Adenosine monophosphate-activated protein kinase (AMPK) is a conserved sensor of intracellular energy activated in response to low nutrient availability and environmental stress. In a screen for conserved substrates of AMPK, we identified ULK1 and ULK2, mammalian orthologs of the yeast protein kinase Atg1, which is required for autophagy. Genetic analysis of AMPK or ULK1 in mammalian liver and Caenorhabditis elegans revealed a requirement for these kinases in autophagy. In mammals, loss of AMPK or ULK1 resulted in aberrant accumulation of the autophagy adaptor p62 and defective mitophagy. Reconstitution of ULK1-deficient cells with a mutant ULK1 that cannot be phosphorylated by AMPK revealed that such phosphorylation is required for mitochondrial homeostasis and cell survival during starvation. These findings uncover a conserved biochemical mechanism coupling nutrient status with autophagy and cell survival. 相似文献

The linear polarization of sunlight multiply scattered by the atmosphere, and cloud particles of Venus has been computed and compared with observations over the wavelength range from the ultraviolet to the infrared region. The following properties of the visible cloud layer are derived: the refractive index of the cloud particles is 1.45 +/- 0.02 at a wavelength of 0.55 micron, and there is an indication of a slight decrease in the value from the ultraviolet to the near-infrared region; the mean particle radius is very near 1 micron, and most of the particles are spherical; the cloud layer occurs high in the atmosphere where the pressure is about 50 millibars (equivalent to an altitude of approximately 20 kilometers on the earth). The results for the index of refraction eliminate the possibility that the visible clouds are composed of pure water or ice. 相似文献

We have used the high optical absorptivity of urban and source particulates to trace their "graphitic" component. The optical absorptivity and the particulate carbon loading show a strong correlation. Analyses of the data indicate that primary soot emissions compose a major fraction of the carbonaceous aerosol and put a low limit on secondary organic material produced in correlation with the ozone concentration. 相似文献

Phytophthora lateralis has been isolated from root and collar lesions in Port‐Orford Cedar (POC) trees (Chamaecyparis lawsoniana) in north‐western France (Brittany). These trees, planted in hedgerows, displayed symptoms similar to the typical symptoms of POC root disease. Until now, the disease has been found outside of the nurseries only in western North America. Aerial symptoms, not associated with root or collar infections, were also observed, and P. lateralis was isolated from branch lesions. Similar symptoms were previously observed only in one POC root disease site, located in the Pacific coast of Oregon where climatic conditions are similar to those occurring in Brittany. The reported aetiology as well as the morphological characteristics (deciduous sporangia) of P. lateralis suggests that this species could be air‐dispersed, as described for P. ramorum, a closely related species. This outbreak of P. lateralis in Brittany in farming landscapes associated with the aerial spread of this pathogen represents a new threat for European countries. 相似文献

The purpose of this study was to determine the impact of the non-steroidal anti-inflammatory drug tepoxalin on canine tumour cell growth and describe the changes associated with tepoxalin treatment. In vitro experiments were performed to assess tepoxalin-associated alterations in tumour cell growth. Clinically achievable tepoxalin concentrations did not significantly alter tumour cell growth in vitro. Vascular endothelial growth factor (VEGF) production and hypoxia-inducible factor-1α dose-dependently increased in vitro in the presence of tepoxalin. A canine osteosarcoma xenograft was used to determine in vivo effects of tepoxalin on tumour growth and angiogenesis. Despite increased VEGF in vitro, there was a significant growth delay associated with tepoxalin treatment. Normal dogs were administered tepoxalin to assess effects on systemic VEGF production, but not found to have significantly increased VEGF. These data suggest that tepoxalin may moderately inhibit tumour growth and may be administered as an analgesic to tumour-bearing dogs. 相似文献

Afforestation of sandy arable soils in northern Europe is likely to lead to an increase in the soil's acidity and changes in the behaviour of the organic matter, and this might affect the ability of the soil to retain heavy metals. It is important to assess the impact of such a change in the land use on the solubility of the heavy metals and to assess the risk of leaching to surface‐ and groundwater and the possible entrapment of heavy metals in the tree canopy. The impact of afforestation was assessed by excavating soil profiles in adjacent 34‐year‐old Norway spruce stands and arable plots at four different sites. We found that after 34 years the pH had decreased and cations were depleted in the topsoil under forest. The aqua regia‐extractable heavy metals were determined, and the heavy metal binding within the soil was assessed using a modified version of the BCR (Community Bureau of Reference) sequential extraction procedure. Higher contents of heavy metal were found in the arable plots in the loamy sand soils. Cadmium was found only in the most mobile fractions. The content of Pb in the subsoil was strongly correlated with the clay content, but not in the topsoil, which suggested that Pb had been added to the topsoil. We found strong correlations between the clay content and the Cu, Ni and Zn in the residual fraction, leading us to conclude that much of the Cu, Ni and Zn is of geological origin. No significant differences in the heavy metal fractionation between forest and arable soil were found, presumably because 34 years of different land use is not long enough to produce such differences. 相似文献

Farmyard manure (FYM) and fertilizer applications are important management practices used to improve nutrient status and organic matter in soils and thus to increase crop productivity and carbon (C) sequestration. However, the long-term effects of fertilization on C, nitrogen (N) and sulfur (S) associated with aggregates, especially on S are not fully understood. We investigated the effects of more than 80 years of FYM (medium level of 40 Mg ka−1 and high level of 60 Mg ka−1) and mineral fertilizer (NPKS and NK) on the concentrations and pools of C, N, and S and on their ratios in bulk soil, dry aggregates and water stable aggregates on an Aquic Eutrocryepts soil in South-eastern Norway. A high level of FYM and NPKS application increased the proportion of small dry aggregates (<0.6 mm) by 8%, compared with the control (without fertilizer). However, both medium and high level of FYM application increased the proportion of large water stable aggregates (>2 mm) compared with mineral fertilizer (NPKS and NK). The total C and N pools in bulk soils were also increased in FYM treatments but no such increase was seen with mineral fertilizer treatments. The increased total S pool was only found under high level of FYM application. Water stable macroaggregates (>2 and 1–2 mm) and microaggregates (<0.106 mm) contained higher concentrations of C, N and S than the other aggregate sizes, but due to their abundance, medium size water stable aggregates (0.5–1 mm) contained higher total pools of all three elements. High level of FYM application increased the C concentration in water stable aggregates >2, 0.5–1 and <0.106 mm, and increased the S concentration in most aggregates as compared with unfertilized soils. Higher C/N, C/S and N/S ratios were found both in large dry aggregates (>20 and 6–20 mm) and in the smallest aggregates (<0.6 mm) than in other aggregate sizes. In water stable aggregates, the C/N ratio generally increased with decreasing aggregate size. However, macroaggregates (>2 mm) showed higher N/S ratios than microaggregates (<0.106 mm). We can thus conclude, that long-term application of high amounts of FYM resulted in C, N and S accumulation in bulk soil, and C and S accumulation in most aggregates, but that the accumulation pattern was dependent on aggregate size and the element (C, N and S) considered. 相似文献

Analyses of the spatial and temporal variations in the natural abundance of 13C are frequently employed to study transformations of plant residues and soil organic matter turnover on sites where long continued vegetation with the C3-type photosynthesis pathway has been replaced with a C4-type vegetation (or vice versa). One controversial issue associated with such analyses is the significance of isotopic fractionation during the microbial turnovers of C in complex substrates. To evaluate this issue, C3-soil and quartz sand were amended with maize residues and with faeces from sheep feed exclusively on maize silage. The samples were incubated at 15 °C for 117 days (maize residues) or 224 days (sheep faeces). CO2 evolved during incubation was trapped in NaOH and analysed for C isotopic contents. At the end of incubation, 63 and 50% of the maize C was evolved as CO2 in the soil and sand, respectively, while 32% of the faeces C incubated with soil and with sand was recovered as CO2. Maize and faeces showed a similar decomposition pattern but maize decomposed twice as fast as faeces. The δ13C of faeces was 0.3‰ lower than that of the maize residue (δ13C −13.4‰), while the δ13C of the C3-soil used for incubation was −31.6‰. The δ13C value of the CO2 recovered from unamended C3-soil was similar or slightly lower (up to −1.5‰) than that of the C3-soil itself except for an initial flush of 13C enriched CO2. The δ13C values of the CO2 from sand-based incubations typically ranged −15‰ to −17‰, i.e. around −3‰ lower than the δ13C measured for maize and faeces. Our study clearly demonstrates that the decomposition of complex substrates is associated with isotopic fractionation, causing evolved CO2 to be depleted in 13C relative to substrates. Two field trials with spring barley (Hordeum vulgare L.) were conducted at two locations in Denmark in order to evaluate the effects of tillage and growth of a catch crop on yield parameters under temperate coastal climate conditions. Ploughing in autumn or spring in combination with perennial ryegrass (Lolium perenne L.) as a catch crop was evaluated on a coarse sand (Orthic Haplohumod) from 1987 to 1992 at three rates of N fertiliser application (60, 90 and 120 kg N ha−1 year−1). Rotovating and direct drilling were also included as additional tillage practices. The experiment was conducted on a 19-year-old field trial with continuous production of spring barley. Ploughing in autumn or spring in combination with stubble cultivation and a catch crop, in addition to minimum tillage, was evaluated in a newly established field trial on a sandy loam (Typic Agrudalf) from 1988 to 1992. Yield parameters and N concentrations in grain and straw were determined. On the coarse sand, N uptake in the grain in ploughed plots without a catch crop was significantly greater when spring ploughed as opposed to autumn ploughed, but grain and straw yields did not differ significantly. Grain yield, straw yield and total N uptake did not differ significantly between direct drilled and autumn ploughed plots, but the trend was for grain yield to be lower with direct drilling. After 19 years of catch crop use, yield parameters in ploughed plots were greater than in plots without catch crops. This was most pronounced in the autumn ploughed plots. Rotovating the catch crop in the spring decreased grain yield compared with underploughing the catch crop in autumn or spring. No significant interactions were found between tillage and N rates. On the sandy loam, grain as well as straw yield and total N uptake were not significantly affected by catch crop or time of ploughing.
