The Rickettsiales Ehrlichia ruminantium (ER) is the causative agent of heartwater, a fatal tick-borne disease of livestock in sub-Saharan Africa and in the Caribbean, posing strong economical constraints to livestock production. In an attempt to identify the most prominent proteins expressed by this bacterium, especially those encoded by the major antigenic protein 1 (map1) multigene family, a proteome map of ER cultivated in endothelial cells was constructed by using two dimensional gel electrophoresis combined with mass spectrometry. Among the sixty-four spots detected, we could identify only four proteins from the MAP1-family; the other proteins detected were mainly related to energy, amino acid and general metabolism (26%), to protein turnover, chaperones and survival (21%) and to information processes (14%) or classified as hypothetical proteins (23%). Additional studies on MAP1-family protein using immunochemical labeling also revealed that these proteins are differentially expressed along the bacterium life cycle, presenting different structural organization. Interestingly, when infectious elementary bodies (EBs) are released from host cells, MAP1 appears to be organized in SDS and heat-resistant dimers and trimers stabilized by disulfide bridges. Overall, the results presented herein not only reveal the first partial proteome map of ER but provide new insights on the expression ER MAP1-family proteins in host endothelial cells.  相似文献

The objective of this study was to evaluate the effect of a single joint lavage with 7.2% or 15% hypertonic saline solutions (HSS) on the tarsocrural joints of healthy calves. The tarsi of 10 calves were randomly lavaged with 7.2% HSS, 15% HSS, or isotonic saline. Synovial fluid samples were collected aseptically on days 1 (before joint lavage), 2, 3, 4, and 8 for complete cytological analysis. Lameness, joint swelling, and pain were recorded daily. Calves were euthanized on day 8 for gross and histological analyses of synovial membranes and articular cartilage. Synovitis was evaluated using a scoring system reflecting inflammatory changes in synovial membranes.Joints irrigated with HSS were more distended and painful compared with isotonic control joints. Swelling decreased consistently in the joints lavaged with 7.2% HSS, whereas it remained unchanged in joints lavaged with 15% HSS. Slight to moderate lameness was observed in the joints lavaged with 15% HSS. In comparison to isotonic saline joints, total protein concentration was significantly increased on day 2 and 3 for the joints lavaged with 7.2% HSS (P ≤ 0.01) and on days 2, 3, and 4 in the joints lavaged with 15% HSS (P ≤ 0.0006). Gross and histological findings revealed that synovitis was more severe in the joints lavaged with 15% HSS but variable in the joints lavaged with 7.2% HSS. No significant differences were observed for the articular cartilage.Fifteen percent HSS is not recommended for joint lavage. Although irrigation with 7.2% HSS may induce a variable synovitis, it was found appropriate for joint lavage. Its effects on septic joints remain undetermined.  相似文献

  • ? The effect of two weeds (grasses and broom, Cytisus scoparius) competition on the growth and functional traits of European beech saplings (Fagus sylvatica) was investigated in an experimental plantation in the French Massif central.
  • ? We hypothesized that grasses would have a much more harmful effect than broom on beech growth through strong competition for soil water and nitrogen.
  • ? A randomized block design was used with three separate blocks, each possessing three types of vegetation; grasses (mainly Agrostis capillaris, Holcus mollis and H. lanatus), broom, and bare soil as a control.
  • ? Two years after plantation beech sapling growth was significantly greater in bare soil than with competition from grasses, and was intermediate on broom. Growth was positively correlated with beech leaf nitrogen content (Nm) and xylem water potential (Ψx). N and Ψx were dependent, with nitrogen uptake probably decreasing with increasing drought. There was no clear correlation between growth and light transmitted 10 cm below the apex of the saplings.

    ? Introduction

    In the context of global environmental changes, better understanding of tree response to soil drought in young mixed species stands is needed to anticipate forest adaptation and management practices for the future.

    ? Materials and methods

    We investigated the functional response of five co-occurring broad-leaved tree species growing in a 15- to 25-year-old mixed stand in northeastern France during the 2006 summer drought. We measured functional traits related to water acquisition (phenology, rooting pattern and vulnerability of xylem to cavitation) and the ecophysiological response (sap flux density, leaf water potential) of these species to soil water shortage.

    ? Results

    Our study highlights contrasted drought response strategies among these species and a trade-off between leaf phenology, resistance of xylem to cavitation and root system depth.

    ? Conclusion

    At this site, a deep root system seemed to be a key functional trait for the species to cope with drought. Quercus robur and Salix capreae can be characterised as drought-avoidance species as they possess a deep root system and therefore did not strongly experience soil drought. Despite deep rooting capacity, Betula pendula did not really avoid soil drought and strongly regulated transpiration during dry periods. Nevertheless, the earliness of budburst of this species contributes to high annual growth rate. In contrast, Carpinus betulus and Fagus sylvatica both displayed typical characteristics of drought-sensitive species.  相似文献

    Thin zeolite films are attractive for a wide range of applications, including molecular sieve membranes, catalytic membrane reactors, permeation barriers, and low-dielectric-constant materials. Synthesis of thin zeolite films using high-aspect-ratio zeolite nanosheets is desirable because of the packing and processing advantages of the nanosheets over isotropic zeolite nanoparticles. Attempts to obtain a dispersed suspension of zeolite nanosheets via exfoliation of their lamellar precursors have been hampered because of their structure deterioration and morphological damage (fragmentation, curling, and aggregation). We demonstrated the synthesis and structure determination of highly crystalline nanosheets of zeolite frameworks MWW and MFI. The purity and morphological integrity of these nanosheets allow them to pack well on porous supports, facilitating the fabrication of molecular sieve membranes.  相似文献

    High-resolution spectra of the CN B2 summation operator +-X2 summation operator + (0,0) band at 390 nanometers yield isotopic ratios for comets C/1995 O1 (Hale-Bopp) and C/2000 WM1 (LINEAR) as follows: 165 +/- 40 and 115 +/- 20 for 12C/13C, 140 +/- 35 and 140 +/- 30 for 14N/15N. Our N isotopic measurements are lower than the terrestrial 14N/15N = 272 and the ratio for Hale-Bopp from measurements of HCN, the presumed parent species of CN. This isotopic anomaly suggests the existence of other parent(s) of CN, with an even lower N isotopic ratio. Organic compounds like those found in interplanetary dust particles are good candidates.  相似文献
