The persistence of plant-derived recombinant DNA in sheep and pigs fed genetically modified (Roundup Ready) canola was assessed by PCR and Southern hybridization analysis of DNA extracted from digesta, gastrointestinal (GI) tract tissues, and visceral organs. Sheep (n = 11) and pigs (n = 36) were fed to slaughter on diets containing 6.5 or 15% Roundup Ready canola. Native plant DNA (high- and low-copy-number gene fragments) and the cp4 epsps transgene that encodes 5-enolpyruvyl shikimate-3-phosphate synthase were tracked in ruminal, abomasal, and large intestinal digesta and in tissue from the esophagus, rumen, abomasum, small and large intestine, liver, and kidney of sheep and in cecal content and tissue from the duodenum, cecum, liver, spleen, and kidney of pigs. High-copy chloroplast-specific DNA (a 520-bp fragment) was detected in all digesta samples, the majority (89-100%) of intestinal tissues, and at least one of each visceral organ sample (frequencies of 3-27%) from sheep and swine. Low-copy rubisco fragments (186- and 540-bp sequences from the small subunit) were present at slightly lower, variable frequencies in digesta (18-82%) and intestinal tissues (9-27% of ovine and 17-25% of porcine samples) and infrequently in visceral organs (1 of 88 ovine samples; 3 of 216 porcine samples). Each of the five cp4 epsps transgene fragments (179-527 bp) surveyed was present in at least 27% of ovine large intestinal content samples (maximum = 64%) and at least 33% of porcine cecal content samples (maximum = 75%). In sheep, transgene fragments were more common in intestinal digesta than in ruminal or abomasal content. Transgene fragments were detected in 0 (esophagus) to 3 (large intestine) GI tract tissues from the 11 sheep and in 0-10 of the duodenal and cecal tissues collected from 36 pigs. The feed-ingested recombinant DNA was not detected in visceral tissues (liver, kidney) of lambs or in the spleen from pigs. Of note, however, one liver and one kidney sample from the pigs (different animals) were positive for a 278-bp fragment of the transgenic cp4 epsps (denoted F3). Examination of genomic libraries from these tissues yielded no conclusive information regarding integration of the fragment into porcine DNA. This study confirms that feed-ingested DNA fragments (endogenous and transgenic) do survive to the terminal GI tract and that uptake into gut epithelial tissues does occur. A very low frequency of transmittance to visceral tissue was confirmed in pigs, but not in sheep. It is recognized that the low copy number of transgenes in GM feeds is a challenge to their detection in tissues, but there was no evidence to suggest that recombinant DNA would be processed in the gut in any manner different from endogenous feed-ingested genetic material. 相似文献

This study investigated the potentiation of porcine circovirus 2 (PCV2)-induced postweaning multisystemic wasting syndrome by porcine parvovirus (PPV) and found it was associated with excessive production of tumor necrosis factor-alpha (TNF-alpha). Colostrum-deprived conventional pigs were inoculated intranasally with PCV2 or PPV alone or in combination (PCV2 and PPV). In vitro assay of TNF-alpha, obtained from alveolar macrophages coinfected with PCV2 and PPV, showed a significant increase in TNF-alpha compared to single infection of macrophages with either PCV2 or PPV alone (P < 0.05). All pigs inoculated with PCV2 and PPV developed severe postweaning wasting syndrome, whereas clinical signs (e.g., weight loss) were present but perhaps less severe in either PCV2- or PPV-inoculated pigs. Compared to the pigs inoculated with PCV2 or PPV alone, pigs inoculated dually with PCV2 and PPV showed significantly (P < 0.05) increased levels of TNF-alpha. Levels of TNF-alpha in the sera were reversely correlated with the body weight in pigs experimentally infected with dual inoculation of PCV2 and PPV (r(s) = -0.92, P < 0.001). Poly(vinyl alcohol) (PVA)/Ag-zeolite nanofiber webs were prepared with different concentrations of Ag-zeolite nanoparticles
by the electrospinning technique. Scanning electron microscopy (SEM), energy-dispersive X-ray spectrometry (EDS), transmission
electron microscopy (TEM), Fourier transform-infrared (FT-IR) spectroscopy, thermogravimetric analysis (TGA), differential
scanning calorimetry (DSC), Instron, and antibacterial activities analysis were utilized to characterize the morphology and
properties of the PVA/Ag-zeolite nanofiber webs. The study results showed that the polymer concentration, applied voltages
and tip-to-collector distances were the main factors influencing the morphology of the electrospun nanofiber webs. The introduction
of Ag-zeolite nanoparticles improved the mechanical properties and thermal stability of the PVA nanofiber webs. TEM data demonstrated
that the Ag-zeolite nanoparticles were well distributed within the nanofiber. FTIR revealed a possible interaction between
the PVA matrix and the Ag-zeolite nanoparticles. These fibers showed an antibacterial efficacy of 99.8 % and over against
Staphylococcus aureus and Klebsiella pneumoniae at Ag-zeolite concentrations of 1 % and over, because of the presence of the

Triphenyl phosphate (TPP) is well known to be one of the most effective flame retardants for acrylonitrile-butadiene-styrene
copolymer (ABS) and its blending resins, such as polycarbonate (PC)/ABS, among various phosphorous-based compounds. However,
TPP can also play a role as a plasticizer, which decreases the mechanical properties of PC/ABS resins at high temperature.
Furthermore considerable amount of TPP has to be evaporated during molding process due on its much lower evaporation temperature.
To overcome these shortcomings, we tried to immobilize TPP by grafting on butadiene moiety of ABS. FT-IR analysis of prepared
TPP-grafted ABS (ABS-g-TPP) comparing with TPP, ABS and their blend confirmed that chemical reactions happened between TPP
and ABS resins and it was attributed to the graft reaction of TPP onto butadiene moieties. Prepared ABS-g-TPP resins were
blended with PC at various compositions to be prepared as testing specimens by injection molding. The physical characteristics
such as mechanical properties, thermal stability, and flame retarding properties of the PC/ABS-TPP graft copolymer were analyzed
through Vicat softening temperature, IZOD impact strength, transmission electron microscope, and UL94 flame retardation tests.
Results showed that PC/ABS-g-TPP resin takes better thermomechanical properties than the existing PC/ABS resins at relatively

Carbon nanotube (CNT)/magnetite (Fe3O4) nanocomposites were successfully fabricated by nanocoating. When the CNT/Fe3O4 nanocomposites were added to a carbonyl (CI) based magnetorheological fluid (MRF), the magnetic rheological properties of
the MRF were enhanced. Such enhancement in magnetic rheological properties of the CI-CNT/Fe3O4 MRF is considered to be attributable to both the superparamagnetic property of the magnetic layer covering the CNT surface
and the high length to diameter ratio of the CNT/Fe3O4 nanocomposite. Impact properties for CI-CNT/Fe3O4 MRF treated Kevlar fabrics were also tested, and improved impact properties were observed with external magnetic fields compared

A grid-based, KIneMatic wave STOrm Runoff Model (KIMSTORM) is described. The model adopts the single flow-path algorithm and
routes the water balance during the storm period. Manning’s roughness coefficient adjustment function of the paddy cell was
applied to simulate the flood mitigation effect of the paddy fields for the grid-based, distributed rainfall-runoff modeling.
The model was tested in 2296 km2 dam watershed in South Korea using six typhoon storm events occurring between 2000 and 2007 with 500 m spatial resolution,
and the results were tested through the automatic model evaluation functions in the model. The average values of the Nash–Sutcliffe
model efficiency (ME), the volume conservation index (VCI), the relative error of peak runoff rate (EQp), and the absolute error of peak runoff (ETp) were 0.974, 1.016, 0.019, and 0.45 h for calibrated storm events and 0.975, 0.951, 0.029, and 0.50 h for verified storm
events, respectively. In the simulation of the flood mitigation effect of the paddy fields, the average values of the percentage

The antifungal effects of citral, eugenol, nerolidol and α-terpineol on Trichophyton mentagrophytes were investigated. Citral over 0.1 mg/ml strongly inhibited the hyphal growth of T. mentagrophytes, and the antifungal activity of α-terpineol was less effective. The morphological changes of the fungus exposed to the terpenes were observed by electron microscopy. The hyphae were distorted and collapsed at 0.2, 0.4 and 1 mg/ml of eugenol, nerolidol and α-terpineol respectively, and cell membrane and organelles were irreversibly damaged at 0.2 mg/ml citral. 相似文献

This study was conducted to determine carbon (C) dynamics following forest tending works (FTW) which are one of the most important forest management activities conducted by Korean forest police and managers. We measured organic C storage (above- and below-ground biomass C, forest floor C, and soil C at 50 cm depth), soil environmental factors (soil CO2 efflux, soil temperature, soil water content, soil pH, and soil organic C concentration), and organic C input and output (litterfall and litter decomposition rates) for one year in FTW and non-FTW (control) stands of approximately 40-year-old red pine (Pinus densiflora S. et Z.) forests in the Hwangmaesan Soopkakkugi model forest in Sancheonggun, Gyeongsangnam-do, Korea. This forest was thinned in 2005 as a representative FTW practice. The total C stored in tree biomass was significantly lower (P < 0.05) in the FTW stand (40.17 Mg C ha−1) than in the control stand (64.52 Mg C ha−1). However, C storage of forest floor and soil layers measured at four different depths was not changed by FTW, except for that at the surface soil depth (0–10 cm). The organic C input due to litterfall and output due to needle litter decomposition were both significantly lower in the FTW stand than in the control stand (2.02 Mg C ha−1 year−1 vs. 2.80 Mg C ha−1 year−1 and 308 g C kg−1 year−1 vs. 364 g C kg−1 year−1, respectively, both P < 0.05). Soil environmental factors were significantly affected (P < 0.05) by FTW, except for soil CO2 efflux rates and organic C concentration at soil depth of 0–20 cm. The mean annual soil CO2 efflux rates were the same in the FTW (0.24 g CO2 m−2 h−1) and control (0.24 g CO2 m−2 h−1) stands despite monthly variations of soil CO2 efflux over the one-year study period. The mean soil organic C concentration at a soil depth of 0–20 cm was lower in the FTW stand (81.3 g kg−1) than in the control stand (86.4 g kg−1) but the difference was not significant (P > 0.05). In contrast, the mean soil temperature was significantly higher, the mean soil water content was significantly lower, and the soil pH was significantly higher in the FTW stand than in the control stand (10.34 °C vs. 8.98 °C, 48.2% vs. 56.4%, and pH 4.83 vs. pH 4.60, respectively, all P < 0.05). These results indicated that FTW can influence tree biomass C dynamics, organic C input and output, and soil environmental factors such as soil temperature, soil water content and soil pH, while soil C dynamics such as soil CO2 efflux rates and soil organic C concentration were little affected by FTW in a red pine stand. 相似文献

This study assessed the altitudinal variations in the anthocyanin and isoflavone contents of six black seed coated soybean
[Glycine max (L.) Merrill] cultivars. The black soybean cultivars Heugcheong, Seonheuk, Geomjeong 1, Geomjeong 2, Cheongja 2, and Cheongja
3 were planted at Milyang (12 m above mean sea level — low altitude) and Muju (600 m — high altitude), Korea on 10 June 2005
and 2006. The total anthocyanin and isoflavone contents and individual components were investigated by reverse-phase high
performance liquid chromatography (HPLC). All black soybean cultivars cultivated in high altitude possessed significantly
higher total anthocyanin (p < 0.01) and isoflavone (p < 0.01) contents than those grown in low altitude. For anthocyanin composition, cyanidin-3-O-glucoside, cyanidin-3-O-galactoside, and peonidin-3-O-glucoside contents were significantly higher while delphinidin-3-O-glucoside contents was significantly lower at high altitude. The composition of individual isoflavones, 6″-O-malonyldaidzin, and 6″-O-malonylgenistin contents significantly increased at high altitude. 相似文献
