Clonal gene therapy: transplanted mouse fibroblast clones express human alpha 1-antitrypsin gene in vivo

A retroviral vector was used to insert human alpha 1-antitrypsin (alpha 1AT) complementary DNA into the genome of mouse fibroblasts to create a clonal population of mouse fibroblasts secreting human alpha 1AT. After demonstrating that this clone of fibroblasts produced alpha 1AT after more than 100 population doublings in the absence of selection pressure, the clone was transplanted into the peritoneal cavities of nude mice. When the animals were evaluated 4 weeks later, human alpha 1AT was detected in both sera and the epithelial surface of the lungs. The transplanted clone of fibroblasts could be recovered from the peritoneal cavities of those mice and demonstrated to still be producing human alpha 1AT. Thus, even after removal of selective pressure, a single clone of retroviral vector-infected cells that expressed an exogenous gene in vitro, continued to do so in vivo, and when recovered, continued to produce the product of the exogenous gene.     

Stock evaluation of giant scallop (Placopecten magellanicus) using high-resolution acoustics for seabed mapping

Survey designs in use for the evaluation of sea scallop stocks do not consider the variability of sediment type, despite strong evidence of its importance for the recruitment and survival of scallops on the sea floor. This study examines the distribution of scallops on Browns Bank, Scotian Shelf, at two test sites, in comparison to sea floor sediment distribution, with particular attention to the effects of small-scale sediment variability on the abundance of the commercially exploited scallop. Important links between scallop abundance, sediment type and habitat structure are described. Scallops are strongly associated with gravel lag deposits, which are readily distinguishable from sand-covered terrain through the use of multibeam backscatter data. There exists a highly significant correlation between scallop survey catch rates and backscatter intensity which can be used for the prediction of scallop stock abundance. Developments in underwater acoustics enable for more precise sea floor mapping and contribute to better estimates of scallop abundance.     

Clinical characteristics, treatment, and outcome of dogs with presumed primary hepatic lymphoma: 18 cases (1992-2008)

OBJECTIVE-To determine outcome of dogs with presumed primary hepatic lymphoma treated with various multiagent, doxorubicin-based chemotherapeutic protocols and identify factors associated with prognosis. DESIGN-Retrospective case series. ANIMALS-18 dogs with presumed primary hepatic lymphoma. PROCEDURES-Medical records were reviewed for information on signalment, treatment, and outcome. RESULTS-8 dogs had a complete remission (CR), with a median remission duration of 120 days. Dogs with leukocytosis, neutrophilia, hypoalbuminemia, hyperbilirubinemia, or a combination of hypoalbuminemia and hyperbilirubinemia were less likely to achieve a CR. Overall median survival time (MST) was 63 days (range, 2 to 402 days). In a multivariate analysis, response to treatment and serum albumin concentration were associated with MST. Dogs that did not achieve a CR had a significantly shorter MST than did dogs that did achieve a CR (13 vs 283 days, respectively). Dogs with serum albumin concentration < 2.5 g/dL at the time treatment was initiated had a significantly shorter MST than did dogs with serum albumin concentration within reference limits (10 vs 128 days, respectively). There was also a positive correlation between serum albumin concentration and survival time (r = 0.74). CONCLUSIONS AND CLINICAL RELEVANCE-Results suggested that dogs with primary hepatic lymphoma that underwent chemotherapy had a poor prognosis, with a low response rate. Dogs that responded to treatment had a better prognosis, and dogs with hypoalbuminemia had a poorer prognosis.     

Increasing net CO2 uptake by a Danish beech forest during the period from 1996 to 2009

The exchange of CO₂ between the atmosphere and a beech forest near Sorø, Denmark, was measured continuously over 14 years (1996-2009). The simultaneous measurement of many parameters that influence CO₂ uptake makes it possible to relate the CO₂ exchange to recent changes in e.g. temperature and atmospheric CO₂ concentration. The net CO₂ exchange (NEE) was measured by the eddy covariance method. Ecosystem respiration (RE) was estimated from nighttime values and gross ecosystem exchange (GEE) was calculated as the sum of RE and NEE. Over the years the beech forest acted as a sink of on average of 157 g C m⁻² yr⁻¹. In one of the years only, the forest acted as a small source. During 1996-2009 a significant increase in annual NEE was observed. A significant increase in GEE and a smaller and not significant increase in RE was also found. Thus the increased NEE was mainly attributed to an increase in GEE. The overall trend in NEE was significant with an average increase in uptake of 23 g C m⁻² yr⁻². The carbon uptake period (i.e. the period with daily net CO₂ gain) increased by 1.9 days per year, whereas there was a non significant tendency of increase of the leafed period. This means that the leaves stayed active longer. The analysis of CO₂ uptake by the forest by use of light response curves, revealed that the maximum rate of photosynthetic assimilation increased by 15% during the 14-year period. We conclude that the increase in the overall CO₂ uptake of the forest is due to a combination of increased growing season length and increased uptake capacity. We also conclude that long time series of flux measurements are necessary to reveal trends in the data because of the substantial inter-annual variation in the flux.
