Single-nucleotide polymorphisms (SNPs) constitute an abundant source of DNA polymorphisms, which have been successfully used
to identify loci that are associated with a particular phenotype. Additionally, such markers could be efficiently used in
combination with doubled haploid technology to improve the efficiency of breeding programmes. Information on such markers
in plants is still scarce. For bread wheat, SNP data are restricted to a few genes. This can be explained by the hexaploidy
of Triticum aestivum which makes SNP discovery difficult. We developed a novel method for SNP discovery in bread wheat. The strategy is based
on the development of highly specific PCR-primers, which were used to sequence 27 lines. SNPs were discovered from sequence
alignment data. Some SNPs were identified by mass spectrometry in a collection of 113 lines, which were both evaluated for
agronomic traits and genotyped at 42 neutral microsatellite loci. Traits investigated include: protein content, the quantity
of high-molecular-weight glutenins and that of the GluBx subunit. The 42 markers were used to infer population structure,
which was included in linear models for association studies. The results of this preliminary study showed 89 SNPs in approximately
20 kbp, i.e., one SNP every 223 bp on average. Six SNPs were genotyped: three were located along the sequence of Glu-B1-1, while three non-synonymous SNPs were located along the sequence of the B homoeologous gene coding for SPA (Storage Protein
Activator). The SNPs from Glu-B1-1 had a significant effect on the studied variables, whereas those of SPA had no effect. Such results might indicate that some haplotypes for Glu-B1-1 are linked to higher protein content, through an increased amount of high-molecular-weight glutenins, especially the GluBx

The objective of this study was to determine if deoxynivalenol (DON) exposure alters the susceptibility of rainbow trout to bacterial coldwater disease caused by Flavobacterium psychrophilum. Rainbow trout were fed a nutritionally complete diet containing corn that was naturally contaminated with DON at a desired concentration of <0.5 (control and pair‐fed treatments), 4 or 6 ppm over 7 weeks to apparent satiation. After 4 weeks, fish were infected by intraperitoneal injection with F. psychrophilum (3.03x106 CFU mL?1) via intraperitoneal injection and monitored for morbidity and mortality. A significant linear reduction in feed intake was associated with increasing dietary levels of DON contamination over the initial 4 weeks. There was a significant reduction (P < 0.05) in cumulative per cent mortality in DON‐fed groups (4.1 ppm, 11%; 5.9 ppm, 7%) in comparison to control (46%) and pair‐fed (25%) groups at 21 days post infection. Mortality of trout pair‐fed the control diet was also significantly lower (P < 0.05) than the control group fed to apparent satiation. A replicate trial using genetically similar fish and the same experimental design produced similar results. These results suggest that DON exposure and restricted feed intake provided a protective effect for rainbow trout infected with F. psychrophilum. 相似文献

Digital maps of soil properties are now widely available. End-users now can access several digital soil mapping (DSM) products of soil properties, produced using different models, calibration/training data, and covariates at various spatial scales from global to local. Therefore, there is an urgent need to provide easy-to-understand tools to communicate map uncertainty and help end-users assess the reliability of DSM products for use at local scales. In this study, we used a large amount of hand-feel soil texture (HFST) data to assess the performance of various published DSM products on the prediction of soil particle size distribution in Central France. We tested four DSM products for soil texture prediction developed at various scales (global, continental, national, and regional) by comparing their predictions with approximately 3 200 HFST observations realized on a 1:50 000 soil survey conducted after release of these DSM products. We used both visual comparisons and quantitative indicators to match the DSM predictions and HFST observations. The comparison between the low-cost HFST observations and DSM predictions clearly showed the applicability of various DSM products, with the prediction accuracy increasing from global to regional predictions.
