The methanol extract of stems of Catalpa ovata G Don exhibits potent in vivo antifungal activity against Magnaporthe grisea (Hebert) Barr (rice blast) on rice plants, Botrytis cinerea Pers ex Fr (tomato grey mould) and Phytophthora infestans (Mont) de Bary (tomato late blight) on tomato plants, Puccinia recondita Rob ex Desm (wheat leaf rust) on wheat plants and Blumeria graminis (DC) Speer f. sp. hordei Marchal (barley powdery mildew) on barley plants. An antifungal substance was isolated and identified as dehydro-alpha-lapachone from mass and nuclear magnetic resonance spectral data. It completely inhibited the mycelial growth of B. cinerea, Colletotrichum acutatum Simmonds, Colletotrichum gloeosporioides Simmonds, M. grisea and Pythium ultimum Trow over a range of 0.4-33.3 mg litre(-1). It also controlled the development of rice blast, tomato late blight, wheat leaf rust, barley powdery mildew and red pepper anthracnose (Colletotrichum coccodes (Wallr) S Hughes). The chemical was particularly effective in suppressing red pepper anthracnose by 95% at a concentration of 125 mg litre(-1).  相似文献

Population density estimates for many animal species are often difficult or expensive to obtain, and they rely on assumptions that, if violated, result in unmeasurable estimation errors. Density estimates also may be unnecessary for research or management purposes, because an index that tracks changes in a population within appropriate time and geographic constraints could provide the information necessary to make management decisions or to evaluate the impact of a control program. We review research on a passive tracking index where observations are made on a series of tracking plots placed on lightly used dirt roads. The number of sets of tracks (individual intrusions) are recorded for each species of interest on each plot on consecutive days. The mean number of intrusions over the plots is calculated for each day for each species. The index is the mean of the daily means. These design and measurement methods present valuable advantages over most traditional tracking plot methods. Because no scents or baits are used as attractants, no conditioning of animals to the plots biases the results. This also permits multiple species, predator and prey alike, to be simultaneously monitored. Using the number of animal intrusions as observations produces results that are far more sensitive to change than tracking surveys where only presence or absence of spoor are recorded for each plot. Of particular importance, the statistical properties inherent to this data structure permit calculation of standard errors, confidence intervals and statistical tests, without subjectively subdividing the data.  相似文献

利用单克隆抗体H19及间接免疫荧光抗体反应(IFA),从经马立克病病毒(MDV)强毒株Mdll的38kd磷蛋白(pp38)基因克隆质粒DNA转染的MDV疫苗毒CVl988株感染的鸡胚成纤维细胞中。筛选到一个点突变株CVI/rpp38(AA)。DNA序列测定结果表明,该点突变株pp38基因ORF的第320位和326位碱基均已从亲本CVl988株的“G”突变为“A”,导致第107和109位氨基酸分别从精氨酸和甘氨酸转变为谷氨酰胺和谷氨酸。这个双氨基酸点突变株在IFA和免疫沉淀反应中与pp38特异性单抗H19都呈现阳性反应。但与另一个pp38特异性单抗T65都完全不反应,表现出与亲本病毒完全相反的结果。本研究发现了MDV的pp38上二个特异性不同的抗原表位,并以确凿的证据证明了第107及109位的谷氨酰胺和甘氨酸分别在这二个抗原表位形成中的关键作用。  相似文献

Anti-Yersinia ruckeri egg yolk immunoglobulin (IgY) was transferred to egg yolk after immunization of White Leghorn hens with formalin-killed whole cells of serovar 1 (RS1154) and serovar 2 (RS1153)Y. ruckeri and its lipopolysaccharide (LPS). The IgY was specific for its homologous LPS in western immunoblot, whereas some protein bands were commonly recognized, even by IgY from eggs of unimmunized hens. Purified LPS from both Y. ruckeri serovar types 1 and 2 had a very poor immunogenicity. The IgY activity was stable when processed into pellet form by a microbial transglutaminase treatment and showed a considerable resistance against acid pepsin for at least 2 h. Feeding specific anti-serovar 1 Y. ruckeri IgY to fish either before or after immersion infection produced marginal reductions in mortalities and in intestine infection. The same IgY did passively protect rainbow trout against infection when administered by intraperitoneal injection 4 h before an immersion challenge.  相似文献

Approximately 7,000 accessions of Korean soybean (Glycine max (L.) Merrill) landraces, largely composed of three collections, the Korea Atomic Energy Research Institute’s soybean (KAS), the Korean Crop Experiment Station’s soybean (KLS) and the Korean Agricultural Development and Technology Center’s soybean (KADTC) collections, have been conserved at the Rural Development Administration (RDA) genebank in Korea. The accessions within collections were classified based on their traditional uses such as sauce soybean (SA), sprouted soybean (SP), soybean for cooking with rice (SCR), and OTHERS. A total of 2,758 accessions of Korean soybean landraces were used to profile and to evaluate genetic structure using six SSR loci. A total of 110 alleles were revealed by at the six SSR loci. The number of alleles per SSR locus ranged from 9 to 39 in Satt187 and Satt_074, respectively. The number of alleles ranged from 87 in the KADTC collection to 96 in the KLS collection, and from 63 in the SCR group to 95 in the SP group. Nei’s average genetic diversity ranged from 0.68 to 0.70 across three collections, and 0.64 to 0.69 across the usage groups. The average between-group differentiation (G st) was 0.9 among collections, and 4.1 among the usage groups. The similar average diversity among three collections implies that the genetic background of the three collections was quite similar or that there were a large number of duplicate accessions in three collections. The selection from the four groups classified based upon usage may be a useful way to select accessions for developing a Korean soybean landrace core collection at the RDA genebank. DNA profile information of accessions will provide indications of redundancies or omissions and aid in managing the soybean collection held at the RDA genebank. The information on diversity analysis could help to enlarge the genetic diversity of materials in breeding programs and could be used to develop a core collection.  相似文献

The mechanisms of Marek's disease virus (MDV) entry to host cells have not yet been analyzed. Heparan sulfate (HS) on the cell surface serves as a receptor for several herpesviruses in mammalian species. In this study, we demonstrated that plaque formation by cell-free MDV is inhibited by the addition of soluble heparin to the cell culture. Moreover, pretreatment of susceptible cells, chicken embryo fibroblasts, with heparinase, partially reduced infectivity of the cell-free MDV. From these results, it was suggested that the MDV entry, at least in the case of cell-free MDV, is dependent on the presence of cell surface glycosaminoglycans, principally HS.  相似文献

To evaluate the transplacental transfer of Theileria sergenti infection in cattle, we used DNA probes to detect T. sergenti in 6 pregnant cows and their calves. All the animals were monitored by parasitologic, serologic, and polymerase chain reaction (PCR) assays for a predicted 875-base-pair (bp) DNA product and a 684-bp amplicon detected by nested PCR in the blood and spleens of aborted fetuses. An open reading frame (ORF) starting at nucleotide 170 and terminating at position 1021 was shown to code for a polypeptide of 283 amino acid residues. All 6 dams and 5 calves were positive for T. sergenti in all tests. One calf was positive only with nested PCR. We conclude that transplacental transmission of T. sergenti is a significant problem. The relevance of the data in the programmed introduction of new (especially pregnant) animals into established clean herds needs serious consideration with regard to control of theileriosis and other tickborne diseases.  相似文献

Until now, continental shelf environments have been monitored with highly localized line-transect methods from slow-moving research vessels. These methods significantly undersample fish populations in time and space, leaving an incomplete and ambiguous record of abundance and behavior. We show that fish populations in continental shelf environments can be instantaneously imaged over thousands of square kilometers and continuously monitored by a remote sensing technique in which the ocean acts as an acoustic waveguide. The technique has revealed the instantaneous horizontal structural characteristics and volatile short-term behavior of very large fish shoals, containing tens of millions of fish and stretching for many kilometers.  相似文献

This study was conducted to evaluate the influence of seed priming on drought tolerance of pigmented and non‐pigmented rice. Seeds of pigmented (cv. Heug Jinju Byeo) and non‐pigmented (cv. Anjoong) rice were soaked in water (hydropriming) or solution of CaCl2 (osmopriming). Seeds were sown in soil‐filled pots retained at 70 (well‐watered) and 35% (drought) water‐holding capacity. Drought stress caused erratic and poor stand establishment and decreased the growth of both rice types. More decrease in plant height and leaf area under drought stress was noted in pigmented rice, whereas decrease in root length and seedling dry weight, under drought, was more obvious in non‐pigmented rice. Pigmented rice maintained more tissue water and photosynthesis and had more polyphenols, flavonoids and antioxidant activity than non‐pigmented rice. Seed priming was effective in improving stand establishment, growth, polyphenols, flavonoids and antioxidant activity; however, extent of improvement was more in pigmented rice under drought. In conclusion, drought caused erratic germination and suppressed plant growth in both rice types. However, pigmented rice had better drought tolerance owing to uniform emergence, and better physiological and morphological plasticity. Seed priming was quite helpful in improving the performance of both rice types under drought and well‐watered conditions.  相似文献
