysimeters and tile-drained field plots gave similar results, although leaching values tended to be slightly higher (by 18%–25%) in the tile-drained field. Overall, both facilities give reliable estimates of nitrogen leaching and are suitable for leaching studies.  相似文献

The objective of the study was to determine the disinfection efficacy of aerosolizing (cold fogging) Virkon S on survival of Stahpylococcus aureus and Salmonella enterica on different surfaces. Two experiments were conducted in different locations. Salmonella enterica and S. aureus were grown in broth culture and then seeded into pre-marked areas in each location and allowed to dry. Virkon S (1%) was aerosolized into the rooms (approximately 1L of per 30 m(3)). Samples were collected pre- and post-fogging for quantitative cultures to evaluate the efficacy of aerial disinfection. The reduction of S. enterica or S. aureus counts ranged from 3.40 to 0.95 log(10) (Salmonella) or 4.92 to 0.02 log(10) (Staphylococcus). The greatest reduction was evident in samples collected from non-porous horizontal surfaces, which were not obstructed from the air flow. These results indicate that fogging with Virkon S could be beneficial in routine disinfection of pre-cleaned surfaces. The benefits of routine use of cold fogging with Virkon S in veterinary hospital settings would include its wide-range antimicrobial action and minimal working-men power required to disinfect large areas. Also, fogging would potentially minimize microbial contamination in the hard to reach areas.  相似文献

Herding semi-domesticated reindeer has economic and social value for Sami people in the northern territories of Fennoscandia. However, with the intensification of reindeer husbandry, interspecies transmission of pathogens between reindeer and domestic animals may become a problem, especially for countries such as Sweden, Norway, and Finland where pestivirus and alphaherpesvirus have been eradicated in domestic ruminants. This study, which included 1158 Swedish reindeer, showed relatively high prevalence of antibodies against bovine viral diarrhoea virus (BVDV) (32%) and bovine herpesvirus-1 (BoHV-1) (53%). Adult animals were more often seropositive for BVDV and BoHV-1 (50% and 78%, respectively) than were calves (18 and 11%, respectively). While the seroprevalence of alphaherpesvirus was similar in different herding districts, pestivirus seropositivity was highest in the South and diminished towards the North of the Swedish reindeer herding area. High correlation of the seropositivity against both pathogens at both individual and herd levels may indicate possible mutual synergetic effects and may be explained by the immunosuppressive nature of the viruses. While alphaherpesvirus seroprevalence was probably related to putative cervid herpesvirus 2 (CvHV-2), the pestivirus infecting reindeer remains undefined. The virus neutralisation test of reindeer sera using different pestivirus strains, revealed higher titres against Border disease virus strains like 137/4 (BDV-1) and Reindeer-1 (BDV-2) than against BVDV-1. However, the virus was not identified by real time RT-PCR in any of the samples (n=276) from seronegative reindeers. The study showed that pestivirus and alphaherpesvirus infections are endemic in the Swedish reindeer population.  相似文献

Pseudodidymella fagi, described from infected leaves of Fagus crenata in Japan, has recently been spreading in Europe, where it causes leaf blotch of European beech Fagus sylvatica. In the years 2008–2016, an outbreak of P. fagi was observed on European beech in Switzerland, Germany and Austria, and it is reported here for the first time from Slovakia where it was detected in 2017. The identification of the pathogen is supported by the morphological analyses of material observed in vivo and in vitro and molecular analyses based on ITS sequences. Detailed morphological characterization of cultures based on European material, on F. sylvatica, is provided here for the first time.  相似文献

(1) Background: The mussel (Mytilus edulis, Mytilus galloprovincialis) is the most widespread lamellibranch mollusk, being fished on all coasts of the European seas. Mussels are also widely grown in Japan, China, and Spain, especially for food purposes. This paper shows an original technique for mussel shell processing for preparation of calcium salts, such as calcium levulinate. This process involves synthesis of calcium levulinate by treatment of Mytilus galloprovincialis shells with levulinic acid. The advantage of mussel shell utilization results in more straightforward qualitative composition. Thus, the weight of the mineral component lies with calcium carbonate, which can be used for extraction of pharmaceutical preparations. (2) Methods: Shell powder was first deproteinized by calcination, then the mineral part was treated with levulinic acid. The problem of shells generally resulting from the industrialization of marine molluscs creates enough shortcomings, if one only mentions storage and handling. One of the solutions proposed by us is the capitalization of calcium from shells in the pharmaceutical industry. (3) Results: The toxicity of calcium levulinate synthesized from the mussel shells was evaluated by the method known in the scientific literature as the Constantinescu phytobiological method (using wheat kernels, Triticum vulgare Mill). Acute toxicity of calcium levulinate was evaluated; the experiments showed the low toxicity of calcium levulinate. (4) Conclusion: The experimental results highlighted calcium as the predominant element in the composition of mussel shells, which strengthens the argument of capitalizing the shells as an important natural source of calcium.  相似文献

Fat type in diet is responsible for specific changes in gut microbiota (GM). Extra virgin olive oil (EVOO) has been shown to be beneficial for blood pressure and to produce effects on GM. To analyze the cause-effect relationship between intestinal microbial changes and blood pressure, we studied the effect of EVOO on fecal microbiota and systolic blood pressure (SBP) levels in spontaneously hypertensive rats (SHR). SHR were fed either an enriched EVOO diet or a standard diet for a period of 12 weeks. At the end of the experimental period, the microbial profiles in the feces were studied in both groups by using PCR-denaturing gradient gel electrophoresis. Real-time PCR was used to quantify the selected bacterial groups. The results demonstrated significant differences when using Lactobacillus (p<0.05), clostridia XIV (p<0.01) and universal (p<0.05) primers. A significant (r=?0.475; p=0.04) inverse correlation between the abundance of clostridia XIV and SBP, which depends on the type of diet, was also observed. Finally, the results suggested an increase in the microbial diversity of the feces of the animals fed the EVOO diet. These results strongly connect the pattern of GM in SHR fed a diet enriched with EVOO to the lower levels of SBP observed in these animals at the end of the feeding period.  相似文献
