Photoelectrons excited by extreme ultraviolet or x-ray photons in the presence of a strong laser field generally suffer a spread of their energies due to the absorption and emission of laser photons. We demonstrate that if the emitted electron wave packet is temporally confined to a small fraction of the oscillation period of the interacting light wave, its energy spectrum can be up- or downshifted by many times the laser photon energy without substantial broadening. The light wave can accelerate or decelerate the electron's drift velocity, i.e., steer the electron wave packet like a classical particle. This capability strictly relies on a sub-femtosecond duration of the ionizing x-ray pulse and on its timing to the phase of the light wave with a similar accuracy, offering a simple and potentially single-shot diagnostic tool for attosecond pump-probe spectroscopy.  相似文献

Single soft-x-ray pulses of approximately 90-electron volt (eV) photon energy are produced by high-order harmonic generation with 7-femtosecond (fs), 770-nanometer (1.6 eV) laser pulses and are characterized by photoionizing krypton in the presence of the driver laser pulse. By detecting photoelectrons ejected perpendicularly to the laser polarization, broadening of the photoelectron spectrum due to absorption and emission of laser photons is suppressed, permitting the observation of a laser-induced downshift of the energy spectrum with sub-laser-cycle resolution in a cross correlation measurement. We measure isolated x-ray pulses of 1.8 (+0.7/-1.2) fs in duration, which are shorter than the oscillation cycle of the driving laser light (2.6 fs). Our techniques for generation and measurement offer sub-femtosecond resolution over a wide range of x-ray wavelengths, paving the way to experimental attosecond science. Tracing atomic processes evolving faster than the exciting light field is within reach.  相似文献

This exogenic test for the detection of enzyme activities (revertase) is helpful in differentiating between leucocytes of leukaemic cattle, on the one hand, and those of intact animals, on the other. The mechanism of such identification is based on differences in enzyme activity in the presence of poly rA/oligo dT and poly dA/oligo dT. The best results were obtained from short-time cultured leucocytes which gave ten times higher margins of incorporation by comparison between intact and preleucotic cattle. The morphological findings obtained by electron microscopy confirmed the results by other authors and enlarged them by determination, for the first time, of RNA filaments and protein rings.  相似文献

Leaf rust of barley, caused by Puccinia hordei Otth, is an important foliar disease in most temperate regions of the world. Sixteen major leaf rust resistance (Rph) genes have been described from barley, but only a few have been mapped. The leaf rust resistance gene Rph7 was first described from the cultivar ‘Cebada Capa’ and has proven effective in Europe. Previously mapped restriction fragment length polymorphism (RFLP) markers have been used to determine the precise location of this gene in the barley genome. From the genetic analysis of a ‘Bow‐man’/‘Cebada Capa’ cross, Rph7 was mapped to the end of chromosome 3HS, 1.3 recombination units distal to the RFLP marker cMWG691. A codominant cleaved amplified polymorphic site (CAPS) marker was developed by exploiting allele‐specific sequence information of the cMWG691 site and adjacent fragments of genomic DNA. Based on the large amount of polymorphism present in this region, the CAPS marker may be useful for the marker‐assisted selection of Rph7 in most diverse genetic backgrounds.  相似文献

Background

Reversible antioxidant depletion is found in hyperthyroid humans, and antioxidant depletion increases the risk of methimazole toxicosis in rats.

Objectives

To determine whether abnormalities in concentrations of blood antioxidants or urinary isoprostanes were present in hyperthyroid cats, and were reversible after radioiodine treatment. To determine whether or not antioxidant abnormalities were associated with idiosyncratic methimazole toxicosis.

Animals

Hyperthyroid cats presented for radioiodine treatment (n = 44) and healthy mature adult control cats (n = 37).

Methods

Prospective, controlled, observational study. Red blood cell glutathione (GSH), plasma ascorbate (AA), plasma free retinol (vitamin A), α‐tocopherol (vitamin E), and urinary free 8‐isoprostanes in hyperthyroid cats were compared to healthy cats and to hyperthyroid cats 2 months after treatment.

Results

Blood antioxidants were not significantly different in hyperthyroid cats (mean GSH 1.6 ± 0.3 mM; AA 12.8 ± 4.9 μM, and vitamin E, 25 ± 14 μg/mL) compared to controls (GSH 1.4 ± 0.4 mM; AA 15.0 ± 6.6 μM, and vitamin E, 25 ± 17 μg/mL). Urinary isoprostanes were increased in hyperthyroid cats (292 ± 211 pg/mg creatinine) compared to controls (169 ± 82 pg/mg; = .006), particularly in hyperthyroid cats with a USG < 1.035. Plasma free vitamin A was higher in hyperthyroid cats (0.54 ± 0.28 μg/mL versus 0.38 ± 0.21 in controls; = .007). Both abnormalities normalized after radioiodine treatment. No association was found between oxidative status and prior idiosyncratic methimazole toxicosis.

Conclusion and Clinical Importance

Increased urinary isoprostane could reflect reversible renal oxidative stress induced by hyperthyroidism, and this requires additional evaluation.  相似文献

The revertase test with exogenous matrix, poly-(rA) . oligo-(dT), for the detection of bovine leukemia virus from crude virus sediments was standardised and miniaturised. An amount of 10 ml of cell culture supernatant of short-term cultured lymphocytes (5 . 10(6) cells/ml) is quite sufficient for testing one cattle sample. The lower sensitivity limit of the test was found to be 10(6) virus particles. The test is properly reproducible, within tolerance limits of +/- 20--30 per cent, provided that optimum lysis conditions be maintained (0.01 per cent triton X-100, 20 minutes, 0--4 degrees C incubation) and under the condition that the protein quantity in 100 microliter test solution does not exceed the threshold of 3--15 micrograms. The specificity of the test is based on the use of free viruses from cell culture supernatant, the optimum temperature of the revertase reaction at 25 degrees C, which actually deviates from that for cellular DNA polymerases, that is 37 degrees C, and magnesium ion concentration which has to be optimum for bovine leucosis virus revertase. Two-hundred heads of cattle, differing by haematological status, were examined, and 56 per cent of them were, clearly, virus producers, among them 95 per cent of all animals with positively established leucosis and 36 per cent of the haematologically intact animals. Examinations of individuals have shown that in repetitive checks, carried out in intervals between two months and one week, the revertase activities varied by something between 0.5 and two magnitudes.  相似文献

The age dependent development of the long bones' diaphyses (humerus, os femoris and os tibiofibulare) was examined macroscopically, morphometrically and histologically using 15 White New Zealand Rabbits (WN) and 15 Chinchilla-Bastard-Experimental-Rabbits (Chbb) in 5 different age groups. The main growth of bone occurs in the first three months of life. The remodelling from juvenile to adult bone began in the period from the 4th-12th week. The different bones show various characteristic remodelling zones for each localization. Until the 12th week the resorption of juvenile bundled bone predominates. Afterwards an increasing bone formation by osteal bone can be observed. The bone growth is terminated between the 17th and 33rd week. The os tibiofibulare in both breeds represents the major part of the compact bone area on the whole cross-section area. Principally the bone-remodelling process proceeds equally in both breeds. Breed dependent differences in the adult bone are seen in the construction of the circumferential lamellae. These are formed considerably more irregularly by the Chbb-rabbits than by the WN-rabbits.  相似文献
