Dust plays a critical role in Earth's climate system and serves as a natural source of iron and other micronutrients to remote regions of the ocean. We have generated records of dust deposition over the past 500,000 years at three sites spanning the breadth of the equatorial Pacific Ocean. Equatorial Pacific dust fluxes are highly correlated with global ice volume and with dust fluxes to Antarctica, which suggests that dust generation in interhemispheric source regions exhibited a common response to climate change over late-Pleistocene glacial cycles. Our results provide quantitative constraints on the variability of aeolian iron supply to the equatorial Pacific Ocean and, more generally, on the potential contribution of dust to past climate change and to related changes in biogeochemical cycles. 相似文献

New technologies based on microarray methods have begun to feature widely in carbohydrate chemistry and biology. These ‘glycoarray’ techniques, which in a number of cases emulate what has been achieved with DNA microarrays, allow for high throughput, quantitative analysis of protein–carbohydrate interactions. Lectin, antibody and enzyme specificity have been evaluated with these new techniques, which also extend to the detection of viruses and bacteria, and serodiagnosis of infection. In the plant field, high throughput mapping of cell wall carbohydrate structures has been reported, giving information not only on the localisation of given glycans within a plant, but also allowing systematic comparison between mutants and species. This review outlines some of the basic principles of ‘glycoarrays’ and illustrates recent reports of their development and application. 相似文献

Uncertainties in soil nitrogen (N) supply and crop N demand present a challenge to farmers deciding on N fertilizer rates. While field studies have documented the improvements in N use efficiency possible with site-specific N management approaches that address these uncertainties, a general understanding of the importance of uncertainty across a wide range of cropping systems is yet to emerge. Here a general model of N rate decision-making is presented which computes the optimal N rate that maximizes expected profit given uncertainties in N supply and demand. The cost of uncertainty is measured as the difference in N rate when soil N supply and crop N demand are unknown versus known perfectly. Eliminating uncertainty in soil N supply (but not crop demand) would reduce average N rates by 5–15% in typical irrigated rice systems, 10–30% in wheat, and 20–40% in maize. Perfect knowledge of potential crop N demand (but not soil supply) would reduce rates by 3–10% in all systems. Simultaneous knowledge of both factors reduced N rates by significantly more than the sum of their individual effects, reflecting important interactions between supply and demand uncertainties. This indicates that the value of information for one factor is inversely related to the level of uncertainty in the other. Studies that separately consider information on soil N supply or crop N demand may therefore underestimate the total benefit of management approaches that address both these uncertainties. Site-specific N management could lead to substantial reductions of N rates without yield loss in a wide range of cropping systems, thereby improving profitability and environmental quality. 相似文献

In previous aqueous enzymatic oil extraction (AEOE) experiments we reported a best free oil yield of 49% of the hexane extracted yield of dry fractionated corn germ. In the current experiments, a dispersion of 10% cooked, dry-fractionated germ in water was treated with α-amylase, glucoamylase and a cellulase complex. Free oil was collected by centrifuging a foam fraction of the dispersion. Several dispersion treatments were tried to evaluate their release of free oil and effect on the production of foam. The foam contained up to 8% free oil (dispersed from germ containing 26% oil) and fines oil (not centrifugally separable), protein and germ particles. Treatment with α-amylase and glucoamylase prior to treatment with the commercial cellulase used in previous AEOE studies increased the free oil yields about 25-61% of the hexane-extractable yield. A preliminary cost analysis indicates that oil separation with the amylase enhanced AEOE appears to be preferable to AEOE alone and profitable if crude corn oil cost exceeds $1.1/kg ($0.50/lb). 相似文献

The antiphospholipid syndrome is an autoimmune disease characterised by recurrent arterial or venous thrombosis, pregnancy morbidity and the persistence of positive antiphospholipid antibodies. Many other clinical manifestations may occur including heart valve disease, livedo reticularis, thrombocytopenia and neurological manifestations such as migraine and seizures. Flours from wheat varieties of differing bread-making quality were fractionated using a sequential salt precipitation technique. The gluten fractions in the different varieties varied in the proportion of HMW, LMW glutenins and gliadins. Their rheological behaviour was examined using constant strain (2%) small deformation oscillation tests over frequencies ranging from 0.005 to 10 Hz, before and after heating at 90 °C. The fractions containing a higher proportion of HMW glutenins were associated with a predominantly elastic character, whereas fractions containing mostly gliadins exhibited a viscous-like behaviour. The frequency dependent rheological behaviour of fractions containing HMW proteins was less susceptible to heat, and their elastic character was maintained after heating, whereas the rheology of intermediate fractions and fractions containing mostly gliadins was more susceptible to heating, indicating a rapid change from viscous to elastic behaviour after heating. 相似文献

The effects of honey bee management, such as intensive migratory beekeeping, are part of the ongoing debate concerning causes of colony health problems. Even though comparisons of disease and pathogen loads among differently managed colonies indicate some effects, the direct impact of migratory practices on honey bee pathogens is poorly understood. To test long- and short-term impacts of managed migration on pathogen loads and immunity, experimental honey bee colonies were maintained with or without migratory movement. Individuals that experienced migration as juveniles (e.g., larval and pupal development), as adults, or both were compared to control colonies that remained stationary and therefore did not experience migratory relocation. Samples at different ages and life-history stages (hive bees or foragers), taken at the beginning and end of the active season, were analyzed for pathogen loads and physiological markers of health. Bees exposed to migratory management during adulthood had increased levels of the AKI virus complex (Acute bee paralysis, Kashmir bee, and Israeli acute bee paralysis viruses) and decreased levels of antiviral gene expression (dicer-like). However, those in stationary management as adults had elevated gut parasites (i.e. trypanosomes). Effects of environment during juvenile development were more complex and interacted with life-history stage and season. Age at collection, life-history stage, and season all influenced numerous factors from viral load to immune gene expression. Although the factors that we examined are not independent, the results illuminate potential factors in both migratory and nonmigratory beekeeping that are likely to contribute to colony stress, and also indicate potential mitigation measures. 相似文献

Science, technology, engineering, and mathematics (STEM) graduate students are often encouraged to maximize their engagement with supervised research and minimize teaching obligations. However, the process of teaching students engaged in inquiry provides practice in the application of important research skills. Using a performance rubric, we compared the quality of methodological skills demonstrated in written research proposals for two groups of early career graduate students (those with both teaching and research responsibilities and those with only research responsibilities) at the beginning and end of an academic year. After statistically controlling for preexisting differences between groups, students who both taught and conducted research demonstrate significantly greater improvement in their abilities to generate testable hypotheses and design valid experiments. Cytotoxic T lymphocyte antigen 4 (CTLA-4) is an essential negative regulator of T cell immune responses whose mechanism of action is the subject of debate. CTLA-4 shares two ligands (CD80 and CD86) with a stimulatory receptor, CD28. Here, we show that CTLA-4 can capture its ligands from opposing cells by a process of trans-endocytosis. After removal, these costimulatory ligands are degraded inside CTLA-4-expressing cells, resulting in impaired costimulation via CD28. Acquisition of CD86 from antigen-presenting cells is stimulated by T cell receptor engagement and observed in vitro and in vivo. These data reveal a mechanism of immune regulation in which CTLA-4 acts as an effector molecule to inhibit CD28 costimulation by the cell-extrinsic depletion of ligands, accounting for many of the known features of the CD28-CTLA-4 system. 相似文献
