To investigate the consequences of long-term N additions on soil CH4 dynamics, we measured in situ CH4 uptake rates, soil profiles and kinetics parameters during the growing season in a temperate deciduous forest in northwestern Pennsylvania (Allegheny College Bousson Environmental Forest). Measurements were made in control and adjacent plots amended with 100 kg N ha–1 year–1 for 8 years. We found that the in situ consumption rates were 0.19±0.02 (mean±SE) for the control and 0.12±0.01 mg CH4–C m–2 h–1 for the N treatment, indicating that consumption had been reduced by 35% after 8 years of N amendments. Despite the large difference in rates of consumption, there were no differences in the CH4 concentration profiles between the control and N-amended plots. Laboratory incubations of CH4 consumption throughout the soil column (organic horizon and mineral soil depths) showed that rates were greatest in the organic horizon of both control and N-amended soils, although consumption was reduced by 42% in the N-amended plot. However, the rate in the organic horizon was only about 50% the rate measured in organic horizons at other temperate forests. The apparent Km [Km(app)] value in the organic horizon of the control plot was fourfold less than the Km(app) value in the organic horizon of another temperate forest, but similar to the Km(app) values in adjacent plots amended with N for a decade. Unlike results for other temperate forests, Km(app) values at Bousson generally did not decrease with soil depth. These results indicate that N cycling strongly controls the CH4-consuming community, and suggest that alterations of the N cycle due to N deposition or addition may alter rates and the location of CH4 consumption by soils, even in soils with high N content and cycling rates.  相似文献

The objective of this study was to use synchrotron transmission FTIR microspectroscopy as a rapid, direct, and nondestructive analytical technique to reveal molecular microstructural-chemical features within tissue in grain barley. The results showed that synchrotron transmission FTIR microspectroscopy could provide spectral, chemical, and functional group characteristics of grain barley tissue at ultrahigh spatial resolutions. The spatially localized structural-chemical distributions of biological components (lignin, cellulose, protein, lipid, and carbohydrates) and biological component ratios could be imaged. Such information on molecular microstructural-chemical features within the tissue can be used for plant breeding programs for selecting superior varieties of barley for special purposes and for prediction of grain barley quality and nutritive value for humans and animals.  相似文献

Since 1999, Cryptococcus gattii has emerged as an important pathogen of humans and animals in southwestern British Columbia. Historically thought to be restricted to the tropics and subtropics, C. gattii has posed new diagnostic and treatment challenges to veterinary practitioners working within the recently identified endemic region. Clinical reports of canine and feline cryptococcosis caused by C. gattii diagnosed between January 1999 and December 2003 were included in this case series. The most common manifestations of disease were respiratory and central nervous system signs. Multivariate survival analysis revealed that the only significant predictor of mortality was the presence of central nervous system signs upon presentation or during therapy. Case fatality rates in both species were high. Further investigation into effective treatment regimes is warranted.  相似文献

This project investigated the use of ultrasonography at first diagnosis of presumptive early bovine respiratory disease (BRD) in feedlot cattle from western Canada. One hundred seventy-four cattle (116 cases and 58 controls) at high risk of developing BRD were enrolled in a prospective longitudinal study over 2 y (2006-2007). Cattle with clinical signs relating to the respiratory system and assessed as sick at the time of feedlot arrival (arrival fever cases) or assessed as sick in the pen 3 to 30 d post-arrival (post-arrival fever cases, post-arrival no fevers cases) were eligible for enrollment. Control animals were identified at the time of case enrollments. Ultrasonography was done using a 3.5 sector transducer at enrollment and at 2, 4, and 6 wk post-enrollment. Lung lesions were identified at least 1 time over the course of the trial in 32/116 (28%) cases and 9/58 (16%) controls. At enrollment, lung lesions were identified in 20/115 (17%) cases and 2/55 (4%) controls (data unreadable n = 4). Post-arrival fever cases (14/48) were the most likely to have a lesion identified using ultrasound. In arrival fever cases, average daily gain (enrollment to last ultrasound, average 34 d) was improved (P = 0.007) in cattle identified with a lesion at enrollment using ultrasound compared with those not identified with a lesion at that time, potentially demonstrating the effects of gut fill at arrival weighing, as these sicker animals may have eaten less prior to arrival and, therefore, had more room for improvement in weight over time due to restoration of normal gut fill. None of the ultrasound time points explored (enrollment, 2, 4, or 6 wk post-enrollment) were associated with the animal health outcomes of interest (subsequent treatment, chronicity, wastage, or mortality) for cattle enrolled at arrival or post-arrival.Ultrasonography using a 3.5 sector transducer was not particularly effective as a prognostic/diagnostic tool for early detection of BRD, but may be useful in targeted populations of animals with respiratory disease of longer duration (such as chronic pens).  相似文献

OBJECTIVE: To evaluate retention of porcine reproductive and respiratory syndrome virus (PRRSV) in houseflies for various time frames and temperatures. SAMPLE POPULATION: Fifteen 2-week-old pigs, two 10-week-old pigs, and laboratory-cultivated houseflies. PROCEDURE: In an initial experiment, houseflies were exposed to PRRSV; housed at 15 degrees, 20 degrees, 25 degrees, and 30 degrees C; and tested at various time points. In a second experiment to determine dynamics of virus retention, houseflies were exposed to PRRSV and housed under controlled field conditions for 48 hours. Changes in the percentage of PRRSV-positive flies and virus load per fly were assessed over time, and detection of infective virus at 48 hours after exposure was measured. Finally, in a third experiment, virus loads were measured in houseflies allowed to feed on blood, oropharyngeal washings, and nasal washings obtained from experimentally infected pigs. RESULTS: In experiment 1, PRRSV retention in houseflies was proportional to temperature. In the second experiment, the percentage of PRRSV-positive houseflies and virus load per fly decreased over time; however, infective PRRSV was found in houseflies 48 hours after exposure. In experiment 3, PRRSV was detected in houseflies allowed to feed on all 3 porcine body fluids. CONCLUSIONS AND CLINICAL RELEVANCE: For the conditions of this study, houseflies did not support PRRSV replication. Therefore, retention of PRRSV in houseflies appears to be a function of initial virus load after ingestion and environmental temperature. These factors may impact the risk of insect-borne spread of PRRSV among farms.  相似文献

OBJECTIVE: To determine whether cattle testing positive for Mycobacterium avium subsp paratuberculosis as determined by microbial culture of feces or antibody ELISA were more likely to have false-positive responses on the caudal fold tuberculin (CFT) test or interferon-gamma (IFN-gamma) assay for Mycobacterium bovis than cattle testing negative for M paratuberculosis. ANIMALS: 1043 cattle from 10 herds in Michigan. PROCEDURE: Feces and blood samples for plasma were collected from cattle > or =24 months old on the day the CFT test was read. Fecal samples were submitted for microbial culture for M paratuberculosis. Plasma samples were tested for antibody against M paratuberculosis, and IFN-gamma after stimulation with purified protein derivative tuberculin from M bovis or M avium. RESULTS: Of 1043 cattle, 180 (17.3%) had positive CFT test results (suspects) and 8 (0.8%) had positive IFN-gamma assay results after stimulation with purified protein derivative tuberculin from M bovis. Forty-five (4.3%) and 115 (11.0%) cattle tested positive for M paratuberculosis as determined by microbial culture of feces and antibody ELISA, respectively. Cattle with positive responses for M paratuberculosis appeared to have an increased likelihood of false-positive results on the CFT test, although this association was not significant. CONCLUSIONS AND CLINICAL RELEVANCE: No significant association was detected among cattle testing positive for M paratuberculosis as determined by microbial culture of feces and antibody ELISA and positive CFT test and IFN-gamma assay results for M bovis.  相似文献

The response of plant cell membranes to vinyl organophosphorus insecticides was studied by determining the release of intracellular materials as a measure of membrane permeability and the uptake of [1-14C]-α-aminoisobutyric acid as a measure of active transport. A pretreatment with chlorfenvinphos (2-chloro-1-(2,4-dichlorophenyl)-vinyl diethyl phosphate) at 0.4 mM or higher concentrations increased the leakage of cell contents from the tissues of pea, corn, and beet, but two other vinyl organophosphorus insecticides, tetrachlorvinphos (2-chloro-1-(2,4,5-trichlorophenyl)-vinyl diethyl phosphate) and phosphamidon (2-chloro-2-diethyl carbamoyl-1-methyl vinyl dimethyl phosphate), had no effect. Simultaneous addition of phospholipids, β-sitosterol, or Ca2+ inhibited in varying degrees the chlorfenvinphos-induced permeability, suggesting that the leakage of cell contents might be due to alteration in membrane structure.Chlorfenvinphos or tetrachlorvinphos at 0.1 mM or higher concentrations inhibited the uptake of α-aminoisobutyric acid. The degree of inhibition varied with different plant species. The inhibition was competitive and was not prevented by phospholipids. However, Ca2+ and other divalent cations were stimulatory to the uptake of α-aminoisobutyric acid, either in the presence or absence of chlorfenvinphos. Chlorfenvinphos also inhibited plant growth in tobacco callus and pea stem assays.From the differences in effective concentration, structural requirement, and interaction with phospholipids, it is suggested that chlorfenvinphos affected the membrane permeability and active transport by different mechanisms. These effects probably underlie its inhibitory action on plant growth.  相似文献

Context

With global change, microclimates become important refuges for temperature-sensitive, range-restricted organisms. In African savannas, woody vegetation on Macrotermes mounds create widely-dispersed microclimates significantly cooler than the surrounding matrix, which buffer against elevated temperatures at the finer scale of mounds, allowing species to persist at the landscape scale. Termite colonies cultivate symbiotic fungi to digest lignin, but the fungi require temperatures between 29 and 32 °C, which termites strive to maintain. Mound-associated vegetation is a hot-spot for elephant herbivory, so removal of woody species cover by elephants could influence mound-associated microclimates, impacting temperature regulation by termites.

Objectives

We explored the interaction between two prominent ecosystem engineers (termites and elephants) to ascertain whether elephant removal of mound woody cover affects (1) external mound-associated microclimate and (2) internal mound temperature.

Methods

We surveyed 44 mounds from three sites in Kruger National Park, South Africa, during an El Niño/Southern Oscillation-induced drought and heatwave, recording whether sub-canopy, external, mound-surface and internal mound temperatures varied with vegetation removal by elephant.

Results

Elephant damage to mound-associated vegetation reduces the fine-scale microclimate effect provided by vegetation on Macrotermes mounds. Despite this, termites were able to regulate internal mound temperatures, whereas internal temperatures of abandoned mounds increased with elevated surface temperatures.

Conclusions

Termites can persist despite loss of mound-associated microclimates, but the loss likely increases energetic costs of mound thermoregulation. Since mound vegetation buffers against drought, loss of widely-dispersed, fine-scale microclimates could increase as megaherbivores remain constrained to protected areas, impacting climate-sensitive organisms and ecosystem function at a range of scales.
