Paeoniflorin (1), isolated from the methanol extract of Paeonia lactiflora, showed a lowering effect on cholesterol level in the experimentally-induced hyperlipidemic rats. Paeoniflorin showed a significantly lowering effect of total cholesterol, LDL and triglyceride levels compared with the control group at the dose of 200 and 400 mg/kg, p.o. once a day for 4 weeks.  相似文献

The primary objective of this study was to investigate if responses of pottedPinus densiflora Sieb. et Zucc. seedlings to ozone exposure could be altered by water deficit stress applied before or during ozone exposure. One-year-old seedlings grown from seeds in pots were used. Water deficit preconditioning was done for ten weeks from May 1, 1998, followed by ozone exposure and simultaneous water deficit for eight weeks. Water deficit was controlled by monitoring xylem water potentials with a pressure chamber. Ozone was fumigated in open top chambers with an eight-hour mean concentration of 0.1 ppm. A 23 factorial design was employed. Dry weights, carbohydrate concentrations, and leaf gas exchanges were measured. In response to the water deficit, growth and stomatal conductance were reduced, while soluble carbohydrate concentrations were enhanced. Interactions between ozone and simultaneous water deficit were significant. Dry weights were significantly decreased by ozone exposure only in well-watered seedlings, suggesting that simultaneous water deficit may alleviate the adverse effects of ozone. This protection from ozone stress observed in water-stressed seedlings resulted from: (1) reduced ozone uptake due to stomatal closure and (2) enhanced TNC (Total Nonstructural Carbohydrates) which acted as a buffer against ozone injury.  相似文献

Differences in the compositions of volatiles from dried omija fruits (Schisandra chinensis Baillon) cultivated in different areas (Mungyeong, Jangsu, Jechon, and Hoengseong) in South Korea were determined by applying principal component analysis to gas chromatography-mass spectrometry data sets. Quantitative assessments revealed that terpene hydrocarbons, such as germacrene D, β-selinene, α-ylangene, β-elemene, α-selinene, and (E)-β-farnesene, were the main volatiles in all omija fruit samples. On the other hand, (E)-β-ocimene, calarene, (E)-β-farnesene, β-selinene, nonanal, 2-methylbutanoic acid, benzoic acid, 2,3-butanediol, and phenethyl alcohol were the major volatile components that contributed to the discrimination between omija fruit samples from the four cultivation areas. In addition, aroma-active compounds in four dried omija fruits were investigated and compared by gas chromatography-olfactometry using aroma extract dilution analysis. (E)-β-Ocimene (floral and herbaceous), α-pinene (pine-like and woody), hexanal (cut grass-like), 5-methylfurfural (burnt sugar-like and sweet), and α-terpinene (minty, green, and fresh) were important aroma-active compounds in all omija samples. Interestingly, the flavor dilution factors of most aroma-active compounds were lower for omija sample cultivated in Hoengseong than for those cultivated in Mungyeong, Jangsu, and Jechon.  相似文献

In this study, we investigated the effects of S-allyl-L-cysteine (SAC), a major sulfur-containing compound present in garlic, on Ki67- and doublecortin (DCX)-positive cells, which were used as a marker for cell proliferation and neuroblast differentiation, respectively, in the mouse dentate gyrus. SAC (300 mg/kg) was intraperitoneally administered to 8-week-old mice once a day for 3 weeks. The animals were then sacrificed at 11 weeks of age. SAC administration significantly increased Ki67-positive nuclei and DCX-immunoreactive neuroblasts in the subgranular zone of the dentate gyrus. Furthermore, serotonin 1A receptor (5-HT(1A)) levels in the hippocampus were also increased. These results suggest that SAC significantly increased cell proliferation and neuroblast differentiation by increasing 5-HT(1A) levels in the dentate gyrus.  相似文献

Hamacanthins, bis (indole) alkaloids, are found in a few marine sponges, including Spongosorites sp. Hamacanthins have been shown to possess cytotoxic, antibacterial and antifungal activities. However, the precise mechanism for the biological activities of hamacanthins has not yet been elucidated. In the present study, the anti-angiogenic effects of 6″-debromohamacanthin A (DBHA), an active component of isolated hamacanthins, were evaluated in cultured human umbilical vascular endothelial cells (HUVEC) and endothelial-like cells differentiated from mouse embryonic stem (mES) cells. DBHA significantly inhibited vascular endothelial growth factor (VEGF)-induced cell proliferation, migration and tube formation in the HUVEC. DBHA also suppressed the capillary-like structure formation and the expression of platelet endothelial cell adhesion molecule (PECAM), an endothelial biomarker, in mES cell-derived endothelial-like cells. To further understand the precise molecular mechanism of action, VEGF-mediated signaling pathways were analyzed in HUVEC cells and mES cell-derived endothelial-like cells. DBHA suppressed the VEGF-induced expression of MAPKs (p38, ERK and SAPK/JNK) and the PI3K/AKT/mTOR signaling pathway. In addition, DBHA inhibited microvessel sprouting in mES/EB-derived embryoid bodies. In an ex vivo model, DBHA also suppressed the microvessel sprouting of mouse aortic rings. The findings suggest for the first time that DBHA inhibits angiogenesis by targeting the vascular endothelial growth factor receptor 2 (VEGFR2)-mediated PI3K/AKT/mTOR signaling pathway in endothelial cells.  相似文献

Objective To evaluate the effect of ephedrine on intramuscular blood flow and hemodynamic parameters during equine anesthesia. Study design Prospective experimental study. Animals Six healthy adult Welsh Mountain ponies (five males, one female, mean weight: 267 kg, range: 213–347 kg). Methods Halothane‐anesthetized ponies received an IV bolus of ephedrine (0.1 mg kg?1), followed 30 minutes later by a second IV ephedrine injection (0.2 mg kg?1). Changes in intramuscular blood flows (IMBF) in upper and lower triceps brachii were measured by laser Doppler flowmetry. Cardiopulmonary measurements were made at intervals for 30 minutes following each injection. Results were compared with values from a control group, similarly anesthetized but given saline in an earlier study. Results Ephedrine at either dose increased heart rate, arterial blood pressure (AP), cardiac index (CI) and intramuscular blood flow (IMBF), the effects on these parameters being significant and long‐lasting following the higher dose. Systemic vascular resistance remained unchanged, and was significantly lower than in the control saline group. PaO2 decreased significantly immediately following the first injection of ephedrine, then remained unchanged for the remainder of the experiment. PaCO2 increased slowly throughout the anesthetic period. One pony developed supraventricular premature complexes following the second injection. No other side effects were seen. Conclusion Ephedrine at dose rates of 0.2 mg kg?1 IV consistently increased in CI, AP, and IMBF in both forelimbs. Clinical relevance Ephedrine may be of use to improve AP, CI and IMBF during halothane anesthesia, although the occurrence of an arrhythmia in one pony is of concern.  相似文献

In this study, we investigated the effects of chronic aluminum (Al) exposure for 10 weeks on cell proliferation and neuroblast differentiation in the hippocampus of type 2 diabetic rats. Six-week-old Zucker diabetic fatty (ZDF) and Zucker lean control (ZLC) rats were selected and randomly divided into Al- and non-Al-groups. Al was administered via drinking water for 10 weeks, after which the animals were sacrificed at 16 weeks of age. ZDF rats in both Al- and non-Al-groups showed increases in body weight and blood glucose levels compared to ZLC rats. Al exposure did not significantly affect body weight, blood glucose levels or pancreatic β-cells and morphology of the pancreas in either ZLC or ZDF rats. However, exposure to Al reduced cell proliferation and neuroblast differentiation in both ZLC and ZDF rats. Exposure to Al resulted in poor development of the dendritic processes of neuroblasts in both ZLC and ZDF rats. Furthermore, onset and continuation of diabetes reduced cell proliferation and neuroblast differentiation, and Al exposure amplified reduction of these parameters. These results suggest that Al exposure via drinking water aggravates the impairment in hippocampal neurogenesis that is typically observed in type 2 diabetic animals.  相似文献
