AIMS: To develop a challenge model for Salmonella Brandenburg infection in pregnant ewes. To compare efficacies of a live attenuated Salmonella Typhimurium mutant, a subunit preparation from a virulent S. Brandenburg isolate, and a commercial multivalent inactivated vaccine in their ability to prevent experimental S. Brandenburg infection. To assess the efficacy of the live attenuated S. Typhimurium mutant against natural S. Brandenburg infection in lambs.

METHODS: Two-year-old ewes were immunised with either a live attenuated vaccine (eye-drop; n=20), a subunit vaccine (n=20) or an inactivated bacterin vaccine (n=20), both administered subcutaneously, or served as unvaccinated controls (n=21). Four weeks later, the sensitising regime was repeated as a booster vaccination, and the ewes were challenged 6 weeks later with a virulent S. Brandenburg isolate, approximately 6 weeks prior to lambing. The presence of clinical signs, abortion or death was noted following challenge. The presence and number of Salmonella spp in faecal samples taken throughout the trial, and in organs collected post mortem, were determined using an enrichment selection procedure, and confirmed by serology and pulsed-field gel electrophoresis (PFGE). Half of the surviving lambs were vaccinated with the live attenuated vaccine and all (n=39) were exposed to natural infection from contaminated pasture.

RESULTS: There was no significant protection against mortality and abortion following vaccination with the live attenuated, subunit and inactivated vaccines following experimental challenge with S. Brandenburg. There was a significant but transient decrease in the number of ewes shedding S. Brandenburg (live attenuated, p=0.05; subunit, p=0.05; inactivated, p=0.01), and in the quantity of these bacteria in the sheep from the vaccinated groups (p<0.05) compared with controls, 6 weeks after challenge. Lambs from the challenged ewes did not shed Salmonella spp after being vaccinated with the live attenuated vaccine, in contrast to some of the controls, when grazed on pasture contaminated with S. Brandenburg.

CONCLUSIONS: The use of live attenuated, subunit and inactivated vaccines did not significantly protect sheep against lethal experimental challenge with S. Brandenburg.  相似文献

Superficial stromal keratitis or pannus is a syndrome of corneal, conjunctival and third eyelid inflammation. Superficial stromal keratitis mainly presents as a subepithelial corneal infiltration of vascular connective tissue, and usually arises from the lateral (temporal) limbal area. In some dogs perilimbal hyperaemia and third eyelid blepharitis can be present without corneal involvement. The most commonly affected breed of dog is the German Shepherd. Most cases of superficial stromal keratitis can be controlled with topical corticosteroids, and only rarely is cryosurgery or superficial keratectomy required to remove excessive pigment and or granulation tissue. The precise aetiology of SSK is unknown, but is likely to be multifactorial, with sunlight being a significant factor. Corneal lipidosis and keratoconjunctivitis sicca can occur secondary to superficial stromal keratitis.  相似文献

Faecal egg count reduction tests and an anthelmintic efficiency assay were used to assess the efficacy of combinations of albendazole sulphoxide and levamisole against populations of Ostertagia and Trichostrongylus sp. which contained different proportions of worms resistant to both benzimidazole and levamisole anthelmintics. Compared to the effects of either drug alone, significantly greater efficacy was obtained using combinations which included dose rates similar to those recommended for the separate components. At these dose rates, the mixtures reduced mean faecal egg counts by 95% or more, and caused a reduction of 68% in adult Ostertagia sp. and more than 95% for 4th stage Ostertagia and T colubriformis. The increased efficacy of the mixtures could be accounted for by actions of the drugs acting independently.  相似文献

The purpose of this article is to review the most relevant features of the physiology, clinical signs, diagnosis, treatment and prevention of canine pseudocyesis (PSC). This is a physiological syndrome, characterized by clinical signs such as: nesting, weight gain, mammary enlargement, lactation and maternal behaviour, which appears in non‐pregnant bitches at the end of metaoestrus. PSC is a frequent finding in domestic dogs. Although it is generally admitted that prolactin (PRL) plays a central role in the appearance of PSC, its precise aetiophysiology is not completely understood yet. A number of clinical studies suggest that at some point of metaoestrus circulating PRL levels rise in overtly pseudopregnant bitches. Individual differences in sensitivity to PRL as well as the existence of molecular variants of canine PRL with different bioactivity versus immunoreactivity ratios may help clarify the aetiopathology of PSC. Diagnosis of PSC is based on the presence of typical clinical signs in metaoestrous non‐pregnant bitches. Considering that PSC is a self limiting physiological state, mild cases usually need no treatment. Discouraging maternal behaviour and sometimes fitting Elizabethan collars to prevent licking of the mammary glands may suffice in these cases. Sex steroids (oestrogens, progestins and androgens) have been traditionally used to treat PSC but the side‐effects usually outweigh the benefits of these medications. Inhibition of PRL release by ergot derivatives [ bromocriptine (10–100 μg/kg per day for 10–14 days], cabergoline (5 μg/kg per day during 5–10 days), metergoline (0.2 mg/kg per day during 8–10 days) has proved to be effective for the treatment of canine PSC. Although some of these ergot derivatives present some untoward side‐effects, they are transient and can usually be managed. Predisposed bitches not intended for breeding should be spayed as ovariectomy is the only permanent preventive measure.  相似文献

Ovariectomized ewes (n=22; 68.76+/-2.34 kg initial body weight; 2.9+/-0.1 initial body condition score) were individually fed one of three diets: (1) control (phytoestrogen-free; n=7), (2) flax containing diet (n=8), or (3) linseed meal (LSM) containing diet (n=7) to investigate the rate of progesterone (P4) clearance. On day 20 of feeding (day 0=initiation of treatment), a P4 releasing device (CIDR) was placed in the vagina and jugular blood samples were obtained prior to CIDR insertion and 15, 30, 60, and 120 min following CIDR insertion. Further, blood samples were obtained daily between days 21 and 24. On day 25, blood samples were retrieved prior to CIDR removal and 2, 5, 10, 15, 30, 60, 120, and 360 min following CIDR removal. There was no difference in initial or final body weight or body condition score and there were no time by diet interactions on P4 clearance. The fractional rate of P4 uptake measured prior to CIDR insertion through day 4 following insertion tended to be greater (P=0.07) in LSM fed ewes (508.75+/-71.37%/min) compared to flax (295.39+/-66.76%/min) and control fed (287.54+/-71.37%/min) ewes. Diet tended (P=0.10) to influence P4 clearance rate when measured from prior to CIDR removal through 120 min following CIDR removal with LSM fed ewes having a greater (1.26+/-0.2) fractional rate constant than flax (0.929+/-0.09) and control fed (0.922+/-0.09) ewes. Flax fed ewes also had more (P<0.01) omega-3 fatty acids and total fatty acids in plasma. Reports of increased pregnancy rates in dairy cows fed flax may relate to P4 metabolism.  相似文献

The aim of this study was to compare fecal microbiome, plasma, fecal and urine metabolomes, and serum biochemistry of adult companion dogs according to body condition scores. Blood, serum/plasma, urine, and fecal samples were collected from 66 clinically healthy, adult companion dogs of either normal weight (NW), overweight (OW), or obese dogs (OB). analyses included fecal microbiome analyses via 16S ribosomal RNA gene amplicon; sequencing, nontargeted plasma, fecal, and urine metabolomics using liquid chromatography/gas chromatography-mass; spectrometry, and serum biochemistry for each dog. Few significant differences in serum biochemistry and fecal microbiome Operational Taxonomic Unit (OTU) were found between weight groups and there was high OTU variation between individual dogs. NW dogs had higher relative abundance of the genus Eubacterium (log-fold change 4.3, adjusted P value?=?.003) and lower relative abundance of the family Bifidobacteriaceae (log-fold change ?3.6, adjusted P value?=?.02) compared to OB dogs. The microbiome of NW dogs had higher OTU richness compared with OB dogs. Metabolome analysis showed 185 plasma, 37 fecal, and 45 urine metabolites that significantly differed between NW and OW or OB dogs. There were notable significant differences in relative abundance of several plasma phospholipid moieties and fecal volatile fatty acids between weight phenotypes. The combinations of host and gut microbiota and metabolic shifts suggest a pattern that could help detection of early metabolic changes in overweight dogs before the development of obesity related disease. The results of this study support the need for continued investigation into sensitive measures of metabolic aberrancies in overweight dogs.  相似文献

In New Zealand the fungus Pithomyces charturum normally produces sporidesmin, a mycotoxin, which is responsible for the hepatogenous photosensitisation disease known as facial eczema. Cultures from an isolate of P. charturum, which does not produce sporidesmin, were examined by cell culture and by dosing to lambs to determine whether other toxic metabolites were produced. Acute and long term toxicity studies were conducted with the toxic response being assessed by weight changes, postmortem and histological examination of tissues, blood biochemistry and haematology tests. An extract from a sporidesmin-producing isolate was highly toxic in cell culture, while extracts of the nonsporidesmin-producing isolate did not cause a cytotoxic response to HEp 2 cells. After dosing with a sporidesmin-producing isolate, lambs developed liver lesions and clinical signs of facial eczema. Serum biochemistry changes occurred which were consistent with sporidesmin poisoning. Lambs dosed with the nonsporidesmin-producing isolate, at the rate of thirty times the number of spores of the sporidesmin-producing isolate, showed no observable toxic effects. All organs were of normal appearance, and histological examination of tissues, blood biochemistry and haematology results showed no abnormal changes. Similarly, long term dosing of extracts of the nonsporidesmin-producing isolate, at a rate equivalent to 100,000 spores/g of grass, produced no indication of a toxic response. It was concluded that the nonsporidesmin-producing isolate of P. churtarum contained no toxic metabolites in significant concentration.  相似文献

Sporidesmin, a mycotoxin produced by some strains of Pithomyces chartarum, is responsible for the hepatogenous photosensitisation disease facial eczema, which causes severe losses in agricultural revenue in New Zealand. A sporidesmin-producing strain of P. chartarum, isolated in New Zealand, was grown in co-culture with a South African strain that does not produce the mycotoxin. Competition occurred between the two strains when grown both on agar plates and on dried ryegrass, with a significant decrease in the total amount of sporidesmin produced. Biological control of toxigenic P. chartarum can thus occur under laboratory conditions, raising the possibility of similar control in the field situation.  相似文献
