Any mammalian spermatozoon that achieves successful in vivo fertilization has to perform almost perfectly in many disparate functions and overcome a series of obstacles imposed by the female reproductive tract. This implies that during formation in the testis and epididymis, the spermatozoa did not incur any morphological, metabolic, immunological or genetic abnormalities. Given that the spermatozoa are such highly differentiated cells, this means that every cellular compartment must not only be intact but must also respond appropriately to intracellular and extracellular signals. Assuming that a spermatozoon possesses this level of perfection, and is able to reach and penetrate an oocyte, it can only be regarded as 'fertile' if the DNA it carries is intact and able to sustain embryonic development. Although the proportion of inseminated spermatozoa that meet these criteria is vanishingly small, the female reproductive tract applies stringent selection criteria during sperm transport and, as a result, the probability of conception around the time of ovulation is very high. If laboratory tests of semen quality could approach the efficacy of the female reproductive tract, it would be possible to predict the odds of spermatozoa meeting the egg; however, this is not possible at present. In this article, I suggest a simple model to illustrate how a battery of laboratory tests could eventually be used to make these predictions.  相似文献

The schistosome Heterobilharzia americana infects dogs, raccoons, and other mammals in the southeastern United States. Migration of eggs into the liver results in parasitic granulomas with varying degrees of fibrosis and inflammation. Recently, hepatic parasitic granulomas in horses were shown to be caused by H. americana infection. In the present study, samples of liver from 11 of 12 horses with hepatic granulomas identified at necropsy (n = 11) or surgical biopsy (n = 1) were used for DNA extraction, polymerase chain reaction amplification and sequencing using primers specific for a portion of the H. americana small subunit ribosomal RNA gene. A polymerase chain reaction amplicon of the correct size was produced from the extracted DNA in 8 of the 11 horses. Amplicons from 5 of the 8 positive horses were sequenced and had 100% identity with H. americana. In all but 2 of the 12 horses, Heterobilharzia was not responsible for the primary clinical disease, and the hepatic granulomas were considered an incidental finding.  相似文献

A 4-year-old, castrated, male domestic ferret (Mustela putorius furo) was evaluated for a possible upper respiratory tract infection and wheezing for approximately 3 weeks. Supportive care for a suspected upper respiratory tract infection was initiated; however, the patient became progressively more dyspneic over the next 48 hours. A possible tracheal mass was identified upon thoracic radiographic imaging of the ferret. Tracheoscopy and a computed tomography scan of the neck and thorax confirmed the presence of a tracheal mass approximately 3 cm from the larynx obstructing 80% of the tracheal lumen. Rather than pursuing surgical resection, the owners elected humane euthanasia. An adenosquamous carcinoma of the trachea was diagnosed after histologic analysis. This tumor type has been diagnosed in the lungs of ferrets that were experimentally exposed to carcinogens found in cigarette smoke; however, there are no previous reports of this or any other tumor type associated with the ferret trachea. A tracheal mass causing a partial upper airway obstruction should be considered within the differential disease diagnosis list when a ferret is presented with dyspnea.  相似文献
