To understand the effect of nematode Meloidogyne exigua infestation on coffee plants, resistant and susceptible coffee seedlings were inoculated with second-stage juveniles of M. exigua, and root metabolites were studied for four time intervals at 0, 24, 48 and 96 h. During this important period for parasite establishment, the concentrations of phenols, carbohydrates, amino acids and alkaloids in the roots were measured, and hydrogen nuclear magnetic resonance spectra of the root extracts were used to identify and quantify other metabolites. One of the most striking changes was the concentration of fumaric acid on resistant plants, which varied from 59 μg g(of root)?1 to 138 μg g(of root)?1 during the first 24 h of the nematode inoculation. The same level of variation was observed much later (96 h) in susceptible plants. Similarly, formic and quinic acid concentrations increased more rapidly in the resistant plants compared to the susceptibles. Sucrose concentrations increased to 370 % in the first 48 h in the resistant plants but showed no significant variation in the susceptible plant. Besides, the concentration of alkaloids was much higher at 24 and 48 h in the susceptibles compared to the resistant plants. These results suggest that the higher production of sucrose as well as formic, fumaric and quinic acids, and the lower production of alkaloids by the resistant cultivar in the first 48 h after the nematode inoculation are associated with the resistance of coffee plants to M. exigua.  相似文献

Over the last two decades, soil cultivation practices in the southern Argentinean Pampas have been changing from a 7 year cash-crop production system alternated with 2–3 years under pasture, to a continuous cropping system. A better understanding of the impact of the period of time a field has been under continuous cropping on a broad spectrum of soil properties related to soil quality is needed to target for sustainable cropping systems. The objectives of this study were to: (i) assess the relationship between physical and chemical soil parameters related to soil quality and (ii) identify soil quality indicators sensitive to soil changes under continuous cropping systems in the Argentinean Pampas.

Correlation analysis of the 29 soil attributes representing soil physical and chemical properties (independent variables) and years of continuous cropping (dependent variable) resulted in a significant correlation (p < 0.05) in 78 of the 420 soil attribute pairs. We detected a clear relationship between hydraulic conductivity at tension h (Kh) and structural porosity (ρe); ρe being a simple tool for monitoring soil hydraulic conditions.

Soil tillage practice (till or no-till) affected most of the soil parameters measured in our study. It was not possible to find only one indicator related to the years under continuous cropping regardless of the cultivation practice. We observed a significant relationship between years under continuous cropping and Kh under no-till (NT) and wheat fallow (p < 0.001, R2 = 0.70). Under these conditions, K−40 diminished as the number of years under continuous cropping increased.

The change in mean weight diameter (CMWD) was the only physical parameter related to the number of years under continuous cropping, explaining 36% of the variability in the number of years under continuous cropping (p < 0.001) The combination of three soil quality indicators (CMWD, partial R2 = 0.38; slope of the soil water retention curve at its inflexion point (S), partial R2 = 0.14 and cation exchange capacity (CEC), partial R2 = 0.13) was able to explain, in part, the years under continuous cropping (R2 = 0.65; p value > 0.001), a measure related to soil quality.  相似文献

Increasing atmospheric CO2 is recognized as a major aspect of global climate change that would have a significant impact on the productivity of major agricultural crops. Two field experiments were done, with the objective of quantifying the response of a short‐duration rice (Oryza sativa) variety (BG‐300) to elevated atmospheric carbon dioxide, in the low elevation, subhumid zone of Sri Lanka. The experiment contained three treatments. In the elevated CO2 treatment, rice was grown at a CO2 concentration of 570 µmol/mol within open top chambers (OTC s). The ambient CO2 treatment included crops grown within OTC s, but maintained at the ambient CO2 concentration of 370 µmol/mol. The third treatment was a crop grown in the open field under ambient CO2 concentration. Grain yields of rice crops grown under elevated CO2 were 24 % and 39 % greater than the respective ambient treatments in the maha (January – March 2001) and yala (May – August, 2001) seasons. Significant increases in total biomass at harvest (23 % and 39 %, respectively, in maha and yala) were more responsible for the above yield increases than the slight increases in the harvest index (4 % and 2 %). Yields of the ambient and open field treatments did not differ significantly. Among the yield components, the number of panicles per hill was significantly higher in the elevated treatment and showed significant positive correlations with grain yield in both seasons. In addition, grain yield showed significant positive correlations with the percentage of filled grains in maha and the number of grains per panicle in yala. Significant increases in the number of tillers per hill under elevated CO2 were responsible for its greater leaf area index and the greater numbers of panicles per hill. Crops under elevated CO2 accumulated biomass faster than those grown under ambient CO2 during the vegetative and grain‐filling stages. The results of this study demonstrate that elevated CO2 causes significant yield increases in rice, even when it is grown in warm, subhumid tropical climates.  相似文献

Objective

To evaluate the onset and duration of hematological changes and the use of Doppler ultrasound (spleen) in dogs sedated with acepromazine or xylazine.

Study design

Clinical study.

Animals

A total of 24 mixed breed dogs aged 1–4 years and weighing 15–25 kg.

Methods

Dogs were randomly distributed into two groups: acepromazine group (AG) which were administered acepromazine (0.05 mg kg?1) intramuscularly and xylazine group (XG) administered xylazine (0.5 mg kg?1) intramuscularly. Sonographic evaluations (morphologic and hemodynamic splenic vascularization) and hematologic tests were performed before drug administration (baseline) and 5, 15, 30, 60, 120, 240, 360, 480 and 720 minutes after drug administration.

Results

A significant reduction occurred in erythrogram variables in AG at 15–720 minutes corresponding with a significant enlargement of the spleen. In XG, a significant reduction was observed in the erythrogram variables at 30–60 minutes without a significant enlargement of the spleen. Hilar diameter did not change over time in either group. Flow alterations were found only in the splenic artery in AG, with a decreased final diastolic velocity observed at 60–120 minutes.

Conclusions

Administration of acepromazine resulted in decreased red blood cell count, hemoglobin, packed cell volume and an increased diameter of the spleen. Xylazine administration resulted in similar hematologic changes but of smaller magnitude and duration and without splenic changes. The absence of significant changes in the Doppler flow parameters of the splenic artery and vein and the hilar diameter suggests that the splenomegaly that was observed in AG was not due to splenic vasodilation. No splenic sequestration occurred after xylazine administration.

Clinical relevance

The results indicate that acepromazine decreases the erythrocyte concentrations by splenic erythrocyte sequestration and concomitant splenomegaly. Xylazine can cause slight hematologic changes, but without splenic changes.  相似文献

The purpose of this study was to evaluate different parameters of the immune status in the mare, during the follicular and the luteal phases of the oestrous cycle, in two consecutive years. Functional competence of peripheral blood neutrophils, such as chemotaxis, phagocytosis and oxidative burst was assessed under physiological cyclic conditions (Exp. I). In the second year of this study (Exp. II), besides peripheral blood neutrophil phagocytosis and oxidative burst analysis, circulating lymphocyte subsets were also characterized. The reproductive status in a total of 17 adult cycling mares was evaluated by ultrasonography and further confirmed by plasma progesterone levels. Chemotaxis tests were performed using porous membranes inserted in transwell chambers. Lipopolysaccharide (LPS) from Escherichia coli and N‐formyl‐Met‐Leu‐Phe (fMLP) were used as chemoattractants. Measurement of phagocytosis and oxidative burst in blood neutrophils were assessed by flow cytometry using commercially available kits. Quantification of T‐lymphocyte subsets was assessed by indirect immunofluorescence staining after incubation with monoclonal antibodies specific for CD2, CD3, CD4 and CD8 cell markers by flow cytometry. Natural killer cells and B cells were estimated mathematically. No significant difference was found in migration, phagocytosis and oxidative burst at either phase of the oestrous cycle. Statistical analysis of total white blood cell counts also showed no significant difference between either phase of the oestrous cycle, although there was a tendency for blood neutrophils to increase in number under the progesterone influence (p = 0.09). Lymphocytic subpopulations did not differ throughout the oestrous cycle. Overall, our results suggest that luteal and follicular phases in cycling mares may not influence the immune status of the mare.  相似文献

This study describes the implications of cardiac ventricular microscopy in Chelonia mydas relating to its ability to dive. For this work, 11 specimens of the marine turtle species C. mydas found dead on the coast of Rio Grande do Norte (Northeast Brazil) were used. After necropsy, fragments of the cardiac ventricular wall were fixed in 10% buffered formaldehyde solution for 24 h and then subjected to routine processing for light and scanning electron microscopy (SEM). The ventricle in this species is formed by the epicardium, myocardium and endocardium. The subepicardial layer consists of highly vascularised connective tissue that emits septa to reinforce the myocardium surface. There is an abundant and diffuse subepicardial nerve plexus shown by immunostaining technique. The thickness of the spongy myocardium and the nature of its trabeculae varied between the heart chambers. The endocardium shows no characteristic elements of the heart conduction system. The valves have a hyaline cartilage skeleton, coated by dense irregular connective tissues characterised by elastic fibres. These findings in the green turtle ventricular microscopy are related to hypoxia resistance during diving.  相似文献

Schinus terebinthifolius Raddi (Anacardiaceae) and Rapanea ferruginea (Ruiz & Pavon) Mez (Myrsinaceae) are two neotropical pioneer trees with wide geographical distribution in South America, highly degree of adaptation to different soil conditions and intense regeneration in areas with anthropic activities. With the aim to recommend the use Schinus and Rapanea in gallery forest restoration programs, we conducted an experiment with the objective to analyze the capacity of these two pioneer trees to tolerate soil flooding, mainly by accessing the effects of flooding on leaf gas exchange, growth and dry matter partitioning. Seedling survival throughout the 56-day flooding period were 100 and 90% for Schinus and Rapanea, respectively. The mean values of stomatal conductance (gs) and net photosynthesis (A) observed in the control seedlings were, respectively, 0.4 mol m–2s–1 and 14 mmolm–2s–1, for Schinus, and 0.5 mol m–2s–1 and 14 mmolm–2s–1, for Rapanea. On day 20 flooding reduced gs and A by 36 and 29% in Schinus, and 81 and 61% in Rapanea. At the end of the experiment, significant decreases were also observed for root and whole plant biomass, in both species. Based on the results, we concluded that seedlings of Schinus and Rapanea can survive and grow throughout a medium period of soil waterlogging, in spite of the alterations observed in their physiological behavior, such as the decreases in stomatal conductance and in whole plant biomass.  相似文献
