Four ruminally and duodenally cannulated Suffolk wether lambs (34.5 +/- 2 kg initial BW) were used in a 4 x 4 Latin square designed experiment to compare effects of supplemental ruminally degradable protein (RDP) vs. increasing amounts of supplemental ruminally undegradable protein (RUP) on ruminal characteristics and site and extent of digestion in lambs. Lambs were fed a basal diet of crested wheatgrass hay (4.2% CP) for ad libitum consumption, plus 1 of 4 protein supplements: isolated soy protein (RDP source) fed to meet estimated RDP requirements assuming a microbial efficiency of 11% of TDN (CON) or corn gluten meal (RUP source) fed at 50, 100, or 150% of the supplemental N provided by CON (C50, C100, and C150, respectively). Neither NDF nor ADF intake was affected (P >/= 0.18) by protein degradability, but they increased or tended to increase (P = 0.07) with increasing level of RUP. Total OM and N intakes were similar (P >/= 0.26) for CON and C100, but increased (P = 0.01) as level of RUP increased. True ruminal OM and ruminal digestibilities of NDF and ADF were not affected (P >/= 0.33) by protein degradability. However, true ruminal N digestibility was greater (P = 0.03) for CON compared with C100. Ruminal ammonia concentrations were greater (P = 0.002) for CON compared with C100 lambs, and increased (P = 0.001) with increasing RUP. Microbial N flows were not affected (P >/= 0.12) by protein degradability or increasing RUP. Likewise, neither ruminal urease activity (P >/= 0.11) nor microbial efficiency (P >/= 0.50) were affected by protein degradability or level of RUP. Total tract OM, NDF, and ADF digestibility was greater (P = 0.05) for C100 compared with CON. Likewise, total tract N digestibility was greater (P = 0.03) for C100 than for CON, and increased linearly (P = 0.001) with increasing RUP. Lambs fed C100 consumed approximately 69% less supplemental RDP (31% less total RDP) than CON, but were able to maintain forage intake and digestion. This lack of response in forage intake would suggest that lambs supplemented with RUP were recycling sufficient N to compensate for an apparent RDP deficiency. Although ruminal degradability of protein has little effect on forage intake or ruminal digestion of nutrients, there is potential to enhance total tract digestion of nutrients by decreasing the ruminal degradability of supplemental protein. 相似文献

The importance of communication skills in veterinary medicine is increasingly recognised. Appropriate communication skills towards the client are of utmost importance in both companion animal practice and production animal field and consultancy work. The need for building a relationship with the client, alongside developing a structure for the consultation is widely recognised and applies to both types of veterinary practice.Results
Veterinary advisory practice in production animal medicine is, however, characterised by a more complex communication on different levels. While the person-orientated communication is a permanent process between veterinarian and client with a rather personal perspective and defines the roles of interaction, the problem-orientated communication deals with emerging difficulties; the objective is to solve an acute health problem. The solution - orientated communication is a form of communication in which both veterinarian and client address longstanding situations or problems with the objective to improve herd health and subsequently productivity performance. All three forms of communication overlap.Conclusions
Based on this model, it appears useful for a veterinary practice to offer both a curative and an advisory service, but to keep these two separated when deemed appropriate. This is a review of prion replication in the context of the cell biology of membrane proteins especially folding quality control in the endoplasmic reticulum (ER). Transmissible spongiform encephalopathies, such as scrapie and BSE, are infectious lethal diseases of mammalian neurons characterised by conversion of the normal membrane protein PrPC to the disease-associated conformational isomer called PrPSc. PrPSc, apparently responsible for infectivity, forms a number of different conformations and specific N-glycosylation site occupancies that correlate with TSE strain differences. Dimerisation and specific binding of PrPc and PrPSc seems critical in PrPSc biosynthesis and is influenced by N-glycosylation and disulfide bond formation. PrPsc can be amplified in vitro but new glycosylation cannot occur in cell free environments without the special conditions of microsome mediated in vitro translation, thus strain specific glycosylation of PrPSc formed in vitro in the absence of these conditions must take place by imprintation of PrPc from existing glycosylation site-occupancies. PrPSc formed in cell free homogenates is not infectious pointing to events necessary for infectivity that only occur in intact cells. Such events may include glycosylation site occupancy and ER folding chaperone activity. In the biosynthetic pathway of PrPSc, early acquisition of sensitivity of the GPI anchor to phospholipase C can be distinguished from the later acquisition of protease resistance and detergent insolubility. By analogy to the co-translational formation of the MHC I loading complex, it is postulated that PrPSc or its specific peptides could imprint nascent PrPc chains thereby ensuring its own folds and the observed glycosylation site occupancy ratios of strains. 相似文献

Electroejaculation in rhinoceroses has historically yielded inconsistent results, with the collection of high-quality, sperm-rich samples rare. The goal of this study was to develop a reliable method of electroejaculation in the rhinoceros by designing a rectal probe that appropriately fits the anatomy of this taxon and refining the procedure. A curved probe handle ending in an oblate, ellipsoid head was built using readily available supplies. A combination of rectal massage, penile massage, and electrical stimulation with a specially designed probe was employed in attempts to collect semen on 14 occasions from greater one-horned rhinoceroses (Rhinoceros unicornis; n = 4), black rhinoceroses (Diceros bicornis; n = 2) and a southern white rhinoceros (Ceratotherium simum; n = 1). During 13 of the 14 attempts, ejaculates were collected in multiple fractions. All but one of the ejaculates contained spermatozoa, and seven ejaculates contained good-quality fractions of semen (-60% sperm motility; > or =20 x 106 spermatozoa/ml) suitable for sperm banking and assisted reproduction procedures. Mean (+/-SEM) values for volume, pH, osmolality, and total sperm number for ejaculates containing good-quality fractions (98.2 +/-21.8 ml, 8.5+/-0.1, 290.4+/-6.7 mOsm, and 37.1+/-12.0 x 10(9), respectively) did not differ (P > 0.05) from those containing only poor-quality samples. Urine and/or erythrocyte contamination was not uncommon in fractions of both ejaculate types. Males producing good-quality samples ranged in age from 7 to 34 yr. None of the samples contained > or =75% morphologically normal spermatozoa. Electroejaculation with a uniquely designed probe consistently produced ejaculates in the rhinoceros. However, the production of high-quality samples continued to be challenging, occurring in only 50% of collection attempts. Regardless, the technology has progressed to a stage at which good-quality semen samples can be produced for sperm banking and assisted reproduction, and thereby can be integrated into intensive rhinoceros management strategies for the ultimate survival of this taxon. 相似文献

Root volume as a grading criterion to improve field performance of Douglas-fir seedlings 总被引:1,自引:0,他引:1
Three seed sources of 2+0 Douglas-fir [Pseudotsuga menziesii (Mirb.) Franco] seedlings were graded into three root-volume categories (<9, 9–13, and >13 cm3) and outplanted to determine (i) differences in survival and growth after one and two growing seasons in the field, (ii) relationship(s) of seedling height after one and two seasons to preplanting nursery root volume, total fresh weight, root-collar diameter, and height, and (iii) differences in field performance due to fertilization at planting. Field survival was >90% among all root-volume categories. Seedlings in the largest category grew significantly better than those in the smaller two categories over two seasons. Apparently, a population of seedlings having a large proportion of high root volumes will outperform a population having a large proportion of low root volumes in the field. Fertilization at the time of planting had no effect on survival or growth because of shallow placement of the fertilizer pellet. The results suggest that targeting root volume, as well as height and diameter, is worthwhile where morphological quality must be maximized to improve field performance. Thus, root volume has potential for use as a criterion for grading seedlings. 相似文献
