When isolated adult oligodendrocytes adhere to a substratum myelinogenesis occurs. Investigation of the mechanism by which this happens indicated that the oligodendrocyte-substratum interaction activated protein kinase C-dependent phosphorylation of myelin basic protein and promoted the synthesis of myelin basic protein. In addition, when agents that activate protein kinase C (second messenger diacylglycerol or a tumor-promoting phorbol ester) were added to nonattached oligodendrocytes, they mimicked the influence of the substratum by inducing phosphorylation of myelin basic protein; and reagents that increase cellular adenosine 3', 5'-monophosphate (cyclic AMP) inhibited phosphorylation of myelin basic protein. Thus, at least in vitro, the interaction between oligodendrocytes and the substratum may mediate myelinogenic events, and phosphorylation of myelin basic protein may be an early requirement in the sequence of steps that ultimately results in myelin formation.  相似文献

Ammonium sulphate is widely used as a sulphur (S) fertilizer, constituting about 50% of global S use. Within nitrogen (N) management, it is well known that ammonium-based fertilizers are subject to ammonia (NH3) volatilization in soils with pH > 7, but this has been overlooked in decision making on S fertilization. We reviewed 41 publications reporting measurements of NH3 loss from ammonium sulphate in 16 countries covering a wide range of soil types and climates. In field experiments, loss was mostly <5% of applied N in soils with pH (in water) <7.0. In soils with pH > 7.0, there was a wide range of losses (0%–66%), with many in the 20%–40% range and some indication of increased loss (ca. 5%–15%) in soils with pH 6.5–7.0. We estimate that replacing ammonium sulphate with a different form of S for arable crops could decrease NH3 emissions from this source by 90%, even taking account of likely emissions from alternative fertilizers to replace the N, but chosen for low NH3 emission. For every kt of ammonium sulphate replaced on soils of pH > 7.0 in temperate regions, NH3 emission would decrease from 35.7 to 3.6 t NH3. Other readily available sources of S include single superphosphate, potassium sulphate, magnesium sulphate, calcium sulphate dihydrate (gypsum), and polyhalite (Polysulphate). In view of the large areas of high pH soils globally, this change of S fertilizer selection would make a significant contribution to decreasing NH3 emissions worldwide, contributing to necessary cuts to meet agreed ceilings under the Gothenburg Convention.  相似文献

ABSTRACT

We provide evidence that a factor contributing to the specific patterns of deforestation in Côte d’Ivoire is the set of spatial units of management used to structure wood extraction. Timber harvests were organized through a system of harvest perimeters, called “chantier,” for nearly a century. Chantiers were initiated as managerial instruments. However, their use as spatial tool for profit maximization led to positional anarchy. The lack of support for sustainable harvests from the legislation, the restoration system, and the tree-based policy governing the use of their content made the chantier the basis of a one-way resources extraction. Managerial scales facilitated clearing of forests. None served to regenerate forests. Using historical and official documents, satellite and cartographic data, we attempted to demonstrate how managerial scales aiming to organize logging have contributed to deforestation. We discuss the phases of fragmentation in the rainforest, the managerial scales, and the environmental context in which they were applied. We summarize the legislation that governed the managerial scales and how these scales organized space but the tree species-based policies and activities resulted in the forest’s dilapidation. The final section addresses the use of these scales in the creation of data for policy making.  相似文献

CASE HISTORY: A 400-cow dairy unit in the Waikato suffered a severe outbreak of facial eczema (FE) despite consistent zinc supplementation and significantly elevated serum zinc concentrations.

CLINICAL FINDINGS: FE prevention had begun in mid-January 2006, via zinc sulphate supplementation in the water. Photosensitisation was reported on 06 April, contemporaneous with a marked increase in the number of Pithomyces chartarum spores in pasture samples. Within 10 days of this first clinical case, 100 affected cows had been dried off, eight of which subsequently died or were culled. Blood samples were collected from ten affected cows; all had serum zinc concentrations >17 µmol/L, and eight had gamma-glutamyl transferase (GGT) activities >200 U/ml, indicating that the photosensitisation was secondary to liver damage, i.e. probably FE. Further investigation identified that this herd had been receiving excess copper supplementation; 4/6 culled cows had liver copper concentrations within or above the marginal range for copper toxicity.

DIAGNOSIS: Severe FE despite zinc supplementation that increased serum zinc concentrations above recommended levels

CLINICAL RELEVANCE: As zinc supplementation significantly reduces apparent copper status, copper supplementation is often used to counteract this. Previous excess copper intake may reduce the efficacy of zinc in preventing FE, thus copper intake should be assessed prior to the start of zinc supplementation.  相似文献

BACKGROUND: The accuracy of predicting the survival of insecticide‐resistant aphids following the application of commonly used insecticides from the carbamate, the pyrethroid, a mix of the two or the neonicotinoid chemical classes was evaluated in a potato field in Scotland. Equal proportions of five genotypes of the peach‐potato aphid, Myzus persicae (Sulzer), with none, resistance to dimethyl‐carbamates, resistance to pyrethroids or combinations conferring resistance to both chemical classes were released into potato field plots. The insecticides were sprayed separately onto these plots, the aphid populations were analysed after 6–8 days and the process repeated. RESULTS: For each assessment after the three separate spray events, plots treated with the carbamate had 48, 147 and 28%, those treated with pyrethroid 53, 210 and 89%, those treated with carbamate/pyrethroid 28, 108 and 64% and those treated with neonicotinoid 43, 55 and 11% of the numbers of M. persicae by comparison with untreated controls. Only the proportions of surviving aphids from the genotype containing no insecticide resistance traits and the genotype containing elevated carboxylesterases matched ratios predicted from the selective advantage afforded by the resistance traits alone. Survival of aphids from the other three genotypes that carried 1–3 of the insecticide resistance traits differed from expectations in all cases, possibly owing to physiological differences, including their vulnerability to predators and hymenopterous parasitoids present at the site and/or their carrying unknown insecticide resistance mechanisms. CONCLUSION: Control strategies based on knowledge of the genetically determined insecticide resistance profile of an M. persicae population alone are insufficient. An important requirement for biologists conserving vulnerable species of wildlife and managing genetic problems associated with small population size is to evaluate existing evidence regarding what is known of a species’ recent population history. For endemic island species in particular, current genetic impoverishment could be due to either a recent population crash or a consequence of an evolutionary history of sustained isolation and small effective population size. Interpreting any given case can often be further complicated by incomplete or contradictory evidence from historical field surveys that might suggest a very different demographic history. Here, we use the case of the Seychelles kestrel (Falco araea), an island endemic previously listed as critically-endangered but now relatively common, to illustrate how genetic data from microsatellite genotypes of 100–150-year-old museum specimens reveals a recent and severe population crash since the 1940s to approximately eight individuals, before the population recovered. We re-interpret the historical population trajectory of the Seychelles kestrel in the light of the minimal intervention required for this species to recover. We examine different ecological explanations for the decline and apparently unassisted recovery of the Seychelles kestrel, review the evidence for similarly unaided recoveries elsewhere and discuss the implications of unaided population recoveries for future species conservation programmes. Demographic profiles from historical genetic signatures can provide highly informative evidence when evaluating past and future recovery efforts for endangered species.  相似文献

A middle-aged rhesus monkey with detailed clinical history exhibited progression of a macular abnormality with a variety of clinical drusen and pigment changes typical of the Cayo Santiago phenotype. Numerous frozen sections of one sample of the macular retina/retinal pigment epithelium (RPE)/choroid showed a single classical druse but extensive single and clustered lipid-filled RPE cells. The monkey exhibited functional outer retinal decline and an insignificant number of 'window defects' as found among signs of relatively benign human macular aging. The clinical and histologically defined results agree if lipid-filled RPE cells are included among the clinically apparent signs of drusen.  相似文献

Heterogeneous environments make it difficult to apply consistent selection pressure because often it is difficult to identify a single or a few superior genotypes across all sets of conditions. However, when the target system is characterized by heterogeneity of environmental stress, varieties developed in high-yielding conditions may fail to satisfy farmers’ needs. Although this type of system is often found in marginal environments of developing countries, heterogeneous environmental conditions are also a feature of organic and low-external-input systems in developed countries. To meet the needs of these systems, breeding programs must decentralize selection, and although decentralized selection can be done in formal breeding programs, it is more efficient to involve farmers in the selection and testing of early generation materials. Breeding within these target systems is challenging, both genetically and logistically, but can identify varieties that are adapted to farming systems in marginal environments or that use very few external inputs. A great deal has been published in recent years on the need for local adaptation and participatory plant breeding; this article reviews and synthesizes that literature.  相似文献

A 21‐year‐old New Forest pony presented for evaluation of lethargy and colic. Transcutaneous abdominal ultrasonography revealed a cavitary, thick walled, mass‐like lesion that appeared continuous with the small intestine. The thick walls of the mass‐like lesion were of heterogeneous echogenicity with hyperechoic foci extending from the hyperechoic luminal surface to within the wall and there was loss of normal wall layering. These findings were confirmed grossly at exploratory celiotomy and histopathological examination of affected tissues confirmed lymphoma. The ultrasound findings facilitated a preoperative presumptive diagnosis that allowed informed decision‐making and aided case management.  相似文献
