Fusarium oxysporum f. sp. lycopersici (FOL) races and F. oxysporum f. sp. radicis lycopersici (FORL), the causal agents of root rot and crown rot diseases, respectively, cause serious economic losses in tomato greenhouses where production is intensive in the West Mediterranean region of Turkey. The isolates were collected from West Mediterranean region of Turkey and were characterized by specific primers based on three races (r1, r2, r3), besides pathogenicity tests in in vivo conditions Additionally, a scheme was developed using newly tested ISSR and SRAP markers to a genotyping database and to determine the possible origin of these pathogens. The present study provided new information on these pathogens based on their races and their dominant existence in this region that has not been reported before. Genetic diversity detected in the same races of the pathogen may be associated with difficulties in controlling the pathogen and a possible resistance formation effort exerted by the pathogen to chemicals used in plant protection in tomato greenhouses. Molecular analyses indicated genetic diversity in pathogen isolates identified as r3, r2 and FORL, which may be associated with abiotic stress to which the pathogens were exposed. 相似文献

Two surveys (2005/2006 and 2009) were conducted in the state of São Paulo, Brazil, to investigate the incidence of ‘Candidatus Liberibacter asiaticus’ and ‘Ca. L. americanus’, two liberibacters associated with citrus huanglongbing (HLB) disease and both transmitted by Diaphorina citri, in orange jasmine (Murraya exotica), a widespread ornamental tree in cities and villages. The graft‐transmissibility of the two species, and their DNA relatedness to citrus‐associated liberibacters, were also investigated. Quantitative PCR was applied to PCR‐positive orange jasmine and HLB‐positive citrus growing in backyards and orchards to assess their inoculum source potentials. Liberibacters were detected in 91 of 786 sampled orange jasmine plants in 10 of 76 sampled locations. PCR‐positive trees exhibited yellow shoots and/or dieback symptoms indistinguishable from those on PCR‐negative trees. ‘Candidatus Liberibacter americanus’ was more common in 2005/2006 (96·6%) and ‘Ca. L. asiaticus’ in 2009 (84·8%). rplJ nucleotide sequences were identical within all populations of either species. Graft transmission succeeded only in homologous host combinations, including ‘Ca. L. americanus’ (2/10) from/to orange jasmine and ‘Ca. L. americanus’ (5/18) and ‘Ca. L. asiaticus’ (5/9) from/to citrus. Symptoms were mild and developed less rapidly in orange jasmine than in citrus, probably as a result of lower liberibacter multiplication rates. Respective titres of ‘Ca. L. americanus’ and ‘Ca. L. asiaticus’ in orange jasmine averaged 4·3 and 3·0 log cells g?1 tissue, compared with 5·5 and 7·3 in citrus. The results indicate that orange jasmine does not favour liberibacter multiplication as much as citrus. However, its importance in HLB epidemics should not be underestimated as it is a preferred host of D. citri and is not under any strict tree‐eradication programme or measures for insect control. 相似文献

Papaya ringspot virus‐type W (PRSV‐W) is the most prevalent and important viral pathogen of cucurbits in Brazil. It can be effectively controlled by the incorporation of genetic resistance into susceptible melon cultivars. The present study identified amplified fragment length polymorphic (AFLP) markers linked to the PRSV‐W resistance Prv1 allele. The susceptible yellow‐fleshed melon‐breeding line AF426prv1 and its nearly isogenic‐resistant line AF426Prv1, which carries the Prv1 allele resident in the Indian cantaloupe U.S. Plant Introduction (PI) 180280, were screened for AFLP marker polymorphisms. Of 30 251 AFLP loci, only three were polymorphic between the nearly isogenic lines. Segregation analyses for these three polymorphic markers and the Prv1 allele using a BC1 population of 197 plants indicated close linkage (0.5% recombination frequency) between marker EK190 (HindIII‐CGA and MseI‐GTG; 190 bp) and Prv1. Thus, EK190 might be a useful marker in breeding programmes aiming to develop melon cultivars resistant to PRSV‐W. The other two markers are closely linked to each other, but distantly linked to Prv1. 相似文献

Magnetic fields (MFs) have been applied for the first time in orchid micropropagation. Protocorm-like bodies (PLBs) – approximately 3 mm in diameter – first derived from leaf segment culture of Phalaenopsis Gallant Beau ‘George Vazquez’, were subcultured every 2 months, and served as initial explants. The proliferation of Phalaenopsis PLBs in liquid medium in the Miracle Pack® culture system was affected by the action of different intensities and polarities of MFs: 0.1, 0.15 and 0.2 Tesla (T) at North (N) and South (S) poles. The MF of 0.1 T – S resulted in the greatest fresh weight of regenerated PLBs. The average number of neo-PLBs formed per clusters in the PLB treated by MF: 0.1–0.2 T was decreased compared to the control exposed to natural MF (5 × 10−6 T). The proliferation of PLBs under 0.15 T – MF at both N and S poles for 2 and 7 weeks demonstrated that a longer duration of exposure to an MF of 0.15 T, regardless of the polarity, resulted in greater biomass of newly formed PLBs and smaller average number of newly formed PLBs. The S pole of MF had stronger effects on Phalaenopsis PLBs proliferation than the N pole did in all treatments. 相似文献

The goal of this study was to evaluate the effect of cold cathode fluorescent lamps (CCFLs) on the growth of Gerbera jamesonii var. ‘Rui Kou’ plantlets in vitro in six different light quality ratios: 100% red CCFL (R), 80% R + 20% blue CCFL (B), 70% R + 30% B, 60% R + 40% B, 100% B and white CCFLs (W). Control radiation was provided by conventional heat-generating plant growth fluorescent lamps (PGFLs). Plantlets under CCFLs showed better plantlet height, SPAD value (i.e., leaf chlorophyll content) and root activity (as assessed by root dehydrogenase activity) than those growing under PGFLs while all other growth parameters were comparable with plants under conventional lighting systems. 相似文献

An attempt was made to understand the molecular systematics and genetic differences between 10 original chrysanthemum cultivars and 11 mutants. The similarity among the cultivars and mutants varied from 0.17 to 0.90 using RAPD analyses, a simple but efficient method to distinguish cultivars and to assess parentage. Two distinct groups were found. Two cultivars were present as a separate group showing differences from all other cultivars. Mutants with different flower colour could be identified at the molecular level using RAPD technique holding promise to identify unique genes as SCAR markers. A high genetic distance among the different chrysanthemums showed that there exists a possibility of introgressing new and novel genes from the chrysanthemum gene pool. 相似文献

Context
Seed dispersal is recognized as having profound effects on the distribution, dynamics and structure of plant populations and communities. However, knowledge of how landscape structure shapes carnivore-mediated seed dispersal patterns is still scarce, thereby limiting our understanding of large-scale plant population processes.Objectives
We aim to determine how the amount and spatial configuration of forest cover impacted the relative abundance of carnivorous mammals, and how these effects cascaded through the seed dispersal kernels they generated.Methods
Camera traps activated by animal movement were used for carnivore sampling. Colour-coded seed mimics embedded in common figs were used to know the exact origin of the dispersed seed mimics later found in carnivore scats. We applied this procedure in two sites differing in landscape structure.Results
We did not find between-site differences in the relative abundance of the principal carnivore species contributing to seed dispersal patterns, Martes foina. Mean dispersal distance and the probability of long dispersal events were higher in the site with spatially continuous and abundant forest cover, compared to the site with spatially aggregated and scarcer forest cover. Seed deposition closely matched the spatial patterning of forest cover in both study sites, suggesting behaviour-based mechanisms underpinning seed dispersal patterns generated by individual frugivore species.Conclusions
Our results provide the first empirical evidence of the impact of landscape structure on carnivore-mediated seed dispersal kernels. They also indicate that seed dispersal kernels generated strongly depend on the effect that landscape structure exerts on carnivore populations, particularly on habitat-use preferences.88.
Energy balance measurements were carried out in a mango orchard during two growing seasons in the semi-arid region of Brazil.
The actual evapotranspiration (ET) was acquired by eddy correlation (EC) and Bowen ratio energy balance (BR) techniques. The
daily energy balance closure in the EC measurements showed an average gap of 12%, with a root mean square error (RMSE) of
1.7 MJ m−2 d−1. Three different correction procedures were tested for closing the energy balance from the EC system: (1) the surface energy
balance residual method (RES), (2) the Bowen ratio determined from the EC fluxes, the combination approach (EC_BR), and (3)
a new regression energy balance closure technique (REG). All closing energy balance methods presented good correlation with
the direct EC measurements, but the trends were not similar. The latent heat fluxes estimated by the BR method—λE
BR—were higher than those from the direct EC measurements—λE
EC. When using the RES method, the half-hour λE
EC measurements represented around 88% of the λE
RES values, as the uncertainties of net radiation—R
n—and soil heat fluxes—G—are propagated into the RES method. The latent heat flux derived from the combination approach—λE
EC_BR—also brings these uncertainties, being the agreements comparable with those for RES method. It was therefore concluded that
a single correction method for EC measurements considering only the latent and sensible heat fluxes does not exist. A new
way to solve the lack of energy balance closure from EC techniques was tested by means of a curve fitting, the REG method.
Considering the REG corrections applied to the energy balance components involving all periods of the day and the average
conditions of the two growing seasons, half-hour values of λE
EC were overmeasured by 18%, H
EC was undermeasured by 17%, and G values required a correction of 466%. The REG method appeared promising because it considers different weights for all energy
balance components in the optimization process. Taking the REG results for the drier second growing season as a reference,
it was concluded that seasonal ET values by the other methods in mango orchard ranged from 7 to 28% higher, showing that turbulent
flux measurements lack accuracy for executing on-farm water-saving programmes and calibrating transient soil water flow models. 相似文献

The objectives of this work were to evaluate the genetic variability of Meloidogyne enterolobii by molecular markers, and develop species‐specific molecular markers for application in detection. Sixteen M. enterolobii isolates from different geographical regions (Brazil and other countries) and hosts were used in this study. The identification and purification of the populations were carried out based on isoenzyme phenotype. The DNA amplification of the intergenic region (IGS) of the rDNA and of the region between the cytochrome oxidase subunit II (COII) and 16S rRNA genes (mtDNA) produced specific fragments of the expected size for this nematode, i.e. 780 and 705 bp, respectively. Intraspecific variability among the isolates was evaluated with three different neutral molecular markers: AFLP, ISSR and RAPD. The results showed a low level of diversity among the isolates tested, indicating that M. enterolobii is a genetically homogeneous root‐knot nematode species. The RAPD method allowed the identification of a species‐specific RAPD fragment for M. enterolobii. This fragment was cloned and sequenced, and from the sequence obtained, a set of primers was designed and tested. The amplification of a 520‐bp‐long fragment occurred only for the 16 isolates of M. enterolobii and not for the 10 other Meloidogyne species tested. In addition, positive detection was achieved in a single individual female, egg‐mass and second stage juvenile of this nematode. This SCAR species‐specific marker for M. enterolobii represents a new molecular tool to be used in the detection of this nematode from field samples and as a routine diagnostic test for quarantine devices . 相似文献

Cardiovascular diseases represent a major cause of disability and death worldwide. Therapeutics are available, but they often have unsatisfactory results and may produce side effects. Alternative treatments based on the use of natural products have been extensively investigated, because of their low toxicity and side effects. Marine organisms are prime candidates for such products, as they are sources of numerous and complex substances with ecological and pharmacological effects. In this work, we investigated, through in vitro experiments, the effects of three diterpenes (pachydictyol A, isopachydictyol A and dichotomanol) from the Brazilian marine alga, Dictyota menstrualis, on platelet aggregation and plasma coagulation. Results showed that dichotomanol inhibited ADP- or collagen-induced aggregation of platelet-rich plasma (PRP), but failed to inhibit washed platelets (WP). In contrast, pachydictyol A and isopachydictyol A failed to inhibit the aggregation of PRP, but inhibited WP aggregation induced by collagen or thrombin. These diterpenes also inhibited coagulation analyzed by the prothrombin time and activated partial thromboplastin time and on commercial fibrinogen. Moreover, diterpenes inhibited the catalytic activity of thrombin. Theoretical studies using the Osiris Property Explorer software showed that diterpenes have low theoretical toxicity profiles and a drug-score similar to commercial anticoagulant drugs. In conclusion, these diterpenes are promising candidates for use in anticoagulant therapy, and this study also highlights the biotechnological potential of oceans and the importance of bioprospecting to develop medicines. 相似文献
