Upland hardwood stands on mesic, escarpment-oriented sites on the Cumberland Plateau region of northeastern Alabama provide a myriad management opportunities. Stands are primarily managed for Quercus, but the high species diversity allows for management that targets multiple species. Stand composition is unique in that dominant species include shade tolerant species such as Acer saccharum, intermediate tolerant Quercus spp. and Carya spp., and intolerant Liriodendron tulipifera. Three replications of five levels of disturbance were created to assess species compositional changes; disturbances included three levels of harvest intensity, a mid-story herbicide treatment, and a control. After eight growing seasons, there were no discernable differences in species richness, diversity or evenness. Importance values based on relative basal area and relative density also changed little, except for clearcuts where L. tulipifera greatly increased. An initial gradient in basal area, canopy cover, and light created by harvesting or thinning dissipated following five growing seasons. Options exist for future stand management, including promoting two-aged or uneven-aged systems. Maintenance of Quercus will require additional tending.  相似文献

The profile of catechol estrogen formation by pig conceptuses from d 14 to 20 of pregnancy (d 0 = 1st d of estrus) was studied using a direct product isolation assay for estrogen-2/4-hydroxylase (E-2/4-H). High performance liquid chromatography was coupled to detection of radioactive products in a flowing system to separate the 2- and 4-hydroxyestradiol (2- and 4-OH-E2) formed from estradiol. The major product was 2-OH-E2; its formation increased with increasing amounts of nicotinamide cofactors and exhibited a preference for NADPH. Maximum velocities were 909 and 154 pmol.mg protein-1.30 min-1, and apparent Michaelis constants were 7.14 and 5.12 microns, for 2- and 4-OH-E2 formation, respectively. Homogenates of d-13 conceptuses produced 1,151 +/- 142 pmol.mg protein-1.30 min-1 of 2-OH-E2. Estradiol-2-hydroxylase was affected (P less than .01) by day of pregnancy, and day effects from d 14 to 20 were described by a fourth-order equation (P less than .001). Conceptuses collected on d 14 produced 119 +/- 15.4 pmol of 2-OH-E2.mg protein-1.30 min-1. By d 15, E-2-activity had declined to 15.7 +/- 1.8 pmol.mg protein-1.30 min-1. Formation of 2-OH-E2 increased to 64.2 +/- 15.4 pmol.mg protein-1.30 min-1 on d 17, remained stable through d 19, and was 39.4 +/- 13.8 pmol.mg protein-1. 30 min-1 on d 20. Therefore, E-2/4-H activity appears to be regulated from d 14 to 20 of pregnancy in pigs; it may participate in the establishment of pregnancy.  相似文献

A recombinant antigen-based single serum dilution enzyme-linked immunosorbent assay (ELISA) was developed to measure the specific antibody activity in sera of dogs with leptospirosis. The recombinant antigen developed and used in the assay was specific for the pathogenic serovars of Leptospira. A linear relationship was found to exist between the predicted antibody titres at a single working dilution of 1:1000 and the corresponding observed serum titres as determined by the standard serial-dilution method. Regression analysis was used to determine a standard curve from which an equation can be derived that allows demonstration of the mentioned correlation. The equation was then used to convert the corrected absorbance readings of the single working dilution directly into the predicted ELISA antibody titres. The assay was proved to be sensitive, specific and accurate as compared to the standard microscopic agglutination test (MAT).  相似文献

ObjectiveTo investigate the contribution of K+ channels on peripheral antinociception induced by ketamine.Study designProspective experimental study.Animals110 male Wistar rats weighing 160–200 g.MethodsThe paw pressure required to elicit limb flexion was designated as the nociceptive threshold. Hyperalgesia was induced by intraplantar injection of prostaglandin E2. All drugs were administered locally into the right hind paw of rats. Ketamine was administered into the right hind paw 2 hours and 55 minutes after local injection of PGE2. Tetraethylammonium was administered 30 minutes prior to ketamine and the other K+ channel blockers, glibenclamide, dequalinium and paxilline, were administered 5 minutes prior to ketamine.ResultsProstaglandin E2 (2 μg per paw) induced hyperalgesia. Ketamine (10, 20, 40 and 80 μg per paw) elicited a local peripheral antinociceptive effect that was antagonized by a specific blocker of ATP‐sensitive K+ channels, glibenclamide (20, 40 and 80 μg per paw). In another experiment, the non‐selective voltage‐dependent K+ channel blocker tetraethylammonium (30 μg per paw) and small and large conductance blockers of Ca2+‐activated K+ channels, dequalinium (50 μg per paw) and paxilline (20 μg per paw), were ineffective at blocking the effect of a local ketamine injection.Conclusions and clinical relevanceAnalysis of these results provides evidence that ketamine, may in part, induce peripheral antinociceptive effects by ATP‐sensitive K+ channel pathway activation.  相似文献
