土壤硝酸盐试纸条 反射仪测定法存在问题及其解决方案

针对浸提方法影响硝酸盐试纸条-反射仪法结果的问题,选择陕西关中的土娄土和渭南的黑垆土为供试材料,通过室内对比试验,对土壤硝酸盐浸提剂、振荡与沉淀时间及水土质量比等因素进行优化。结果表明,浸提剂类型、振荡与静置时间均对试纸条-反射仪法的土壤硝酸盐测试结果产生较大影响,而水土质量比对测定结果无显著影响,最终确定利用饱和硫酸钙作为浸提剂、振荡10 min、静置5 min为最佳前处理方法,在此条件下,硝酸盐试纸条-反射仪法测试结果准确,测试效率高。     

GBP5 promotes NLRP3 inflammasome assembly and immunity in mammals

Inflammasomes are sensory complexes that alert the immune system to the presence of infection or tissue damage. These complexes assemble NLR (nucleotide binding and oligomerization, leucine-rich repeat) or ALR (absent in melanoma 2-like receptor) proteins to activate caspase-1 cleavage and interleukin (IL)-1β/IL-18 secretion. Here, we identified a non-NLR/ALR human protein that stimulates inflammasome assembly: guanylate binding protein 5 (GBP5). GBP5 promoted selective NLRP3 inflammasome responses to pathogenic bacteria and soluble but not crystalline inflammasome priming agents. Generation of Gbp5(-/-) mice revealed pronounced caspase-1 and IL-1β/IL-18 cleavage defects in vitro and impaired host defense and Nlrp3-dependent inflammatory responses in vivo. Thus, GBP5 serves as a unique rheostat for NLRP3 inflammasome activation and extends our understanding of the inflammasome complex beyond its core machinery.     

Direct detection of projectile relics from the end of the lunar basin-forming epoch

The lunar surface, a key proxy for the early Earth, contains relics of asteroids and comets that have pummeled terrestrial planetary surfaces. Surviving fragments of projectiles in the lunar regolith provide a direct measure of the types and thus the sources of exogenous material delivered to the Earth-Moon system. In ancient [>3.4 billion years ago (Ga)] regolith breccias from the Apollo 16 landing site, we located mineral and lithologic relics of magnesian chondrules from chondritic impactors. These ancient impactor fragments are not nearly as diverse as those found in younger (3.4 Ga to today) regolith breccias and soils from the Moon or that presently fall as meteorites to Earth. This suggests that primitive chondritic asteroids, originating from a similar source region, were common Earth-Moon-crossing impactors during the latter stages of the basin-forming epoch.     

The occurrence and distribution of Salmonella enteritidis and other serovars on California egg laying premises: a comparison of two sampling methods and two culturing techniques

Between the summer of 1998 and the winter of 2000, Salmonella analysis was performed on 2128 single and 532 pooled manure drag swabs obtained from 133 California commercial egg laying farms. The isolation of Salmonella from all rows and from all flocks using single or pooled swabs was 80% and 92%, respectively. Hence, there was no statistical difference between single vs. pooled swabs in terms of identifying Salmonella on a row or flock basis. A total of 14 serogroups comprising 44 serotypes were isolated from 123 of 133 farms. When the top 10 serotypes were considered, there was no significant difference in the range of serotypes isolated by the two culturing methods. The overall S. enteritidis prevalence for California flocks was 10.5% (14/133). The overall row prevalence for S. enteritidis for all the farms was 1.1% (24/2128), and the overall pool prevalence was 2.4% (13/532). Sixty percent (12/20) of the S. enteritidis isolates from the positive farms were phage type 4, and 40% (8/20) represented five other phage types (1, 6B, 7, 8, and 28).     

Flow cytometric evaluation of canine bone marrow differential cell counts

Abstract: Three flow cytometric techniques were evaluated for determination of differential cell counts on canine clinical bone marrow specimens. Techniques included staining bone marrow specimens with 2'7'-dichlo-rofluorescein (DCF) or 3,3'-dihexyloxacarbocyanine iodide (DiOC6) and evaluation of forward-angle light scatter vs. side-angle light scatter plots. Flow cytometric evaluation of bone marrow cells stained with DCF failed to separate bone marrow cells into distinct cell populations. Staining with DiOC6 resulted in separation of bone marrow cells into populations of mature and immature erythroid cells, mature and immature myeloid cells, and lymphocytes. The scatter plot method resulted in identification of mature and immature erythroid cells, immature myeloid cells, metamyelocytes, and bands and segmenters. Lymphocytes could not be differentiated from mature erythroid cells by the scatter plot method. When the results of the DiOC6 method and the scatter plot method were compared with manual bone marrow differential cell counts, the scatter plot method had more similar mean values and higher correlation coefficients. The scatter plot method has the potential of providing rapid semiquantitative assessment of bone marrow differential cell counts in dogs for specimens that contain low numbers of lymphocytes.     

Effects of strain and different skip-a-day feed restriction periods on the growth performance of broiler chickens

The effects of 6, 10 and 14 days of skip-a-day feed removal during the starter and grower periods on the growth performance of Ross 308 and Hubbard broiler chickens were studied in a 49-day production period. Ross 308 was superior to Hubbard in weight gain, market weight and feed utilization but consumed more feed, deposited more abdominal fat and had a higher mortality rate. Overall, feed removal improved feed efficiency, had insignificant effects on abdominal fatness and mortality rate and reduced feed intake, weight gain and market weight. On each restricted-feeding regime, the birds gained as much weight as their fully fed counterparts during the period of resumed full feeding but were unable to compensate fully for the weight loss and were lighter at the end of the trial. It is suggested that for profitable broiler production under tropical conditions, Ross 308 and skip-a-day feed removal for less than 6 days from 7 to 17 days of age be considered.     

THE USE OF INTRAPERITONEAL FLUID AS A SIMPLE TECHNIQUE FOR ENHANCING SONOGRAPHIC VISUALIZATION OF THE CANINE PANCREAS

Sonographic evaluation of the pancreas was performed in ten healthy research dogs following intraperitoneal fluid administration. Physiological saline solution was aseptically introduced into the peritoneal cavity of each dog to create an acoustic window that would facilitate sonographic delineation of normal pancreatic tissues. Abdominal ultrasound examinations were performed in all dogs before and after the production of the diagnostic hydroperitoneum. Sonograms of the pancreas and other abdominal organs were scored on the basis of image clarity by experienced and novice ultrasonographers. Statistical analysis was then used to compare the effectiveness of each preparation method. A significant difference (P < 0.05) was established between the scores from the diagnostic hydroperitoneum sonograms and the baseline scans performed without intraperitoneal fluid placement. Sonographic visualization scores for the pancreas and other abdominal organs were increased following production of the diagnostic hydroperitoneum. These results indicate that the technique of diagnostic hydroperitoneum will significantly improve ultrasonographic visualization of the pancreas, and may prove useful in the diagnosis of canine pancreatic disease.
