Sporozoites of Cryptosporidium parvum were incubated in 1:10 dilutions of immune or non-immune, heat-inactivated lamb serum specimens or serum fractions. The infectivity of treated sporozoites was assessed by inoculating them, per rectum, into five-day-old rats followed by histological examination of their intestines at either three or five days after infection. The infectivity of sporozoites treated with heat-inactivated whole sera was greatly reduced. This neutralisation had both specific and non-specific components. The former was associated with the IgG fraction of hyperimmune serum raised against sporozoites and the latter with a heat-stable, non-dialysable component present in both IgG-depleted hyperimmune serum and uninfected, gnotobiotic serum. 相似文献

The purpose of this study was to assess agreement between a wet reagent and a dry reagent analyzer. Thirteen analytes (albumin, globulin, alkaline phosphatase, alanine aminotransferase, amylase, urea nitrogen, calcium, cholesterol, creatinine, glucose, potassium, total bilirubin, and total protein) for both canine and feline serum were evaluated. Concordance correlations, linear regression, and plots of difference against mean were used to analyze the data. Concordance correlations were excellent for 8 of 13 analytes (r > or = 0.90); the correlations for albumin, potassium, and calcium were clinically unreliable. The linear regression analysis revealed that several analytes had slopes significantly different from unity, which was likely related to methodological differences. Compared to the wet reagent analyzer, the dry reagent analyzer showed excellent agreement for alkaline phosphatase, alanine aminotransferase, amylase (feline), urea nitrogen, cholesterol, creatinine, glucose, total bilirubin (canine), and total protein. Climate change is predicted to become a major threat to biodiversity in the 21st century, but accurate predictions and effective solutions have proved difficult to formulate. Alarming predictions have come from a rather narrow methodological base, but a new, integrated science of climate-change biodiversity assessment is emerging, based on multiple sources and approaches. Drawing on evidence from paleoecological observations, recent phenological and microevolutionary responses, experiments, and computational models, we review the insights that different approaches bring to anticipating and managing the biodiversity consequences of climate change, including the extent of species' natural resilience. We introduce a framework that uses information from different sources to identify vulnerability and to support the design of conservation responses. Although much of the information reviewed is on species, our framework and conclusions are also applicable to ecosystems, habitats, ecological communities, and genetic diversity, whether terrestrial, marine, or fresh water. 相似文献

- 1. Fisheries bycatch affects many species of marine mammals, seabirds, turtles and other marine animals.
- 2. New Zealand's endemic Hector's dolphins overlap with gillnet and trawl fisheries throughout their geographic range. The species is listed as Endangered by the IUCN. In addition, the North Island subspecies has been listed as Critically Endangered.
- 3. Estimates of catch rates in commercial gillnets from an observer programme (there are no quantitative estimates of bycatch by amateur gillnetters or in trawl fisheries) were used in a simple population viability analysis to predict the impact of this fishery under three scenarios: Option (A) status‐quo management, (B) new regulations announced by the Minister of Fisheries in 2008 and (C) total protection.
- 4. Uncertainty in estimates of population size and growth rate, number of dolphins caught and other model inputs are explicitly included in the analysis. Sensitivity analyses are carried out to examine the effect of variation in catch rate and the extent to which fishing effort is removed from protected areas but displaced to unprotected areas.
- 5. These methods are applicable to many other situations in which animals are removed from populations, whether deliberately (e.g. fishing) or not (e.g. bycatch).
- 6. The current Hector's dolphin population is clearly depleted, at an estimated 27% of the 1970 population. Population projections to 2050 under Options A and B predict that the total population is likely to continue declining. In the case of Option B this is driven mainly by continuing bycatch due to the much weaker protection measures on the South Island west coast.

The seroprevalence and seasonal trend of antibody titers against equine monocytic ehrlichiosis (Potomac horse fever) were determined in apparently healthy horses in selected areas of Illinois in 1986. Sera from 1,367 horses (6 months to 29 years old) were evaluated for the presence of antibodies against Ehrlichia risticii with indirect immunofluorescence. The majority (88%) of the horses were Thoroughbred or Standardbred racehorses. The number of horses with antibodies against E risticii was 229/1,367 (16.75%). The titers in these horses ranged from 1:10 to 1:640. As the year progressed, the number of seropositive horses (titers greater than or equal to 1:10) and the magnitude of the titers increased significantly, both reaching a maximum in July and August, respectively (P less than 0.05). A relationship between seropositivity and gender was not detected. In the year prior to sampling, 56.8% of the seropositive horses had not been ill, whereas 0.8% had diarrhea, an episode of acute abdominal pain, or laminitis. It was concluded that a large number of horses in Illinois are exposed to E risticii, that maximal exposure occurs in July, and that the most common form of the disease in Illinois is not associated with clinical signs. 相似文献

A visual decay assessment of Pinus radiata wood, which was part of a framing timber in a house in the North Island of New Zealand, indicated the presence of surface
decay. Microscopic observations, employing confocal laser scanning microscopy (CLSM), scanning electron microscopy (SEM) and
transmission electron microscopy (TEM), provided evidence of decay by cavity-forming soft rot (SR) fungi. A comparison of
ray tracheids (RT) with axial tracheids (AT) indicated that RT were considerably more resistant to SR than AT. In the heavily
degraded regions of wood, where axial tracheid walls contained abundant SR cavities, the walls of RT contained only a few
or no cavities. An assessment of lignin concentration in the cell walls by a combination of TEM, confocal fluorescence and
UV microscopy provided evidence of greater lignin concentration in the secondary wall of RT as compared to AT, which may explain
the observed greater resistance of RT to soft rot.
