Global warming caused by an increase in the concentrations of greenhouse gases, is the direct result of greenhouse gas-induced radiative forcing. When a doubling of atmospheric carbon dioxide is considered, this forcing differed substantially among 15 atmospheric general circulation models. This study evaluated the phytochemical stability and organoleptic attributes of an ascorbic acid-fortified muscadine grape juice as affected by dense phase CO2 processing (DP-CO2) and addition of thyme polyphenolic cofactors (Thymus vulgaris; 1:100 anthocyanin-to-cofactor molar ratio) in efforts to prevent phytochemical losses that occur during storage of anthocyanin-containing beverages, especially in the presence of carbonyl compounds commonly produced during thermal processing and storage. DP-CO2 processing insignificantly altered initial juice phytochemical and antioxidant content, whereas thermal pasteurization reduced anthocyanins (263 mg/L), ascorbic acid (42 mg/L), soluble phenolics (266 mg/L), and antioxidant capacity (6 microM Trolox equivalents/mL). Similar trends were observed during storage, and data showed that increasing the CO2 level from 8 to 16% during DP-CO2 was instrumental in reducing juice phytochemical and antioxidant degradation. Copigmentation was instrumental in retaining higher anthocyanin, soluble phenolics, and antioxidant capacity during storage without affecting initial juice aroma and flavor characteristics. Moreover, on the basis of overall likeability scores, panelists preferred copigmented juices, which had increased juice color intensity and masked the detrimental color fading that occurred during storage, especially when compared to thermally pasteurized juices. DP-CO2 and copigmentation were effective strategies to reduce phytochemical and color deterioration that occurred in muscadine juice during storage without affecting their organoleptic attributes. 相似文献

Dietary intake is a major route of exposure to perfluorinated compounds (PFCs). Although fish and seafood contribute significantly to total dietary exposure to these compounds, there is uncertainty with respect to the effect of cooking on PFC concentrations in these foods. Eighteen fish species purchased from markets in Toronto, Mississauga, and Ottawa, Canada were analyzed for perfluorooctanesulfonamide (PFOSAs)-based fluorochemicals and perfluorinated acids (PFAs) in raw and cooked (baked, boiled, fried) samples. Of 17 analytes, perfluorooctanesulfonic acid (PFOS) was detected most frequently; concentrations ranged from 0.21 to 1.68 ng/g ww in raw and cooked samples. PFOSAs were detected only in scallops at concentrations ranging from 0.20 ng/g ww to 0.76 ng/g ww. Total concentrations of PFAs in samples were 0.21 to 9.20 ng/g ww, respectively, consistent with previous studies. All cooking methods reduced PFA concentrations. Baking appeared to be the most effective cooking method; after baking samples for 15 min at 163 C (325 degrees F), PFAs were not detected in any of the samples. The margin of exposures (MOE) between the toxicological points of reference and the dietary intake of perfluorocarboxylates (PFCAs) and PFOS in fish and seafood muscle tissue were greater than 4 orders of magnitude. This indicates that reducing consumption of fish muscle tissue is not warranted on the basis of PFC exposure concerns at the reported levels of contamination, even for high fish consuming populations. 相似文献

Commercial soy-based foodstuffs, including beverages ( n = 15), cow's milk supplemented with soy isoflavones ( n = 1), snacks ( n = 1), and biscuits ( n = 2), were analyzed to find any link between alterations in protein quality, safety (antigenicity), functionality (antioxidant activity), and food processing. Protein content was analyzed by the Kjeldhal method and available lysine by OPA assay. Chromatographic (RP-HPLC) and electrophoretic (SDS-PAGE) protein profiles were obtained to monitor modifications in the structure of soy allergens. The antigenicity was estimated by immunoblotting against soy total antibodies. Total phenol content was measured by Folin-Ciocalteu, while peroxyl radical scavenging activity of the sample was determined by ORAC FL assay. Protein content did not differ of those declared by the producers. Lysine availability was higher in liquid soy beverages compared to that in other soy foodstuffs studied here. 7S and 11S soy allergens were detected by RP-HPLC and SDS-PAGE, respectively. Both data indicated changes in soy protein patterns due to processing of instant powdered soymilk, soy snacks, and biscuits. Immunoblotting assay showed modifications in the antigenic response of these foodstuffs based on soy, suggesting that their processing had altered the structure of soy allergens. RP-HPLC, SDS-PAGE, and immunoblotting resulted in adequate analytical approaches for detecting changes in protein structure due to processing and adulteration. The aim was to assess the ability of bicarbonate-extractable P (Olsen P) to estimate total plant-available P (TPAP) in reclaimed marsh soils (Aeric Endoaquepts) which differed widely in P buffering capacity (PBC). Total plant-available P was estimated as the cumulative P uptake for a final concentration of 0.02 mg P/L in the soil solution which is the typical P requirement for field crops. The Olsen P estimated for that concentration was adopted as the critical level for crop production. We found that TPAP was better predicted by anion exchange resin-extractable P (AER-P) (65% of variance accounted for) than by Olsen P, probably because the effectiveness of the AER depends on the soil P buffering capacity, a factor that greatly influences the availability of P to plants. The critical Olsen P level was found to depend on those soil properties affecting the relationship between sorbed P and P in soil solution, viz. the P buffering capacity of soil, the Na/Ca mole ratio in the 1:1 soil:water extract, which explained 63 and 84% of the variance in the critical level, respectively, and the affinity of the sorbing surfaces for P. The Totonac homegarden is a traditionally designed agroecosystem mixing different elements, such as cultivated and wild plants, and livestock. Our objective was to understand the role and importance of homegardens as a strategy for subsistence and natural resources management. Anthropological fieldwork was carried out in Coxquihui, Veracruz, Mexico, a Totonac community. Conventional sampling using a questionnaire yielded a sample of 40 individuals, each representing a family group. Personal interviews, life stories, observations, and field transects enriched survey information. Fieldwork permitted identification of four types of Totonac homegardens: backyards, cropping fields, acahuales or fallow fields, and fences or field edges. Each of these gardens yields an array of products and services important for several cultural roles and natural resource management aims. Totonacs see land as the dominant and most critical resource. A great deal of terrain is steeply sloped and soils are poor. Homegardens play a key role in a production system that minimizes these site limitations, striking a balance between resource maintenance and subsistence needs. Their functions are ecological, to foster a multistrata vegetation cover, and a continuous supply of organic matter to the soil; economic, serving as living storehouses where diverse products (food, timber, firewood, forage, animals, ceremonial supplies, medicinal products), are kept through the annual cycle; and social, performing various social roles such as growing medicinal, ritual, and edible plants, thus supporting beliefs and culture continuity. Studies like this contribute to a better understanding of Totonac culture and native ecology, and give ideas for a better land management. 相似文献

ABSTRACT: High-pressure technology is used as an alternative to heat processing because of its inactivating effect on microorganisms and enzymes. However, it can also alter the structure of other muscle proteins. The present study compares the effects of high pressure (300 MPa, 7°C, 20 min) on the proteolytic degradation and alterations in the myofibrillar proteins of sardine and blue whiting muscle. Also, muscle homogenates and enzyme extracts were pressurized in order to evaluate the high-pressure effects on unprotected proteolytic enzymes outside the whole muscle structure. Peak proteolytic activity was found to occur at 55°C in both species. The peak activity pH was pH 3 for the sardine and pH 8 for the blue whiting; the main enzyme families being aspartic proteases in the former and alkaline serine proteases in the latter. Pressurization lowered activity levels at the peak activity pH and temperature in the fish muscle (by 30.8% in the sardine and by 9.5% in the blue whiting) and also slightly in the enzyme extracts (by 16.8% in the sardine and by 19.4% in the blue whiting). The electrophoretic profiles disclosed higher protein degradation in the pressurized muscle. Overall, the observed changes in proteolytic activity can be attributed not only to the effect of high pressure on the enzymes, but also and mainly, to the effect on other muscle proteins. 相似文献

Establishment and persistence of Escherichia coli in the intestine of rainbow trout. Oncorhynchus mykiss (Walbaum), was investigated. Infection was achieved by ingestion of contaminated feed but not by bath exposure. At 15°C. E. coli was found to increase in number in the intestine offish after an initial decline, and could still be detected after 4 days. At 6°C it was detected for 2 days but the numbers declined steadily. A similar trend was observed when extracted gut content was inoculated with E. coli in vitro; after an initial decline, bacterial growth recovered. Gut content in vitro also was a suitable environment for plasmid-mediated transfer of resistance. Oxytetracycline resistance was transferred successfully from Aeromonas salmoniclda to E. coli. This could not be reproduced in vivo, perhaps because the inoculum of A. salmoniclda achieved per fish was not high enough as it did not withstand drying well on the feed pellets. Introduction by gavage was also tried but failed to establish an A. salmonicida infection. These results show that at temperatures around 15°C, the presence of E. coli in fish need not be an indicator of recent passage through polluted waters. It may be a consequence of infection established many days before, and perhaps some distance away. Furthermore, although transfer of antibiotic resistance from a fish pathogen did take place in the environment of fish intestinal content, the likelihood of this being a high-risk route for acquisition of resistance by bacteria that can affect humans is low. 相似文献
