Aeromonas hydrophila is known to be causative agent of an infection named as Bacterial haemorrhagic septicaemia or red pest in freshwater fish. The aim of this study was to develop and validate the glycoprotein‐based fish vaccine against Aeromonas hydrophila. For this aim, after identification and characterization of A. hydrophila isolates from fish farms, one A. hydrophila isolate was selected as vaccine strain. Antigenic glycoproteins of this vaccine strain were determined by Western blotting and glycan detection kit. The connection types of these glycoproteins were examined by glycoprotein differentiation kit. Two glycoproteins, molecular weights of 19 and 38 kDa, with SNA connection type were selected for use in vaccination trials. After their purification by SNA‐specific lectin and size‐exclusion chromatography, protection studies with purified proteins were performed. For challenge trials, four experimental fish groups were designated: Group I (with montanide), Group II (with montanide and ginseng), Group III [with Al(OH)3] and Group IV [with Al(OH)3 and ginseng]. The survival ratings of fish were determined, and protection was calculated as 21.56%, 29.41%, 69.83% and 78.88% in groups I, II, III and IV, respectively. In conclusion, A. hydrophila glycoproteins with Al(OH)3 and ginseng could be used as a safe and effective vaccine for fish. 相似文献

Many fish species undergo natural starvation periods. Adaptation to starvation is possible through the activation of behavioral, biochemical and physiological mechanisms. Knowledge of the effect of dietary nutrients on the intermediary metabolism during starvation and refeeding can be useful to improve fish health and optimize aquaculture production. To analyze the effect of dietary nutrients on liver metabolism of Siberian sturgeon (Acipenser baerii) submitted to starvation and refeeding, four isoenergetic diets differing in nutrient composition were designed: LP-St (38 % protein, 12 % lipid, 36 % carbohydrate), HP-St (44 % protein, 10 % lipid, 30 % carbohydrate), LP-L (38 % protein, 18 % lipid, 25 % carbohydrate) and HP-L (44 % protein, 16 % lipid, 22 % carbohydrate). Four groups of fish were fed 3 weeks to satiety with the corresponding diet, starved for 2 weeks and then refeed 5 weeks to satiety on the same diet. Starvation mobilized the hepatic lipid store to a greater extent than glycogen. Starvation increased superoxide dismutase activity irrespective of the diet, while low protein diets (LP-St and LP-L) increased catalase activity. The oxidative damage decreased after 5 weeks of refeeding. Refeeding the starved fish on the HP-St diet promoted the greatest growth performance. In addition to reporting for the first time the effect of diet composition on growth, liver composition and antioxidant activities in Siberian sturgeon submitted to starvation and refeeding, our findings suggest that refeeding on HP-St diet stimulated the use of dietary carbohydrates and allowed a protein sparing effect in Siberian sturgeon. 相似文献

Edwardsiella tarda has long been known as a pathogen that causes severe economic losses in aquaculture industry. Insights gained on E. tarda pathogenesis may prove useful in the development of new methods for the treatment of infections as well as preventive measures against future outbreaks. In this report, we have established the correlation between the presence of virulence genes, related with three aspects typically involved in bacterial pathogenesis (chondroitinase activity, quorum sensing and siderophore‐mediated ferric uptake systems), in the genome of E. tarda strains isolated from turbot in Europe and their phenotypic traits. A total of 8 genes were tested by PCR for their presence in 73 E. tarda isolates. High homogeneity was observed in the presence/absence pattern of all the strains. Positive results in the amplification of virulence‐related genes were correlated with the detection of chondroitinase activity in agar plates, in vivo AHL production during fish infection and determination of type of siderophore produced by E. tarda. To the best of our knowledge, this is the first study carried out with European strains on potential virulence factors. Furthermore, we demonstrated for the first time that E. tarda produces the siderophore vibrioferrin. 相似文献

Protein degradability in forage legumes is of global importance because utilization efficiency of forage has economic and environmental consequences. However, there are no published studies on the effect of legume stand structure on differences in crude protein (CP) fractions. The main objective of the present research was therefore to investigate differences in CP fractions in leaves and stems of lucerne (Medicago sativa L.) during the growing season. Stand traits were measured over 2 years, and forage was sampled at the early bud and early flower stages in the first, second and third cuts. Stems had significantly higher concentrations (in g kg?1 CP) of non‐protein (fraction A: 430 g kg?1 CP) and indigestible nitrogen (fraction C: 92 g kg?1 CP) than leaves and had lower relative content of true protein (fraction B: 478 g kg?1 CP). In the total forage (stems and leaves combined), about 80% of the variation in CP fractions was explained by year, cut and maturity. Year was the most important factor, particularly for the B fractions. Cut was the second‐most important factor; its main effect was that the relative abundance of fraction A declined from 394 g kg?1 CP in the first cut to 293 in g kg?1 CP the third cut. Maturity increased the amounts of indigestible fraction C and protein fractions B1 and B3. This was associated with the leaf weight ratio, which had an inverse relationship with maximal stem length and dry matter yield. Variation partitioning showed that 75% of CP fraction variability associated with cut, maturity and year could be explained by the evaluated stand traits. Dutch elm disease (DED) spread across Europe and North America in the 20th century killing most natural elm populations. Today, breeding programmes aim at identifying, propagating and studying elm clones resistant to DED. Here, we have compared the physiology and biochemistry of six genotypes of Ulmus minor of variable DED resistance. Leaf gas exchange, water potential, stem hydraulic conductivity and biochemical status were studied in 5‐year‐old trees of AB‐AM2.4, M‐DV2.3, M‐DV2 × M‐CC1.5 and M‐DV1 and 6‐year‐old trees of VA‐AP38 and BU‐FL7 before and after inoculation with Ophiostoma novo‐ulmi. Leaf water potential and net photosynthesis rates declined, while the percentage loss of hydraulic conductivity (PLC) increased after the inoculation in susceptible trees. By the 21st day, leaf predawn and midday water potential, stomatal conductance to water vapour and net photosynthesis rates were lower, and PLC was higher in trees of susceptible (S) genotypes inoculated with the pathogen than in control trees inoculated with water, whereas no significant treatment effect was observed on these variables in the resistant (R) genotypes. Fourier transform infrared spectroscopy analyses revealed a different biochemical profile for branches of R and S clones. R clones showed higher absorption peaks that could be assigned to phenolic compounds, saturated hydrocarbons, cellulose and hemicellulose than S clones. The differences were more marked at the end of the experiment than at the beginning, suggesting that R and S clones responded differently to the inevitable wounding from inoculation and repeated sampling over the experimental course. We hypothesize that a weak activation of the defence system in response to experimental wounding can contribute to the susceptibility of some genotypes to O. novo‐ulmi. In turn, the decline in shoot hydraulic conductivity and leaf carbon uptake caused by the infection further exacerbates tree susceptibility to the fungus. 相似文献

Screening brassicaceous plants as biofumigants for management of Phytophthora cinnamomi oak disease

Brassicaceous plants rich in glucosinolates have been used as biofumigants for the management of soilborne pathogens. Efficacy of Brassica plant tissue has mainly been attributed to toxic isothiocyanates released upon the hydrolysis of glucosinolates. Management of Phytophthora cinnamomi, the causal agent of oak root rot in rangeland ecosystems using biofumigation, is promising, but requires further validation. The biofumigation activity of 14 brassicaceous plants was evaluated under experimental conditions. All evaluated plants rich in sinigrin suppressed (100%) the mycelial growth of P. cinnamomi, while plants rich in aromatic or other aliphatic glucosinolates had little or no suppressive effect. Simulating soil amendment in field conditions, the effects on natural soil artificially infested with P. cinnamomi chlamydospores were examined with Brassica juncea, Eruca vesicaria and Lepidium sativum, three species with different glucosinolate profiles. Only B. juncea decreased the viability of chlamydospores significantly in comparison with untreated soil only 1 day after biofumigation, whereas E. vesicaria needed 8 days to reach significance and L. sativum had no effect at all. Despite the decreases in soil inoculum, biofumigation with B. juncea did not prevent the root infections in a highly susceptible host (Lupinus luteus). However, biofumigation with plants rich in sinigrin, such as B. juncea, decreased P. cinnamomi soil inoculum under the experimental minimum threshold for oak disease expression. Although biofumigation should be considered as an effective measure to be incorporated in integrated control of the oak disease, biofumigation by itself would not be effective enough for the substantial suppression of P. cinnamomi inoculum. 相似文献
