OBJECTIVE: To isolate and characterize the eighth component of the complement system (C8) in cattle. SAMPLE POPULATION: Fresh plasma obtained from beef cattle. PROCEDURES: Plasma samples were fractionated, using sequential precipitation and ion-exchange and gel-filtration chromatography, to yield C8. The protein was identified throughout the procedure on the basis of its hemolytic function. Electrophoresis in polyacrylamide gels was used to determine molecular weight and composition of polypeptide chains. Reconstitution of classical and alternative complement pathways was used to characterize the hemolytic function of bovine C8. RESULTS: The bovine C8 protein consisted of a disulfide-bonded alpha-gamma heterodimer that was noncovalently associated with a beta chain. Apparent molecular weight of the alpha, beta, and gamma chains under reducing conditions were 66, 61, and 23 kd, respectively. In the classical pathway of activation, bovine C8 and the ninth component of the complement system (C9) had species incompatibility with human C8 and C9 on sheep erythrocyte target cells. CONCLUSIONS: A simple 4-step fractionation procedure provided good yield of bovine C8 from plasma. The isolated protein was structurally comparable to C8 from other species. Purified bovine C8 may be useful in functional hemolytic assays to investigate the roles of complement-mediated lysis in the pathogenesis of inflammatory diseases and the killing of susceptible microorganisms. 相似文献

Seven mathematical models, which have been advanced for describing phosphate sorption by soils, are studied here with respect to their statistical behaviour in estimation. These seven non-linear regression models include the Langmuir equation and two extensions of it, the Freundlich equation and two extensions of it, and Gunary's equation. Measures of non-linear behaviour, such as the Bates & Watts (1980) curvature measure of intrinsic non-linearity and the Lowry & Morton (1983) asymmetry measure of non-linearity, were calculated for each model in combination with each of six data sets. It was found that the Freundlich equation and the extension of it proposed by Sibbesen behaved best, with Gunary's equation also having acceptable statistical properties, whereas the Langmuir equation and its extensions behaved worse, exhibiting properties which indicate that the estimators of their parameters would be severely biased and non-normal in distribution. It is believed that similar conclusions may apply to other processes involving surface adsorption. 相似文献

The aim of this paper is to assess the greenhouse gas (GHG) mitigation potential of croplands and grasslands in Great Britain under different management practices. We consider the feasible land management options for grass and cropland using county level land‐use data with estimates of per‐area mitigation potential for individual and total GHGs, to identify the land management options with the greatest cost‐effective mitigation potential. We show that for grasslands, uncertainties still remain on the mitigation potential because of their climatic sensitivity and also their less intensive management. For croplands in Great Britain, the technical mean GHG mitigation potentials for all cropland management practices range from 17 Mt CO2‐eq. per 20 yr to 39 Mt CO2‐eq. per 20 yr. There are significant regional variation in all cases, with the greatest potentials in England, negligible potential in Wales and intermediate potential in Scotland, with country differences largely driven by the areas of cropland and grassland in each country. Practices such as agronomic improvement and nutrient management are the most promising options because of their impact on N2O emissions and also their larger potential at low cost. In terms of annual emissions from agriculture, calculated mitigation potentials are small, where the technical mitigation potential of agronomy and nutrient management strategies are ca. 4.5 and 3.8%, respectively (agricultural emissions account for ca. 9% or 47.7 Mt CO2‐eq., of total Great Britain GHG emissions, Department of Energy and Climate Change, UK). However when compared with the land use, land‐use change and forestry sector (LULUCF) emissions, nutrient management would reduce further emission reductions by approximately half of the 2005 LULUCF sink (i.e. ?1.6 Mt CO2‐eq. per year). 相似文献

Tricyclazole (5-methyl-1,2,4-triazolo[3,4-b]benzothiazole) inhibits melanin synthesis in Pyricularia oryzae at concentrations less than 0.01 μg/ml. The primary site of inhibition in the biosynthetic pathway occurs between scytalone and vermelone. Accumulation of several metabolites derived from melanin precursors along branch pathways is associated with inhibition of melanin biosynthesis. At low tricyclazole concentrations (0.01–1 μg/ml), predominant accumulation of 2-hydroxyjuglone and 3,4-dihydro-3,4,8-trihydroxy-1-(2H)-naphthalenone (3,4,8-DTN) occurs as a result of the primary block between 1,3,8-trihydroxynaphthalene and vermelone. As the concentration of tricyclazole is increased from 1 to 10 μg/ml, flaviolin accumulation is markedly enhanced, whereas that of 3,4,8-DTN and 3,4-dihydro-4,8-dihydroxy-1-(2H)-naphthalenone is depressed, indicating possible secondary sites of inhibition in the main and branch pathways. Five melanin-deficient mutants of P. oryzae that phenotypically resemble the tricyclazole-treated wild-type strain were nonpathogenic or rarely infected two rice varieties. Three of the mutants studied were genetically defective in the melanin biosynthetic pathway at the site blocked by tricyclazole in the wild type. The wild-type strain converted both scytalone and vermelone to melanin; whereas the three mutants and the tricyclazole-treated wild type converted only vermelone to melanin. We derive a general model, based on principles of biochemical kinetics and allometry, that characterizes the effects of temperature and body mass on metabolic rate. The model fits metabolic rates of microbes, ectotherms, endotherms (including those in hibernation), and plants in temperatures ranging from 0 degrees to 40 degrees C. Mass- and temperature-compensated resting metabolic rates of all organisms are similar: The lowest (for unicellular organisms and plants) is separated from the highest (for endothermic vertebrates) by a factor of about 20. Thirty-six mice were inoculated intranasally with Bordetella parapertussis organisms (isolated from sheep with chronic non-progressive pneumonia) to study their deposition and clearance in the lower respiratory tract. The deposition of the organism was greater in the trachea than in the lungs. At 48 hours after inoculation, almost 100% of organisms were cleared from the trachea but only 55% of organisms were cleared from the lungs. This result correlates well with the morphological changes seen previously in the pulmonary parenchyma and airways of mice and lambs experimentally infected with this organism. 相似文献
