Random amplified polymorphic DNA (RAPD) markers have been used in a variety of genetic studies in fisheries and aquaculture. Most population studies are performed without preliminary data demonstrating the Mendelian inheritance and reproducibility of RAPD markers. In this study, the inheritance and reproducibility of RAPD markers was examined in two consecutive generations of common carp, Cyprinus carpio L. Variability and segregation of RAPD markers were investigated in one F1 progeny and three F2 progenies. Seventy-four RAPD markers were generated by five primers using DNA extracted from the initial ornamental (koi) common carp female and wild-type colour common carp male. Fifty-five of these RAPD markers were transmitted to the F1 progeny and the inheritance patterns were analysed. Twenty RAPD markers were fully reproducible and demonstrated dominant simple Mendelian inheritance patterns in two consecutive generations. Twenty-four RAPD markers were not reproducible in all progenies. Thirteen markers displayed inheritance ratios in the progenies that did not fit simple Mendelian inheritance patterns. Non-reproducibility of RAPD markers and distorted ratios may be caused by the absence of amplification, poor amplification or by the appearance of artefact bands. Random amplified polymorphic DNA markers with poor reproducibility and non-Mendelian inheritance can lead to misinterpretations of data in population studies, resulting in errors in the estimation of genetic diversity within and between individual populations. Therefore, it is recommended to first identify the set of reproducible RAPD markers that demonstrate Mendelian inheritance before application of the RAPD technique in population studies. 相似文献

An experiment was conducted to evaluate the effects of the RN genotype on skeletal muscle characteristics in pigs sharing otherwise the same polygenic background. Animals were genotyped for RN on the basis of RTN (Rendement Technologique Napole) records using segregation analysis methods. Samples of longissimus (L) and semispinalis capitis (S) muscles were taken from 39 rn+/rn+, 38 RN-/rn+ and 37 RN-/RN- pigs slaughtered at 108 +/- 8.6 kg live weight. Activities of lactate dehydrogenase (LDH), citrate synthase (CS), and beta-hydroxy-acyl-coenzyme A dehydrogenase (HAD) were measured on both muscles to assess glycolytic, oxidative, and lipid beta-oxidation capacities, respectively. Histological examinations and chemical analyses were performed on L muscle. The energetic metabolism of the white L muscle was more oxidative in RN-/RN- than in rn+/rn+ pigs, as shown by increased CS and HAD activities (P < .001), decreased LDH activity (P < .001), larger cross-sectional area of IIA (P < .05) and IIB-red (P < .05) fibers, higher relative area of IIA fibers ( P < .05), and lower relative area of IIB-white fibers (P < .001). No significant difference was found between heterozygous and homozygous carriers of the RN- allele, except for CS activity, which was lower in RN-/rn+ than in RN-/RN- pigs. In L muscle, the RN- allele led to a large increase in glycolytic potential (+3.5 phenotypic SD between homozygotes) and lightness (+.7 SD), and a decrease in ultimate pH, dry matter, and protein contents (-1.7 to -2 phenotypic SD for these three traits), with an almost completely dominant effect. No differences were found between genotypes for intramuscular fat and hydroxyproline contents. In the red S muscle, the presence of RN- had no influence on enzyme activities. These results indicate that the RN genotype greatly influences compositional and histochemical traits and metabolic enzyme activities in a muscle type-dependent manner, with a completely or incompletely dominant effect of the RN- allele. 相似文献

Little information is available in Turkey on Q fever, a zoonose caused by Coxiella burnetii and transmitted from domestic ruminants. This study aimed at investigating the seroprevalence in sheep flocks from three provinces (Bursa, Balikesir and Canakkale). Serosurvey was undertaken on 42 flocks, which were categorised by sizes. Sera were collected randomly from specific age groups within the young population. CHEKIT Q-fever ELISA kit was used to identify the infection in sheep. The results showed that 20% (n=151) of sheep were seropositive. A total of 34 flocks (81%) revealed at least one seropositive animal. Higher seroprevalence was observed in Balikesir region. Larger flocks resulted more infected than medium and small flocks. An association was found between seropositivity and age, when the primiparous ewes (1-year old) had higher antibodies rates than newborn sheep (aged less than 10 months) or biparous ewes (2 years old). These results showed that Q fever infection was common and circulating in the studied region, hence encourage efforts to propose measures that could reduce the spread and the zoonotic risk. 相似文献

Kittens immunized with purified native FeLV-gp70 or -gp85 envelope proteins developed ELISA, but not virus neutralizing, antibodies in their serum to both whole FeLV and FeLV-gp70. Kittens vaccinated with envelope proteins and infected with feline sarcoma virus (FeSV) developed smaller tumors than nonvaccinates, but a greater incidence of persistent retroviremia. Similarly, FeLV-gp70 and -gp85 vaccinated kittens were more apt to become persistently retroviremic following virulent FeLV challenge exposure than nonvaccinates. Kittens vaccinated with inactivated whole FeLV developed smaller tumors after FeSV inoculation and had a lower incidence of persistent retroviremia than nonvaccinates. The protective effect of inactivated whole FeLV vaccine against persistent retroviremia was also seen with FeLV challenge-exposed cats. Protection afforded by inactivated whole FeLV vaccine was not associated with virus neutralizing antibodies, although ELISA antibodies to both whole FeLV and FeLV-gp70 were induced by vaccination. 相似文献

No precocious metamorphosis or other morphogenetic effects were seen following topical treatment of fifth instar Spodoptera exempta with pre- cocene II. [3H]-Precocene II penetrated the larvae rapidly following topical application. An uptake of 47% was recorded in the first hour and an average of 27% for the first 3 h. Radioactivity in the haemolymph reached a maximum 4 h after treatment at 6–8% of the applied dose. A maximum titre of precocene of 1.1 × 10?4 M was observed 2 h after treatment and a biological half-life of around 1 h was recorded for the first 6 h. The metabolite precocene 11–3, 4-dihydrodiol appeared in the haemolymph with a maximum titre of 1.1 × 10?4 M at 4 h suggesting metabolism via the 3, 4-epoxide. At the same time an unidentified metabolite was observed possibly corresponding to the 6- and 7-(O)-desmethylated products described by others. Very large quantities of highly polar materials were observed in the haemolymph throughout the experiment and it is thought that these corresponded to conjugates. The evidence suggests that peripheral detoxication mechanisms do not operate sufficiently rapidly to prevent the establishment of a high titre of precocene. This, however, did not affect the corpora allata in this insect, whereas in a sensitive species that has been studied by other workers, necrosis of the glands would have resulted. Rainbow trout Oncorhynchus mykiss (Walbaum) are usually cultured at high densities to maximize production, but little is known about the physiological and behavioural consequences of high‐density fish culture. The purpose of this study was to develop quantitative correlates of activity for fish held under conditions of increasing density. Fifteen hatchery‐reared rainbow trout (mean fork length = 432.3 ± 9.2 mm) were implanted with activity (electromyogram; EMGi) transmitters and randomly assigned to each of three replicate tanks. Original tank densities (15 kg m?3) were then increased to 30 and finally to 60 kg m?3 at weekly intervals by adding additional fish. Remote telemetry signals indicated that activity increased with increasing stocking density. Fish were relatively inactive during the middle of the day, with diel activity patterns not differing among treatments. Fish were more active during periods of darkness, with activity increasing with increasing stocking density. Relationships between swimming speed, EMGi activity and oxygen consumption were developed using a respirometer and used to estimate oxygen consumption of the fish in the density treatments. Average oxygen consumption estimates increased with increasing density treatments as follows: low density = 75.6 mg kg?1 h?1; medium density = 90.0 mg kg?1 h?1; and high density = 102.6 mg kg?1 h?1. Telemetry permits quantification of the effects of increasing density on fish activity. Physiological telemetry devices may provide a useful tool for remotely monitoring animal welfare correlates under controlled conditions for fish exposed to different husbandry conditions and may prove a valuable tool for the aquaculture industry. 相似文献

A total of 2053 import and 164 export samples from 425 production plants were examined over a 9-year period (1992-2000) for the presence of Listeria monocytogenes (L. monocytogenes) in Switzerland. Overall, 282 samples (12.2%) and 85 plants (20.5%) harbored the pathogen. The highest isolation risk was for marinated fish (38%); the lowest was in cured- and dried-meat products.Unconditional fixed-effect logistic regression was used to identify the main hazards associated with the presence of L. monocytogenes. The plant-level model considered potential risk factors for a positive culture operating at the production-plant level by including a random effect of plant and year.Food category was the only significant factor; sampling site, country of origin and season were not significant. Marinated fish was a strong predictor for positive culture, whereas cooked- and cured-meat products were protective. Plant and year effects were significant. Control measures should be focused on specific food items in each production plant. 相似文献
