This study evaluated the acute effect of the administration of coffee brew in the activity of phase II antioxidant enzymes in the hepatic tissue of rats. A single dose of this beverage increased the activity of the enzymes SOD, CAT, and GPx; the maximum increase occurred 1 h after administration (19.1, 22.1, and 25.1%, respectively). These changes were statistically significant (p < 0.05), the response was shown to be dose-dependent (p < 0.05), and the return to basal levels took >4 h from the intervention, suggesting a long-term effect. The total antioxidant capacity of the hepatic tissue also exhibited a peak 1 h after the intervention (6.5%), but the increase was not statistically significant and the response was not dose-dependent due to the low exposure to coffee. These results indicate that coffee increases the activities of antioxidant enzymes, improving protection against oxidative stress. 相似文献

Purpose
Standard assays for phytotoxicity provide a reductionist view on the performance of plants under toxic stress. To address two of the most prominent issues in plant toxicity studies, our aims were (1) to assess how well physiological and biochemical parameters complement standard toxicological endpoints when testing natural soils and (2) to assess the suitability of three commonly used control soils as comparative references.Material and methods
We compared the performance of Zea mays and Helianthus annuus in three control soils (artificial Organisation for Economic Co-operation and Development (OECD) soil, standard LUFA 2.2 soil, and turf-perlite) against three natural soils representing a gradient of contamination (from a deactivated uranium mine). Standard endpoints (emergence and biomass) were estimated, along with pigment content, photosynthetic parameters, cellular injury, and proline content.Results and discussion
The toxicological profile of natural soils was highly dependent on the control soil used as reference; also, plant physiological performance was influenced by the soils’ properties. We discuss the need to interpret and combine multiple lines of evidence as a way to increase the degree of confidence one classifies soils based on their ecotoxicity, and this is where the integration of physiological and biochemical parameters bring added value.Conclusions
When facing large variability in soil characteristics, it is best to collect and integrate as much information possible to strengthen conclusions about phytotoxicity of natural soils. Obviously, this refutes reductionist views and places the final conclusion in the hands of expert judgment.443.

Methylxanthinic alkaloids and phenolic compounds are related to the therapeutic properties of Ilex paraguariensis infusions. Considering the known vascular tropism of xanthines, an aqueous extract (mate) and caffeine were evaluated on blood vessel formation, in connection with the analysis of those secondary metabolites, which was performed in young and mature leaf samples collected in three cultivation systems located in the southern region in Brazil (Santa Catarina State). Samples of young and mature leaves from a monoculture cultivation system (MC) showed the highest content of phenolic compounds (149.68 microg/mL, young leaves; 135.50 microg/mL, mature leaves) and caffeine (young leaves, 148.07 microg/mL; mature leaves, 244.63 microg/mL) as compared to samples from agroforesty (AF) and shaded-native (NT) cultures. Theophylline was not detected in samples by reverse phase-high performance liquid chromatography, and mature leaves showed lower theobromine amounts (11.46 microg/mL). Treatments performed with mate aqueous extract and caffeine (1.03-4.12 microM/disk) in the yolk sac vascular membranes of 2-day-old chick embryos revealed pro-vasculo- and angiogenic properties as well as embryonic growth enhancement. These findings, uncoupled from any detectable embryotoxic effect, suggest a potential therapeutic and/or prophylactic use in cardiovascular disorders for caffeine and related constituents of mate plant extracts, an issue that waits further studies. 相似文献

Thermal analysis could rapidly and easily predict nutritional value of ruminant feeds. The hypothesis is that crambe meal (CM) has a quality similar to that of soybean meal (SM), and the objective of this study was to determine the nutritional characteristics of CM and compare them to those of SM. CM had greater concentrations of phytic acid (26.3 vs. 16.0 g/kg) and phenol compounds (615 vs. 393 mg gallic acid (GAE)/kg) than SM. In vitro dry matter (DM) digestibility was lower for CM than SM (752 vs. 975 g/kg DM). Cumulative in vitro gas production at 48 h of incubation (14.1 vs. 19.4 mL/100 mg substrate DM), and energy release (4.5 vs. 5.7 kJ/g substrate DM) were lower for CM than SM. CM had a higher concentration of low digestible fiber, hence degradability of DM was lower and the proportion of indigestible fraction was greater for CM than SM. High concentrations of indigestible compounds were likely responsible for lower gas production of CM compared to SM. These results suggest CM quality is lower than that of SM and that thermal analysis is a useful tool to precisely determine the nutritive value of oilseed residues. 相似文献

Choline is an essential nutrient in poultry diets because it performs various important metabolic functions. The objective of this study was to re‐evaluate the choline requirements of male broiler chickens from 1 to 21 days of age at two levels of methionine. Two assays using 2,160 Cobb® chickens (1,080 in each assay) were conducted. The study design was completely randomized and consisted of six treatments and six replicates, with 30 animals per experimental unit. The semipurified basal diet was formulated with corn, soya bean meal, soya protein concentrate, starch and sugar, providing 390 mg/kg choline and 0.593% digestible methionine (requirement level) in Assay 1 and a reduction of about one‐quarter in the requirement level of digestible methionine (0.440%) in Assay 2. Choline chloride (62.5%) was added by a supplementation technique to both basal diets to compose crescent levels of choline supplementation (715, 1,040, 1,365, 1,690 and 2,015 mg/kg). The weight gain responses were fitted using quadratic polynomial (QP) and broken‐line (BL) models. The ideal intake of choline (mg/bird.day) was estimated from the first intercept of the QP with the BL plateau (BL + QP). The results showed that the diet with the 25% reduction in digestible methionine limited the maximum weight gain by approximately 10%. The choline requirements of broilers from 1 to 7, 1 to 14 and 1 to 21 days of age were 27,013, 44,458 and 62,535 mg/bird.day, respectively, for the requirement level of digestible methionine and 26,796, 41,820 and 56,578 mg/bird.day for the broilers receiving the diet with the 25% reduction in digestible methionine. 相似文献

Rumen micro‐organisms are capable of producing microbial protein from ammonia and carbon skeleton, and non‐protein nitrogen (NPN) may be one of the sources of ammonia. Alternative source of NPN is the slow release of ammonia sources in which the product is the extrusion of starch with urea. This work aimed to determine the effects on nutrient intake, ingestive behaviour, digestibility, nitrogen balance, ruminal pH, rumen ammonia nitrogen, volatile fatty acids (VFA) and blood parameters with increased levels of extruded urea (50, 60, 70 and 80 g/100 kg of body weight [BW]) in beef cattle diet. Four rumen cannulated crossbred steers with initial mean weight of 336 ± 47 kg in a 4 × 4 Latin square design were distributed. Diets were formulated with 400:600 g/kg roughage:concentrate ratio on dry matter based and provided once per day, being used whole corn silage as roughage. There were no effects on nutrient intake (kg/day), ingestive behaviour, apparent digestibility, nitrogen balance, volatile fatty acid (VFA) and blood parameters in extruded urea treatment groups. Similar results were observed on time spent on feeding, rumination and idleness. There were positive linear effects (p = 0.022) on rumen pH in the time of 8 hr after feeding and also on plasma concentration of the extruded urea levels (p = 0.039); whereas a linear negative effect (p = 0.030) was observed on ammonia nitrogen for the collection time of 2 hr after feeding. Increased levels of extruded urea could maintain nutrient intake, digestibility, ingestive behaviour, rumen pH and blood parameters in normal conditions. In conclusion, we recommend the extruded urea use with values up to 80 g/100 kg BW in confined beef cattle that receive balanced diets with 140 g/kg of crude protein. 相似文献

Long-term agricultural sustainability and water quality may be threatened by inadequate land management. Carbon (C) losses at the catchment scale largely depend on land use and management practices. In “Abelar” farm (A Coruña, NW Spain), swine slurry was directly discharged during a period of about 30 years onto agricultural maize fields and rangeland under cattle production. In 1998, the agricultural fields and rangeland were planted to stands of Eucalyptus globulus. The purpose of this work was to assess the concentrations and losses of total, inorganic, and organic carbon at the outlet of a small catchment (10.7 ha) in the period from March 2007 to December 2010. Nearly 190 water samples were collected; specifically eleven in 2007, sixty-eight in 2008, fifty-six in 2009, and fifty-three in 2010. Total runoff volumes from the catchment were also measured during the 3.7-year study period. Except for a peak recorded in November 2008, total carbon values (TC) ranged from 1.51 to 6.14 mg L?1, total inorganic carbon (TIC) varied between 0.54 and 3.30 mg L?1, and total organic carbon (TOC) was between 0.38 and 2.83 mg L?1; the exception was due to an occasional pig slurry discharge occurring in 2008, which drove the greatest values of the three carbon forms. The season of the year influenced concentrations of total, organic, and inorganic carbon. Total, organic, and inorganic carbon loads exported at the catchment outlet were estimated at the monthly and yearly time scales. Overall, 10 years after planting to the forest, dissolved and total carbon losses were rather small. 相似文献

Rumen acidosis is a common metabolic disorder occurring when organic acid production exceeds clearance capacity, reducing ruminal pH. The occurrence of acidosis has been directly correlated to the ratio of concentrate to forage in the diet. However, rates of substrate fermentation and acid absorption vary at different locations in the reticulo-rumen. The objective of this study was to determine the pH and redox potential (Eh) in different locations of the reticulo-rumen using 16 ruminally cannulated steers (309 ± 43 kg) receiving different supplementation levels of quebracho extract (QT; Schinopsis balansae) within a grower type diet (CP: 13.4%; total digestible nutrients [TDN]: 70.4%; and ME: 2.55 Mcal/kg, dry matter [DM] basis). Animals were randomly assigned to one of four dietary treatments: QT at 0%, 1%, 2%, and 3% of DM (QT0, QT1, QT2, and QT3, respectively), containing about 0%, 0.7%, 1.4%, and 2.1% of condensed tannins (CT), DM basis, respectively. Animals were adapted to the basal diet for 12 d before being introduced to predetermined treatments for 4 weeks (wk), with diets provided twice daily to allow ad libitum intake. Weekly measurements of ruminal fluid pH and Eh were taken 4 h post-feeding using a portable pH meter with two probes (pH and redox) in four locations of the reticulo-rumen (reticulum, cranial sac, dorsal sac, and ventral sac). Data were analyzed using a random coefficients model with the pen as a random effect and wk as repeated measures, with DM intake included as a covariate. There was no interaction among treatments, location, and wk (P ≥ 0.882) on reticulo-ruminal pH. Overall, ruminal pH was lower for QT0 and QT1 compared to QT3 (P < 0.001). The pH in the reticulum was greater than those of the ventral and dorsal sacs (6.05 vs. 5.94, 5.89, respectively; P ≤ 0.001) but similar to cranial sac (6.00). Reticular pH was positively correlated with the ruminal locations (≥0.78; P < 0.001). The linear equation to estimate ruminal mean pH using reticulum pH had an intercept and slope different from zero (P ≤ 0.04), but CT (% DM) was not different from zero (P = 0.15), root mean square error of 0.15, and R2 of 0.778: 0.723 (±0.36) + 0.857 (±0.059) × reticulum pH + 0.033 (±0.023) × CT. The Eh was lower for QT0 in week 1 than all other treatments (P < 0.001). We concluded that reticulo-ruminal pH differs among locations in the rumen regardless of QT supplementation level and days on feed, with reticular pH being the highest. 相似文献
