In future, prolonged summer drought and heat will constitute a major risk for the cultivation of shallow‐rooting beech in Central Europe and will negatively affect the productivity of beech forests. In a pot experiment under controlled conditions, the influence of long‐term (28 d) water deprivation on nitrogen (N), carbon (C), phosphate (Pi), and ascorbate (ASC) concentrations was examined in leaves and fine roots of beech seedlings (Fagus sylvatica L.) from six provenances originating from Central Europe (Germany: Neidenstein and Illertissen, intermediate habitats), the Balkan peninsula (Croatia: Zagreb and Gospic, wet habitats), and Southeast Europe (Bulgaria: Kotel, Greece: Paikos; dry habitats). The goal of the study was to identify beech provenances well adapted to water limitation during summer drought events. Our results suggest that N might be involved in the alleviation of water scarcity, whereas Pi might become a limiting factor for forest growth during drought periods. Drought stress resulted in significant changes of ASC pools in leaves and fine roots and the ASC redox state. Under well‐watered and under drought conditions, ASC in leaves was the most important factor causing differences between the provenances examined. Finally, a link between P nutrition and the capacity of antioxidative stress defense by ascorbate could be highlighted. Based on observations from this study, beech seedlings from three origins (Paikos, Zagreb, and Neidenstein) might constitute beech provenances well adapted to water shortage in summer. This conclusion is drawn from the high potential of these provenances to alleviate oxidative stress during water shortage.  相似文献

Anaplasma (A.) phagocytophilum is a tick-transmitted obligate intracellular bacterium and has been identified in a wide range of mammalian species, causing febrile disease in some. Few reports show that it can also cause granulocytic anaplasmosis in cats. As data on the occurrence of A. phagocytophilum in cats from Germany is limited, a total of 326 serum and 306 EDTA-blood samples from cats from Germany were screened by direct (Giemsa-stained blood/buffy coat smears, real-time PCR) and indirect (IFAT) methods. Of 274 Giemsa-stained blood smears which could be evaluated none was positive for morulae, but one blood sample (< or =0.1%; 1/306) was positive for A. < or = phagocytophilum-DNA in PCR. Antibodies (cutoff > or = 1:64) were detected in 53 out of 326 samples (16.2%). Altogether, the results show a high seroprevalence rate of anti-A. phagocytophilum antibodies in cats in Germany while the low detection rate of this bacterial agent by direct methods is similar to those of other studies on A. phagocytophilum infections in cats.  相似文献

Bovine neonatal pancytopenia (BNP) is mainly characterized by multiple haemorrhages, thrombocytopenia and leukocytopenia as a result of bone marrow depletion. BNP can be induced in healthy calves through application of colostrum from BNP donors, proofing that BNP is mediated to maternal alloantibodies. Alloantibody binding to bovine blood cells is present in sera and colostra of BNP donors and is probably initialized by vaccination with a certain BVD vaccine. To understand etiology and pathomechanisms of BNP, we closely characterized disease inducing antibodies regarding immunoglobulin subclass and binding specificities to peripheral blood derived leukocytes and platelets. By exact phenotyping the targeted blood cell subsets, including platelets for the first time, we investigated that BNP alloantibodies are exclusively of IgG1 subclass. Interestingly, IgG1 of BNP colostra bound to 70% leukocytes and 100% platelets irrespective of different bovine breeds and cellular maturity of all specimens tested. Furthermore, staining pattern on platelets as well as leukocyte subsets by BNP-IgG1 alloantibody exposed 100% reactivity to platelets, granulocytes and monocytes. Interestingly, the main part of T-helper cells was not bound by colostral alloantibodies. Our results point to a crucial role of IgG1 antibodies in BNP and to a target antigen that is expressed by all cells of myeloid lineage, but only partially by the lymphoid lineage.  相似文献

Dry matter (DM) production of multispecies swards compared to perennial ryegrass (Lolium perenne; PRG) swards under intensive grazing warrants investigation as it is relatively unknown. A 5 × 2 factorial experiment, with five sward types and two defoliation methods, was used to investigate the effect of grazing versus cutting on dry matter (DM) production of multispecies and PRG‐only swards. Five sward types were established namely: a PRG‐only sward, receiving 250 kg N ha?1 year?1 (PRG250), and a PRG‐only sward (PRG90), a two‐species sward with PRG and white clover (Trifolium repens; PRGWC), a six‐species sward (6S) and a nine‐species sward (9S), each receiving 90 kg N ha?1 year?1. Cutting plots measured 1.95 × 10 m and grazing plots measured 10 × 10 m. All plots were harvested concurrently every 21–30 days from April‐November for two years (2015 and 2016). A strip from the grazing plots was cut for DM yield determination prior to turning in cattle for grazing. There was an interaction between sward type and defoliation method (p < .01), whereby there was no effect of defoliation method on the PRG‐only swards, however, the annual DM production of PRGWC, 6S and 9S swards reduced under grazing compared to cutting (p < .001; on average 1,929 kg DM/ha lower). Multispecies swards had lower DM production under grazing compared to cutting, while the DM yield of PRG‐only swards was unaffected by defoliation method.  相似文献

Single-ascospore-derived isolates ofMycosphaerella fijiensis Morelet from false horn ‘Agbagba’ plantain leaves obtained from five different villages in southern Nigeria were stimulated to mate under artificial conditions. Pairs of isolates were incubated under blacklight on potato dextrose agar (PDA) with surface-sterilized plantain leaves or on PDA with autoclaved plantain leaves. Some isolates were observed to be sexually compatible by their ability to produce spherical to bulb-shaped fruiting body structures (FBS) and ascospores on pairing. FBS were observed to measure between 39–65 μm (smallest diameter) and 39–104 μm (largest diameter; mean 55.3×71.1 μm) in diameter, whereas ascospore lengths measured between 13.0 and 14.9 μm. Length of incubation time required for FBS production was dependent on the pair of isolates involved, the average being 40.1 days. Hyperthyroidism can increase the renal excretion of magnesium and thus cause hypomagnesemia in various species. Anaerobically collected blood samples from 15 hyperthyroid and 40 normal, healthy cats were analyzed with an ion-selective electrode analyzer and a serum biochemical analyzer. There was no significant difference in ionized or total serum magnesium concentration between the 2 groups, but there was a significant difference (P = 0.004) in the ratio of ionized to total serum magnesium concentrations between the healthy cats and the hyperthyroid cats with thyroxine (T4) concentrations at or above the median. There was a significant correlation (r = 0.894, P = 0.000) between the ionized and total magnesium concentrations in the hyperthyroid cats. The hyperthyroid cats had a significantly lower (P = 0.003) total serum protein concentration than the healthy cats. A significant negative correlation (r = -0.670, P = 0.006) was detected between the ionized magnesium and logarithmically transformed total T4 concentrations in the hyperthyroid cats, which suggests that the severity of hyperthyroidism may contribute to a decrease in the ionized magnesium concentration.  相似文献

We explored the applicability of the 13C bicarbonate dilution technique for determination of energy expenditure (EE) in young bulls in comparison to whole body indirect calorimetry (IC). Twelve bulls of a F2 German Holstein x Charolais cross (4.5 months, 332 ± 16 kg BM) received a diet providing 1000 kJ ME d− 1 kg BM− 0.75 and 4.3 g crude protein d− 1 kg BM− 0.75. Bulls were housed in respiration chambers and received an intravenous bolus of NaH13CO3 (A: 3 μmol kg BM− 1 (n = 2), B: 7 μmol kg BM− 1 (n = 4), C: 17.5 μmol kg BM− 1 (n = 6), 99 at.% 13C) into the jugular vein to measure EE. Simultaneously, EE was determined by IC. After the 13C administration blood samples and breath gas were collected from the animals in the respiration chamber during a 24-h period (7.00–7.00 h). The recovery of 13C in breath CO2 (% of 13C dose) was irrespective of NaH13CO3 dose (A: 69.7 ± 2.7%, B: 70.5 ± 4.5%, C: 75.0 ± 4.9%; P > 0.05). Only small amounts of 13C were excreted in urine (3.4 ± 2.6%) and feces (2.0 ± 1.3%). The EE determined by the 13C bicarbonate method using breath and blood 13C recovery rates as correction factors was not different from that measured by IC (816 ± 81 [blood] or 827 ± 101 [breath] vs. 820 ± 90 kJ d− 1 kg BM− 0.75). Bland–Altman analysis showed a 95% confidence interval for EE of ± 99 and ± 109 kJ d− 1 kg BM− 0.75 based on blood and breath 13C recovery, respectively. In conclusion, the 13C bicarbonate dilution method is appropriate to obtain reliable estimates of EE in young bulls using blood CO2 or breath CO2 under standardized experimental conditions, i.e. in the fasting state.  相似文献
