The main objective of the present work was to study the effect of cryopreservation of European eel sperm both on the sperm viability and the spermatozoa head morphology. Spermatozoa morphology was evaluated with computer-assisted morphology analysis after collection in fresh samples, after adding the freezing medium containing dimethyl sulfoxide as cryoprotectant and, finally, after the cryopreservation process and thawing. Cell viability was assessed, in both fresh and thawed samples, by Hoechst 33258 staining. Computer-assisted sperm analysis (CASA) was used to determine the percentage of motile cells and to measure motility parameters in sperm samples. A significant decrease of head perimeter (12.56%) and area (17.90%) was detected from spermatozoa in fresh to thawed samples, indicating that cells do not recover the original size after the cryopreservation process. CASA was used to measure the percentage of motile cells (51.9%) and spermatozoa motility parameters such as curvilinear, straight line and angular path velocities, as well as beating cross frequency. This technique was employed in the fresh sperm samples but proteins present at the freezing medium (L-alpha-phosphatidylcholine) made impossible to use this last technique in thawed samples. When sperm viability was assessed by Hoechst staining, a significant decrease of approximately 15% (73.10 vs 58.26%) of alive spermatozoa was registered from fresh to thawed samples. The percentage of motile cells measured by CASA in fresh samples (51.9%) was lower than the percentage of alive cells determined by Hoechst stainning, suggesting the existence of different batches of spermatozoa in different stages of development, even during the eight to tenth weeks of treatment, when the highest sperm quality was found.  相似文献

A quantum-cascade long-wavelength infrared laser based on superlattice active regions has been demonstrated. In this source, electrons injected by tunneling emit photons corresponding to the energy gap (minigap) between two superlattice conduction bands (minibands). A distinctive design feature is the high oscillator strength of the optical transition. Pulsed operation at a wavelength of about 8 micrometers with peak powers ranging from approximately 0.80 watt at 80 kelvin to 0.2 watt at 200 kelvin has been demonstrated in a superlattice with 1-nanometer-thick AlInAs barriers and 4.3-nanometer-thick GaInAs quantum wells grown by molecular beam epitaxy. These results demonstrate the potential of strongly coupled superlattices as infrared laser materials for high-power sources in which the wavelength can be tailored by design.  相似文献

Particle size fractions of soils from the surface 6 cm of two adjacent grassland plots which, as a result of different fertilizer treatments since 1897, have either a mor or a mull humus form were analysed using solid-state 13C nuclear magnetic resonance spectroscopy and fractionation of organic N by steam distillation. In the mor humus soil, which had received 180 kg (NH4)2SO4 ha?1 annually and was pH 4.3, there was more C and N in the larger particle size fractions than in the mull humus soil (pH 5.8). The NMR spectra of correspondingly sized soil fractions were similar for both soils. The intensities of NMR signals between 0 and 40 ppm (alkyl-C) and between 160 and 200 ppm (carbonyl-C) increased with decreasing particle size. The intensities of the NMR signals between 60 and 90 ppm (0-alkyl-C) and between 90 and 110 ppm (acetal- and ketal-C) decreased with increasing particle size. Comparison of the NMR spectra of the >2000 um fractions from both soils with those of dried grass litter from the same plots indicated the exclusive plant origin of the C in the largest size fraction of the soils. NMR resonances between 40 and 60 ppm were attributed to alkyl-amino-C because their intensities agreed with the amino-N determinations obtained during organic N fractionation. During incubation in soil microcosms, the larger sized fractions decomposed more rapidly than the smaller fractions. However, all the correspondingly sized particle fractions from the two soils decomposed at the same rate except the >2000 pm fractions. The largest size fraction from the mor humus soil decomposed faster than that from the mull humus soil. This difference in decomposition rate could not be attributed to differences in the chemical composition of the >2000 pm fraction.  相似文献

BACKGROUND: The UK government has published plans to reduce UK agriculture's greenhouse gas (GHG) emissions. At the same time, the goal of global food security requires an increase in arable crop yields. Foliar disease control measures such as fungicides have an important role in meeting both objectives. RESULTS: It is estimated that UK winter barley production is associated with GHG emissions of 2770 kg CO2 eq. ha?1 of crop and 355 kg CO2 eq. t?1 of grain. Foliar disease control by fungicides is associated with decreases in GHG emissions of 42–60 kg CO2 eq. t?1 in UK winter barley and 29–39 kg CO2 eq. t?1 in UK spring barley. The sensitivity of these results to the impact of disease control on yield and to variant GHG emissions assumptions is presented. Fungicide treatment of the major UK arable crops is estimated to have directly decreased UK GHG emissions by over 1.5 Mt CO2 eq. in 2009. CONCLUSION: Crop disease control measures such as fungicide treatment reduce the GHG emissions associated with producing a tonne of grain. As national demand for food increases, greater yields as a result of disease control also decrease the need to convert land from non‐arable to arable use, which further mitigates GHG emissions. High-power and highly directional semiconductor microcylinder lasers based on an optical resonator with deformed cross section are reported. In the favorable directions of the far-field, a power increase of up to three orders of magnitude over the conventional circularly symmetric lasers was obtained. A "bow-tie"-shaped resonance is responsible for the improved performance of the lasers in the higher range of deformations, in contrast to "whispering-gallery"-type modes of circular and weakly deformed lasers. This resonator design, although demonstrated here in midinfrared quantum-cascade lasers, should be applicable to any laser based on semiconductors or other high-refractive index materials.  相似文献

The aim of this study was to evaluate the effect of porcine luteinizing hormone (pLH) given at oestrous onset in gilts, by different routes and doses, on the interval between onset of oestrus and ovulation (IOEO) and reproductive performance using a single fixed‐time artificial insemination (FTAI). A total of 153 gilts were submitted to oestrous detection at 8‐h intervals and assigned to three groups: control – without hormone application and inseminated at 0, 24 and 48 h after oestrous onset; VS2.5FTAI – 2.5 mg pLH by the vulvar submucosal route at oestrous onset and a single FTAI 16 h later; IM5FTAI – 5 mg pLH by the intramuscular route at oestrous onset and a single FTAI 16 h later. More VS2.5FTAI gilts (47.1%; p < 0.05) ovulated within 24 h after oestrous onset than control gilts (25.5%) whereas IM5FTAI gilts had an intermediate percentage (31.4%; p > 0.05). The IOEO tended to be shorter (p = 0.06) in VS2.5FTAI (30.2 ± 1.4 h) than in control (34.7 ± 1.4 h) gilts, but there was no difference (p > 0.05) between control and IM5FTAI (32.8 ± 1.4 h) gilts. Farrowing rate was not different (p > 0.05) among treatments. Total born piglets (TB) was lower (p < 0.05) in VS2.5FTAI (12.3 ± 0.4) than in control gilts (14.1 ± 0.4), whereas intermediate TB was observed in IM5FTAI gilts (13.3 ± 0.4). Due to the advancement of ovulation, reduction of the hormonal dose and the ease of application, the vulvar submucosal route would be the best option for FTAI protocols, but their negative impact on litter size remains to be elucidated. Taking into account the good fertility results obtained in IM5FTAI gilts whose ovulation was not advanced, the possibility of a single FTAI without any hormonal treatment should be further investigated, to establish reliable FTAI protocols for gilts.  相似文献

Seventy-eight farms in south-west England, originally visited in 1970–72, were revisited in 1983 in order to identify changes in land use and re-seeding over the c. 12-year period; changes in sward composition were assessed on fifty-nine of these farms. The results were also compared with Ministry of Agriculture, Fisheries and Food (MAFF) Census results and MAFF Seeds Branch data.
The proportion of tillage had declined from 29 to 24%. This was in agreement with census results for the south-western counties, but contrasted with those for England and Wales as a whole, which showed an increase from 38.8 to 41.7% tillage. It is indicative of grass becoming more predominant in south-west England as east England has become more dominated by tillage cropping.
There had also been a decline in the amount of short-term grassland. In 1970–72 17% of the grassland was intended to last from 1 to 4 years; in 1983 the proportion was only 10%. That these intentions were being carried out was confirmed by the fact that in 1983 there was more grassland in each of the age categories 5·8,9·20 and over 20 years. The trend towards older grassland confirms that shown by regional and national census data, and the trend away from short-term swards is in line with deliveries of herbage seed which indicate reduced use of Italian ryegrass (Lolium multiflorum).
Botanical composition had changed appreciably, with the proportion of Lolium perenne having increased in swards of all ages, e.g. from 35 to 51% in swards 5–8 years old and from 17 to 31% in those aged over 20 years. Other sown species, predominantly Dactylis glomerata and Phleum pratense , had declined markedly in swards over 4 years old. The proportions of Agrostis spp., Festuca spp. and Poa trivialis had fallen, but Holcus lanatus had increased slightly. There had been a marked reduction in infestations of perennial broad-leaved weeds, particularly Ranunculus , but also Rumex and Cirsium spp.  相似文献

AIM: To investigate the anthelmintic properties of New Zealand native flax (Phormium tenax) for cattle, using a faecal egg count reduction test (FECRT).

METHODS: Twenty-six heifer calves with high (>300 epg) faecal nematode egg counts (FEC) were paired into two groups and fed either chopped flax leaves ad libitum, or barley straw at an equivalent dry matter (DM) rate, from Day 0–7; all were supplemented with 1.5 kg/head/day cereal feed supplement. On Days 8–10, all heifers were fed a common diet of 3.5 kg DM/ head/day barley straw and 1.5 kg/head/day cereal feed supplement. Heifers were weighed and faecal samples were collected on Days —6, 0, 7, and 10; samples were analysed in triplicate for FEC. The nutritive value of the flax and straw was analysed.

RESULTS: Both the flax and straw had low dry matter digestibility (DMD) and protein content. Although the flax-fed claves grew more than the controls, this may have been related to gut fill. Total daily faecal egg output was similar for flax and straw groups on Day 7 (14.7 vs 15.0 x 106 eggs/day, respectively) and Day 10 (14.9 vs 15.1 x 106 eggs/day, respectively). There was no difference in the change in FEC with time between the calves fed flax or straw diets.

CONCLUSION: Consumption of flax leaves did not reduce FEC in calves with a mixed nematode infection.  相似文献
