Day-old broiler chicks were fed a calcium-deficient diet for 30 days. Gross, microscopic, and radiographic examinations of the proximal tibiotarsus were done at 2, 3, and 4 weeks of age. In 2-week-old chicks, the growth-plate proliferating-prehypertrophied zone (P-PHZ) was variably lengthened and disorganized, cartilage columns of the degenerating hypertrophied zone and metaphyseal primary spongiosa were shortened, and bone spicules of the secondary spongiosa were bordered by increased osteoid. In 3-week-old chicks, lesions were either similar or more pronounced, with the additional finding of metaphyseal peritrabecular fibrosis. In 4-week-old chicks, the P-PHZ decreased in relative length, osteoid seams and fibrous connective tissue were less prominent, and tibial dyschondroplasia-like lesions were present. Results demonstrated that dietary calcium deficiency produced rachitic lesions, that the lesions were very different from those of phosphorus deficiency or calcium excess, and that the lesions were variable over time. 相似文献

Efficiently controlling soil water content with irrigation is essential for water conservation and often improves potato yield. Volumetric soil water content (θv) in relation to irrigation, plant uptake, and yield in potato hills and replicated plots was studied to evaluate four water management options. Measurements of θv using a hammer driven probe were used to derive a θv index representing the relative θv status of replicated plots positioned along a hill slope. Time series for θv were determined using time domain reflectometry (TDR) probes at 5 and 15 cm depths at the center, shoulder, and furrow locations in potato hills. Sap flow was determined using flow collars in replicated field plots for four treatments: un-irrigated, sprinkler, surface drip, and sub-surface drip irrigation (40 cm depth). Irrigated yields were high/low as the θv index was low/high suggesting θv excess was a production problem in the wetter portions of the study area. The diurnal pattern of sap flow was reflected in the θv fluctuation it induces at hill locations with appreciable uptake. Hill locations with higher plant uptake were drier as was the case for the 5 cm (dry) depth relative to the 15 cm (wet) depth and for locations in the hill (dry) relative to the furrow (wet). The surface drip system had the lowest water use requirement because it delivers water directly to the hill locations where uptake is greatest. The sub-surface drip system wetted the hill gradually (1-2 days). Measurement of the θv index prior to experimental establishment could improve future experimental design for treatment comparisons. 相似文献

Mycoplasma gallisepticum was isolated from several turkey flocks at different locations in the United States that were clinically affected with respiratory disease. Five of these isolates from four series of outbreaks had patterns similar to the 6/85 vaccine strain of M. gallisepticum by random amplified polymorphic DNA (RAPD) analysis using three different primer sets, whereas with a fourth primer set (OPA13 and OPA14), only two of the isolates were similar to 6/85. Results obtained by sequencing portions of the pvpA, gapA, and mgc2 genes and an uncharacterized surface lipoprotein gene indicated that the field isolates had DNA sequences that ranged from 97.6% to 100%, similar to the 6/85 results. In some of the outbreaks there was an indirect association with the presence of commercial layers in the area that had been vaccinated with this vaccine strain, but there was no known close association with vaccinated birds in any of the outbreaks. Turkeys were challenged with two of the field isolates and with 6/85 vaccine strain. Turkeys challenged with the field isolates developed respiratory disease with airsacculitis and a typical M. gallisepticum antibody response, whereas birds challenged with 6/85 developed no respiratory signs or lesions and developed only a weak antibody response. Although these isolates were very similar to the 6/85 vaccine strain, it was not possible to prove that they originated from the vaccine strain-it is possible that they could be naturally occurring field isolates. 相似文献

Gnotobiotic pigs were dosed orally with Campylobacter sputorum subspecies mucosalis, either alone, or combined with rotavirus or non-pathogenic Escherichia coli and Streptococcus bovis to study the behaviour of C s mucosalis in defined conditions, to assess intracellular parasitism of enterocytes by C s mucosalis, and if possible to establish an experimental model of porcine intestinal adenomatosis. C s mucosalis colonised the gut of gnotobiotic pigs, persisting for up to 47 days after infection, but did not induce adenomatosis. Despite evidence of limited penetration of the mucosa up to two days after infection, the majority of C s mucosalis remained in the gut lumen. Rotavirus did not enhance invasion of enterocytes by C s mucosalis. The presence of E coli and S bovis caused an increase in the total numbers of C s mucosalis in the gut, but did not affect their distribution. One-day-old poults fed a vitamin D3-deficient diet were examined for clinical, biochemical, and morphological changes at 14 days of age. Changes in these parameters were compared at 15.5 and 17 days of age after one of the following vitamin D-replacement therapies was provided: water-soluble vitamin-mineral packs that contained vitamin D3; vitamin D3 in the feed; or vitamin D, 1,25-dihydroxyvitamin D3 in the feed. The vitamin D3-deficient poults were lame and had significant decreases in weight gain and in longitudinal skeletal growth. None of the therapies alleviated all these changes, but clinical lameness subsided in poults provided 1,25-dihydroxyvitamin D3. Calcium concentrations were significantly improved by all therapies. Treatment with vitamin D3 in the feed and water significantly increased 25-hydroxyvitamin D3 concentrations in vitamin D-deficient poults. The growth plate zones, with the exception of the mineralized hypertrophied zone, were all increased in length and not modified by treatment.
