(-)-Epigallocatechin gallate (EGCG), a polyphenolic compound found in green tea, is a promising chemopreventive agent against cancer due to its strong antiproliferative effects on cancer cells; however, its possible toxicity and carcinogenicity must be investigated before EGCG can be used as a dietary supplement for chemoprevention. The inhibition of gap junctional intercellular communication (GJIC) is strongly associated with carcinogenesis, particularly the tumor promotion process; thus, we investigated the effects of EGCG on GJIC in WB-F344 normal rat liver epithelial (RLE) cells. EGCG, but not (-)-epicatechin (EC), another polyphenol found in green tea, inhibited GJIC in a dose-dependent and reversible manner in RLE cells. EGCG also induced the phosphorylation of connexin 43 (Cx43), a major regulator of GJIC. The phosphorylation of extracellular signal-regulated protein kinase 1/2 (ERK1/2) was also observed in EGCG-treated RLE cells. The inhibition of GJIC and phosphorylation of Cx43 and ERK1/2 by EGCG were completely blocked by U0126, a pharmacological inhibitor of mitogen-activated protein kinase/ERK kinase. EGCG generated a larger amount of hydrogen peroxide than EC in a dose-dependent manner. Furthermore, catalase partially inhibited the EGCG-induced inhibition of GJIC and the phosphorylation of Cx43 and ERK1/2. Potato microtubers were treated with rindite to investigate the effect on dormancy breaking. Postharvest application of rindite by fumigation with 2 ml rindite for 48 hr or 4ml for 24 hr significantly reduced the dormancy period of potato microtubers using a 32 x 15 x 17 cm tightly sealed plastic box. Approximately 2 weeks after the treatments microtubers of all cultivars, Atlantic, Superior, Lemhi Russet, Red Dale and Kennebec started to sprout. The efficiency of the treatments were the greatest for the cv. Lemhi Russet, intermediate for cv. Superior and least for cv. Red Dale. In all cultivars of potato microtubers, more decay was observed the earlier rindite treatment occurred after harvesting, therefore potato microtubers should be treated with rindite at least 2 weeks after harvest when the skin of microtubers is mature. The data indicates that the dormancy of potato microtubers with well-matured skin can be effectively broken with an optimum treatment of rindite. 相似文献

本实验于2004年在东北林业大学进行。用指甲油印记法在光学显微镜下观察了杜鹃花属6个种(牛皮杜鹃 Rh.aureaum,兴安杜鹃Rh. dauricum, 照白杜鹃Rh. micranthum,迎红杜鹃Rh. mucronulatum,苞叶杜鹃 Rh. Redowskianum 和大字杜鹃Rh. schlippenbachii)的表皮指纹特征。对叶片的形态特征包括叶片下表皮气孔类型、保卫细胞和副卫细胞的特征、叶片两面的普通细胞(形状和垂周壁特征)和表皮毛进行了较详细的描述。所研究的6种杜鹃花中,有3种气孔类型,其中兴安杜鹃、照白杜鹃、和迎红杜鹃为单细胞型气孔(Pericytic 型),苞叶杜鹃为不规则型气孔(Anomocytic 型),牛皮杜鹃和大字杜鹃是平轴式气孔(Diacytic 式)。不同种间,单细胞型和平轴式气孔的副卫细胞类型在形状和表面特征上有所不同。普通表皮细胞的表面观为从四边形、六边形, 多边形或不规则型, 垂周壁呈平直或弯曲状。兴安杜鹃、照白杜鹃和迎红杜鹃3种的叶片两面均有腺鳞。所有这些叶片特点表明上述6种杜鹃花的表皮指纹具有种的特性。图4表2参29。 相似文献

In this study we investigated the effects of ultrasonic wave treatment on the extraction yield of acid-soluble collagen from
sea bass skins. Two extraction methods were compared: a 24 h acid treatment using 0.5 M acetic acid (1:200 sample/acid, w/v)
and an extraction using ultrasonic treatment after the addition of a 0.5 M acetic acid solution. The results indicated that
the extraction yield of collagen increased with the ultrasonic treatment, with the extraction rate increasing rapidly at higher
amplitudes of ultrasonic treatment. The subunit compositions of the collagen extracted by ultrasonic treatment were analyzed
by sodium dodecyl sulfate-polyacrylamide gel electrophoresis, which revealed that the α1(α3), α2, and β chains of collagen
were present early in the ultrasonic treatment. An unknown component, believed to be a product of collagen degradation induced
by the ultrasonic treatment, was detected only after a longer treatment time. The component extracted by the ultrasonic treatment
was determined to be collagen based on the finding that there were no changes in the main components of collagen, specifically,

ABSTRACT: As part of our study of the isolation of antihypertensive agents derived from natural marine products, the bioactivity of 10 edible Korean seaweeds were screened by angiotensin converting enzyme (ACE) inhibitory and peroxynitrite assays. Among the crude extracts of selected seaweeds, including five Phaeophyta ( Ecklonia stolonifera , E. cava , Pelvetia siliquosa , Hizikia fusiforme , and Undaria pinnatifida ), four Rhodophyta ( Gigartina tenella , Gelidium amansii , Chondria crassicaulis , and Porphyra tenera ) and one Chlorophyta ( Capsosiphon fulvescens ), the ethanol extracts of E. stolonifera , E. cava , P. siliquosa , U. pinnatifida , and G. tenella exhibited significant inhibitory properties against ACE at more than 50% inhibition at a concentration of 163.93 µg/mL. Phloroglucinol 1 , eckstolonol 2 , eckol 3 , phlorofucofuroeckol A 4 , and dieckol 5 had been isolated previously, and triphlorethol-A 6 and fucosterol 7 were isolated for the first time from E. stolonifera. Also, the ACE inhibitory and peroxynitrite scavenging properties of phlorotannins 1–6 were evaluated, along with fucosterol 7 obtained from E. stolonifera . Among profound peroxynitrite scavenging compounds 1–6 , phlorotannins 3 , 4 and 5 were also determined to manifest marked inhibitory activity against ACE, with 50% inhibition concentration (IC50 ) values of 70.82 ± 0.25, 12.74 ± 0.15, and 34.25 ± 3.56 µM, respectively. 相似文献

Astaxanthin is a powerful biological antioxidant and is naturally generated in a great variety of living organisms. Some studies have demonstrated the neuroprotective effects of ATX against ischemic brain injury in experimental animals. However, it is still unknown whether astaxanthin displays neuroprotective effects against severe ischemic brain injury induced by longer (severe) transient ischemia in the forebrain. The purpose of this study was to evaluate the neuroprotective effects of astaxanthin and its antioxidant activity in the hippocampus of gerbils subjected to 15-min transient forebrain ischemia, which led to the massive loss (death) of pyramidal cells located in hippocampal cornu Ammonis 1-3 (CA1-3) subfields. Astaxanthin (100 mg/kg) was administered once daily for three days before the induction of transient ischemia. Treatment with astaxanthin significantly attenuated the ischemia-induced loss of pyramidal cells in CA1-3. In addition, treatment with astaxanthin significantly reduced ischemia-induced oxidative DNA damage and lipid peroxidation in CA1-3 pyramidal cells. Moreover, the expression of the antioxidant enzymes superoxide dismutase (SOD1 and SOD2) in CA1-3 pyramidal cells were gradually and significantly reduced after ischemia. However, in astaxanthin-treated gerbils, the expression of SOD1 and SOD2 was significantly high compared to in-vehicle-treated gerbils before and after ischemia induction. Collectively, these findings indicate that pretreatment with astaxanthin could attenuate severe ischemic brain injury induced by 15-min transient forebrain ischemia, which may be closely associated with the decrease in oxidative stress due to astaxanthin pretreatment. 相似文献

A series of 3-chloro-2-(4-chloro-2-fluorophenyl)-4,5,6,7-tetrahydro-2H-indazole derivatives containing various substituted isoxazolinylmethoxy groups at the 5-position of the benzene ring were synthesized and their herbicidal activities assessed under greenhouse and flooded paddy conditions. Among them, compounds having a phenyl or cyano substituent at the 3-position of the 5-methyl-isoxazolin-5-yl structure demonstrated good rice selectivity and potent herbicidal activity against annual weeds at 16-63 g AI ha(-1) under greenhouse conditions. Field trials indicated that these two compounds controlled a wide range of annual weeds rapidly with a good tolerance on transplanted rice seedlings by pre-emergence application. They showed a low mammalian and environmental toxicity in various toxicological tests. 相似文献

The effects of topical administration of polysaccharides isolated from fungus, Phellinus gilvus (PG) on the healing of rat dermal wounds were assessed. In 10 Sprague-Dawley (SD) rats, six 6 mm diameter defects were made with a punch biopsy appliance. After 24 hr, test substances were applied to the defects twice a day: 0.025, 0.25, and 2.5% polysaccharides from PG (PG0.025, 0.25, and 2.5 groups), Madecassol ointment (MC group), aqueous gel (AG group) and no treatment (control group). Six days postoperatively, the contraction and reepithelialization of the wound surface were assessed. Wound diameter was significantly reduced in all PG groups (P<0.05). Complete epithelialization and macrophages were noted in the PG0.25 group, as compared to the control group. We conclude that polysaccharides isolated from PG have significant dermal wound healing effects, and this investigation suggests the potential clinical application of PG as a wound healing agent. 相似文献
