The objective was to examine the effect of the additives acetic acid, lactic acid, sodium bicarbonate, sodium chloride, and sodium hydroxide on the hydrolysis of whey protein isolate with subcritical water. A screening experimental design was used to study the effect of temperature, time, and additives. The most influential additive, sodium bicarbonate, along with temperature and time was used in a second experimental design to predict the treatment conditions to maximize the degree of hydrolysis and production of free amino acids. The maximum degree of hydrolysis achieved was 50% at a concentration of 1.24 M sodium bicarbonate, 291 °C, and 28 min. The highest concentration of total amino acids was 83.0 mg/g of whey protein isolate with 0.83 M sodium bicarbonate at 264 °C for 29 min. Compared to water alone, sodium bicarbonate increased the degree of hydrolysis 4-fold and the production of amino acids by 44% and decreased peptides' molecular weight. 相似文献

In this study, the hypothesis that food-derived opioid peptides besides β-casomorphin 7 might modulate the production of mucin via a direct action on epithelial goblet cells was investigated in HT29-MTX cells used as a model of human colonic epithelium. Seven milk whey or casein peptides, a human milk peptide, and a wheat gluten-derived peptide with proved or probable ability to bind μ- or δ-opioid receptors were tested on the cell culture. Significantly increased secretion of mucins was found after exposure to six of the assayed peptides, besides the previously described β-casomorphin 7, as measured by an enzyme-linked lectin assay (ELLA). Human β-casomorphin 5 and α-lactorphin were selected to study the expression of mucin 5AC gene (MUC5AC), the HT29-MTX major secreted mucin gene. α-Lactorphin showed increased expression of MUC5AC from 4 to 24 h (up to 1.6-fold over basal level expression), although differences were statistically different only after 24 h of exposure. However, this increased expression of MUC5AC did not reach significance after cell treatment with human β-casomorphin 5. In conclusion, six food-derived peptides have been identifed with described or probable opioid activity that induce mucin secretion in HT29-MTX cells. Plant growth–promoting rhizobacteria (PGPR) have been reported to stimulate the growth and yield of grain crops, particularly when nutrient supply is poor. However, the mechanisms underlying stimulation of plant growth may vary depending not only on growth conditions and crop management but also on plant and bacterial species. The present study assessed the effect of an inoculation with single or multiple PGPR strains on phosphorus (P)‐solubilization processes in the soil and on grain yield in wheat. Single inoculation with Bacillus subtilis OSU‐142, Bacillus megaterium M3, or Azospirillum brasilense Sp245 increased grain yield by 24%, 19%, and 19%, respectively, while a mixed inoculation with OSU‐142, M3, and Sp245 increased grain yield by 33% relative to noninoculated plants. Single inoculations with Paenibacillus polymyxa RC05 or Bacillus megaterium RC07 were less effective. Single or mixed treatments with OSU‐142, M3, and Sp245 increased the concentrations of the labile and moderately labile P fractions in rhizosphere soil. The growth‐stimulating effect of OSU‐142, M3, and Sp245 was also reflected by higher P concentrations in most plant organs. Among all inocula tested, the highest plant P acquisition was obtained in the presence of M3 and accompanied by the highest microbial P levels and the highest phosphatase activities in the rhizosphere soil. In conclusion, seed inoculation with mixed PGPR strains may effectively substitute for a part of P‐fertilizer application in extensive wheat production, and in particular M3 appears to improve the solubilization of inorganic soil P. 相似文献

We used polymerase chain reaction-denaturing gradient gel electrophoresis (PCR-DGGE) to compare bacterial community patterns
obtained with target DNA extracted from a soil by direct and indirect methods. For this purpose, two direct extraction methods,
i.e. cell lysis by bead beating and cell disruption by grinding in liquid N, and two indirect methods, i.e. cell extraction
followed by DNA extraction, and combined RNA/DNA extraction from the bacterial cell fraction, were performed. Crude extracts
were purified and amplified using universal bacterial primers. PCR products were then analysed by DGGE, and similarity between
the profiles obtained was determined by unweighted pair group with mathematical averages clustering. The results showed clear
profiles that presumably represented the dominant bacterial fractions in the samples. The profiles generated by all four methods
were similar, indicating that the methods were of approximately equal efficiency in the extraction of target DNA representative
of the soil bacterial community. However, the patterns of clustering also indicated that different populations of bacteria
could be detected in the same soil using different soil DNA extraction methods. The application of two dilution levels of
DNA in PCR-DGGE showed that the most stable profile of the soil bacterial community could be generated by the direct methods.
The indirect methods gave clustered profiles at both dilution levels. It is likely that these methods extracted DNA from a
major, easily desorbed, bacterial fraction, consisting of low-density populations. PCR-DGGE was found to be a suitable technique
with which to assess differences in methods for DNA extraction from soil, which can be further used for the determination
of microbial community diversity at the molecular level.

The purpose of this work was to determine the influence of vinification technique (maceration temperature and clarification method), storage temperature, and length of storage time on the phenolic compounds and color of young red wines. Multivariate analysis of variance and principal component analysis pointed to significant differences among all of the variables according to vinification technique and length of storage time. Storage temperature did not cause significant differences between some of the variables. The best color characteristics were obtained when low-temperature maceration wines were clarified with polyvinylpyrrolidone. Color quality also improved with lower storage temperature. 相似文献

The insecticide-nematicide carbofuran was incorporated in alginate-based granules to obtain controlled-release (CR) properties. The basic formulation [sodium alginate (1.61%)-carbofuran (0. 59%)-water] was modified by addition of sorbents. The effect on carbofuran release rate, caused by the incorporation of natural and acid-treated bentonite (0.5 and 1.0 M H(2)SO(4)) in alginate formulation, was studied by immersion of the granules in water under shaking. The time taken for 50% of the active ingredient to be released into water, t(50), was longer for those formulations containing natural bentonite (6.1 h) or acid-treated bentonite (9.0 and 11.7 h for 0.5 and 1.0 M H(2)SO(4) treatments, respectively) than for the preparation without bentonite (4.7 h). It appears from the results that the release of carbofuran from the various formulations is controlled by a diffusion mechanism according to the n values obtained, which were close to 0.5 in all cases. The mobility of carbofuran from alginate-based CR formulations was investigated by using soil columns packed with a clay soil (53% clay and 0.08% organic matter). Two alginate-based CR formulations containing natural bentonite or acid-treated bentonite (0.5 M H(2)SO(4)) were compared to technical grade carbofuran. The use of alginate-based CR formulations resulted in a reduction of the leached amount of carbofuran compared with the total amount of pesticide leached using the technical product (50 and 75% for CR granules containing natural and acid-treated bentonite, respectively). Alginate-bentonite CR formulations might be efficient systems for reducing carbofuran leaching in clay soils, which would reduce the risk of groundwater pollution. 相似文献

A red wine with appropriate basic quality characteristics for aging was stored in oak barrels for 12 months and then bottled and aged for a further 6 months. The same ambient conditions of temperature and humidity were maintained throughout the entire aging process. The barrels used were made from three different species of oak by four different cooperages and had been used for at least two years. Analysis of variance and principal component analysis were run on the values for hexoses, pentoses, total anthocyanins, ortho-diphenols, low- and high-polymer polyphenols, and color parameters to study the behavior of the monosaccharides and polyphenols in response to the factors of aging time, the oak variety employed, and the source cooperage where the barrels had been made. Time trends for all the phenolic components were directly related to aging time, with low-polymeric polyphenols (LPPs) being the most affected by wood type and source cooperage. Wine color was defined by a basic red color which decreased with aging time in the barrel and was altered by yellowish pigment components differing for each of the barrels in which oxidative aging took place and by increased stability of the blue copigments. Principal component analysis showed that samples of the same source wine aged in different barrels tended to be grouped together according to each of the aging intervals considered. 相似文献

Purified lutein diesters deposited on commercial nonporous glass beads were solubilized in supercritical CO(2) in a computerized batch extractor, and their solubilities were compared to their solubilities in hexane. Densities of 0.7, 0.8, and 0.9 g/mL were evaluated without modifiers. Both pressure and temperature increased solubility, although temperatures >50 degrees C promoted carotenoid loss as determined by mass balance. Solubility was enhanced by the use of modifiers and was related to their log P. Chloroform (log P = 2) increased 2.8 times the amount of solubilized lutein diesters compared to pure CO(2) at the same extraction conditions (0.9 g/mL and 40 degrees C) to yield 65% of the amount extracted with hexane. Supercritical CO(2) extraction of lutein diesters could represent a cleaner technology as compared to the current industrial use of hexane with important ecological and health-related implications. 相似文献

Peanut and its derivatives, especially peanut butter, are extensively consumed in many countries, mainly in the United States, which is also the major exporter of these products. trans-Resveratrol is present in peanuts, and recently this compound has been quantified in peanut butter. It is well-known that there are beneficial effects of trans-resveratrol and its glucoside, the piceid, in health. The absorption of trans-resveratrol has been proven in animals, and certain studies show that the absorption of some phenols is enhanced by conjugation with glucose, so that it could be possible that trans-piceid would be more absorbed than its aglycon (trans-resveratrol). In our work, we have identified the presence of trans-piceid in peanut butter with a new method to quantify trans-resveratrol and trans-piceid (3-beta-glucose of trans-resveratrol). This fact is very interesting because the glucosilated form could be more easily absorbed by the intestinal gut; in this way trans-piceid would exercise its beneficial function more efficiently than trans-resveratrol. To our knowledge, this is the first time that trans-piceid has been quantified in peanut butter. In order to evaluate the influence of different N rates on percent N derived from fertilizer (%Ndff) at different growth stages, on yields and the percent N use efficiency (%NUE) values of two winter wheat varieties (durum and bread), field experiments on fallow were carried out at four different locations in Central Anatolia, in the 1991–1992 and 1992–1993 growing seasons. At each site the rates of N (0, 40, 80 and 120?kg N/ha) were applied as ammonium sulphate [(NH4)2SO4] using a Latin Square experimental design with four replicates. The total amounts of N fertilizer were applied once after seedling emergence at all experimental sites. Labelled (15NH4)2SO4 fertilizer was applied to sub-plots from which %Ndff values were determined at tillering, booting, grain filling, and harvest stages. Yield sub-plots received unlabelled (NH4)2SO4 from which total dry matter (seed and straw) and N yields were determined. Also the %NUE values were calculated by the 15N and "difference" methods at the harvest stage. Stored soil moisture at 0–90?cm depth, evapotranspiration and water use efficiency values were calculated as well. The results obtained showed that (1) Gerek-79 variety used both the applied N fertilizer and the available soil moisture more efficiently, (2) the percent NUE values obtained overall were generally less than 20 for both varieties and (3) with the 15N method, less variable %NUE values were obtained in comparison to the difference method. 相似文献
