This study aimed at examining the histological structure of the pecten oculi in the adult yellow‐legged gull, Larus michahellis, and at two moments of postnatal development: during the posthatch (nestling) and juvenile periods. Particular attention was paid to differences in the diameter of vessels, the thickness of the basement membrane, and ultrastructural features of endothelial and pigmented stromal cells. Capillary endothelial cells displayed numerous microvillous‐like folds projecting from their internal and external surfaces. Intercellular spaces between capillaries were occupied by pigmented stromal cells. The ultrastructure of pecten oculi underwent noticeable changes during postnatal development. The examination of the capillaries in nestlings, juveniles, and adults revealed that the formation process of vessels and pigmented stromal cells did not complete itself in the posthaching phase. The prominent feature of endothelial cells of capillaries in nestlings was that the microvilli were longer than in juvenile and adult cells, and the capillary lumen was therefore reduced. In this sense, their pigmented stromal cells showed fewer melanosomes, lacked intercellular spaces, and cellular junctions could still be observed. These results provide evidence that the pecten oculi during the posthatching phase maintains immature morphological features consistent with a role of pigmented stromal cells in the blood‐retina barrier.  相似文献

Forty percentage of UK peatlands have been drained for agricultural use, which has caused serious peat wastage and associated greenhouse gas emissions (carbon dioxide (CO2) and methane (CH4)). In this study, we evaluated potential trade-offs between water-table management practices for minimizing peat wastage and greenhouse gas emissions, while seeking to sustain romaine lettuce production: one of the most economically relevant crop in the East Anglian Fenlands. In a controlled environment experiment, we measured lettuce yield, CO2, CH4 fluxes and dissolved organic carbon (DOC) released from an agricultural fen soil at two temperatures (ambient and +2°C) and three water-table levels (−30 cm, −40 cm and −50 cm below the surface). We showed that increasing the water table from the currently used field level of −50 cm to −40 cm and −30 cm reduced CO2 emissions, did not affect CH4 fluxes, but significantly reduced yield and increased DOC leaching. Warming of 2°C increased both lettuce yield (fresh leaf biomass) and peat decomposition through the loss of carbon as CO2 and DOC. However, there was no difference in the dry leaf biomass between the intermediate (−40 cm) and the low (−50 cm) water table, suggesting that romaine lettuce grown at this higher water level should have similar energetic value as the crop cultivated at −50 cm, representing a possible compromise to decrease peat oxidation and maintain agricultural production.  相似文献

ABSTRACT: Genes localized at Salmonella pathogenicity island-1 (SPI-1) are involved in Salmonella enterica invasion of host non-professional phagocytes. Interestingly, in macrophages, SPI-1-encoded proteins, in addition to invasion, induce cell death via activation of caspase-1 which also cleaves proIL-1β and proIL-18, precursors of 2 proinflammatory cytokines. In this study we were therefore interested in whether SPI-1-encoded type III secretion system (T3SS) may influence proinflammatory response of macrophages. To test this hypothesis, we infected primary porcine alveolar macrophages with wild-type S. Typhimurium and S. Enteritidis and their isogenic SPI-1 deletion mutants. ΔSPI1 mutants of both serovars invaded approx. 5 times less efficiently than the wild-type strains and despite this, macrophages responded to the infection with ΔSPI1 mutants by increased expression of proinflammatory cytokines IL-1β, IL-8, TNFα, IL-23α and GM-CSF. Identical macrophage responses to that induced by the ΔSPI1 mutants were also observed to the infection with sipB but not the sipA mutant. The hilA mutant exhibited an intermediate phenotype between the ΔSPI1 mutant and the wild-type S. Enteritidis. Our results showed that the SPI-1-encoded T3SS is required not only for cell invasion but in macrophages also for the suppression of early proinflammatory cytokine expression.  相似文献

The significance of infection with Equid herpesvirus 2 (EHV-2) remains unresolved, mainly due to its widespread distribution, and frequent isolation of the virus not only from diseased animals, but also from clinically normal horses. It has been suggested that EHV-2 exerts its effects on the host indirectly, through predisposition to secondary infections. The aim of this study was to determine kinetics of EHV-2 infection among foals and to investigate the role that EHV-2 may play in development of Rhodococcus equi pneumonia on one farm. Serial blood samples were collected from 43 foals over a period of 10 months. Viral load of EHV-2 in blood was determined using quantitative realtime PCR assay. All but 2 foals were positive for EHV-2 on at least one occasion. The majority (88%) of foals became infected with EHV-2 within the first 2 months of life. Once infected, most foals (86%) remained positive for EHV-2 on all subsequent samplings. The load of EHV-2 varied between individual foals and between different sampling times for each foal. There was a significant difference in EHV-2 load between samples collected from healthy foals and foals suspected of R. equi pneumonia only for 2-month-old foals, but not for foals of other ages. The results of this study extend our knowledge of EHV-2 epidemiology among foals.  相似文献

Each living cell of a plant produces photons in certain conditions. Under normal physiological conditions, cell photon emission is stationary and minimal. Disturbance in the oxidative homeostasis by biotic stress is manifested by increased ‘biophoton’ production. Such biophoton responses of plants may be used as an integral indicator of the degree of oxidative homeostasis misbalance. Our results demonstrate that biophoton generation has been much higher in a resistant potato variety than in a susceptible one till 10 h after Phytophthora infestans inoculation. In contrast, ultra-weak luminescence from detached susceptible potato and moderately resistant pelargonium leaves increased from 1–4 to 4–5 days after inoculation with Phytophthora infestans or Botrytis cinerea, respectively. Pre-treatment of susceptible potato leaves with a defence inducer, arachidonic acid, resulted in a transient burst of light in response to P. infestans lasting for 30–45 h post inoculation (hpi). This study presents the potential adaptation of functional imaging of ultra-weak luminescence to monitor time-dependent free radical processes during disease development and its application to draw conclusions on plant resistance to pathogens of different lifestyle. Moreover, it has been shown that imaging of temporal biophoton generation from potato leaves treated with arachidonic acid might be a helpful marker in mapping oxidative changes leading to systemic acquired resistance (SAR).  相似文献

Black root rot (BRR) caused by Thielaviopsis basicola as well as Tomato spotted wilt virus (TSWV) are the most serious problems in tobacco growing regions. We crossed the breeding line WGL 3 carrying BRR resistance derived from N.glauca with the line PW-834 the resistance of which to TSWV was transferred from cultivar Polalta. Anthers obtained from F1 hybrid plants were cultured to induce haploids combining resistance to Th. basicola and TSWV. Flow cytometry analysis revealed 242 haploids and 2 spontaneous doubled haploids among regenerants. All haploids were cloned and then evaluated for BRR as well as TSWV resistance. The presence of pathogens was detected by microscopic evaluation of roots or using DAS-ELISA test. Microscopic assessment showed that, 132 haploids had no symptoms of Th. basicola which, together with the absence of symptoms in the F1 hybrids, indicated a dominant monogenic mode of inheritance. At the same time only 30 haploids demonstrated resistance to TSWV. SCAR markers associated with TSWV resistance gene detection was applied. The results indicate that small proportion of TSWV-resistant haploids is probably due to the influence of deleterious genes flanking the resistance factor that reduced vitality of gametophytes. Altogether, 24 haploids showed multiple resistance to Th. basicola and TSWV.  相似文献

After 1989, the use of potassium (K) mineral fertilizers in the Czech Republic dropped from ~55 kg K ha?1 to a mean rate of ~6.5 kg in the last decade. In order to test alternative solutions for K fertilization, the trioctahedral mica mineral zinnwaldite (8% K), orthoclase (10% K) and waste mica from Cínovec (Kru?né Hory Mts/Erzgebirge, Czech Republic), consisting primarily of zinnwaldite, were applied as the only K sources for spring barley. The minerals were treated in three different types of high-energy mills under different working conditions. Application rates in the range 139–820 mg K kg?1 were tested in quartz sand cultures. In all treatments, plant growth, total plant biomass and the K content in the plant tissues increased in the order zinnwaldite > waste mica > orthoclase. K fractionation and K plant uptake were significantly influenced by the milling method used. The effect of 195 mg K kg?1 as zinnwaldite on K uptake from K-depleted soils was positive; however, it was smaller than in sand cultures because of the relatively high content of non-exchangeable K in the soils. Direct use of waste mica as a K fertilizer is limited by the increased fluorine and heavy metal content.  相似文献

This study identified the indigenous criteria used by livestock farmers in urban and peri-urban areas of Kampala to assess the nutritional quality of available feed resources. Focus group discussions and questionnaire interviews (with a total of 120 livestock farming households) were conducted. The findings showed that banana peels, leftover food and own-mixed feeds were the most commonly used feed resources for cattle, pigs and chickens, respectively. Farmers use several indigenous criteria to judge the nutritional quality of the available feed resources. These included perceived effects on disease resistance, feed intake, growth/body condition, hair coat appearance, faecal output, faecal texture and level of production, among others. According to farmers, animals offered with a feed resource of good nutritional quality are more resistant to diseases, ingest much of the feed, gain weight with well-filled bodies, have smooth hair coats, produce large quantities of faeces that are not too firm or watery and exhibit good performance (lactating cows produce more milk, sows produce piglets of good body size, hens lay more eggs of normal size, etc.). Although this indigenous knowledge exists, farmers put more importance on availability and cost as opposed to nutritional quality when choosing feed resources. This explains why banana peels were among the feed resources perceived to be of low nutritional quality but, at the same time, were found to be the most commonly used. Hence, there is a need to sensitise farmers on the importance of nutritional quality in ensuring better and efficient utilisation of the available feed resources.  相似文献
