Two isolates of Trichoderma viride (L4 and S17A) were assessed for biological control of Allium white rot (AWR) with different onion accessions and cultivars, alone and in combination with a tebuconazole-based seed treatment or composted onion waste. In glasshouse tests, 23 new bulb-onion accessions from previous work to detect resistance to Sclerotium cepivorum showed no differences in susceptibility to AWR but, when combined with S17A, disease was reduced by up to two-thirds over all accessions. Trichoderma viride L4 and S17A also reduced the proportion of infected plants for five commercial bulb-onion cultivars and one advanced breeding line by at least one-third. Further glasshouse tests using a salad-onion cultivar showed that L4, S17A, tebuconazole or composted onion waste controlled AWR and at least halved the proportion of diseased plants. Combination treatments of T. viride with either tebuconazole or compost enhanced control and, in some treatments, disease was almost eliminated. In field trials, control of AWR by S17A was significant for 17 out of 18 individual or mixed bulb-onion accessions, with disease reduced overall by more than half. In another field experiment, S17A failed significantly to reduce AWR for two out of three commercial bulb-onion cultivars, while tebuconazole reduced the final proportion of AWR-infected plants over all cultivars from 0·47 to 0·09. Combining S17A and tebuconazole resulted in a similar level of AWR to using tebuconazole alone. The use of T. viride in an integrated strategy with other treatments to enhance control of S. cepivorum is discussed.  相似文献

The polymerase chain reaction (PCR) technique was utilized to obtain internal transcribed spacer ribosomal DNA (ITS rDNA) and small-subunit (18S) rDNA sequences from UK isolates of Spongospora subterranea f.sp. nasturtii , a plasmodiophorid pathogen of watercress ( Rorippa nasturtium-aquaticum ). ITS sequence data obtained from S. subterranea isolated from a range of UK sites were found to be identical. PCR primers were designed using these sequences and were shown to be capable of specific amplification of S. subterranea f.sp. nasturtii DNA from plant tissue and from water samples containing zoospores of the pathogen. As little as 5 ng total genomic DNA from infected plant material, or 1000 zoospores, was required for consistently successful amplification of DNA. A filtration-based method for obtaining pathogen DNA for PCR from watercress-bed water was developed.  相似文献

A non-invasive imaging modality, computed tomography (CT), was used to visualize changes in nasal turbinates of anesthetized pigs over a 12-week observation period (pigs were 14 wk of age at study week 0). Normal, non-infected pigs were compared to pigs with mild challenge-induced atrophic rhinitis (AR) in order to detect subtle differences in morphology. To determine feasibility for time course studies in future experiments, morphometric quantitation at the level of the 2nd premolar (turbinate area ratio or TAR) in cross-section CT images at multiple timepoints was done. Additionally, at study termination, the TAR determined from CT images, magnetic resonance imaging (MRI), and wet tissue (WT), were compared to each other and to the standard subjective measure, visual scoring. There were no statistically significant differences between the control and AR groups at CT imaging dates of 0, 3, 6, 9, or 12 wk (P = 0.182). However, a statistically significant decrease in TAR measurements over time (P = 0.015) was observed in both groups, with lower mean values observed on Weeks 3 and 6 before rebounding to baseline values at study termination. At Week 12 (termination of the study), the TAR measurements derived from CT, MRI, and WT were not statistically different from one another (P = 0.220) and the treatment group-by-method interaction was not significant (P = 0.800). This provided evidence of equivalency of the techniques. Mean values for normal and infected groups were not significantly different based on either TAR imaging methods (P = 0.552) or visual scores (P = 0.088). Thus, the current study demonstrated that CT was an acceptable alternative imaging modality which could be used for quantitation of turbinate changes in snouts of live pigs to provide data comparable to tissue taken at necropsy. Computed tomographic imaging would allow non-invasive tracking of disease or treatment responses within individual animals over time. Morphometric analysis of the TAR was equivalent between the CT, MRI, and WT specimens.  相似文献

Outbreaks of diarrhoea in two groups of Saanen.kids aged 4 to 6 weeks were ascribed to giardiasis, although Cryptosporidia were also present. In all, 45 out of 65 kids were affected, two animals died, and the remainder eventually recovered. Aspects of the epidemiology and diagnosis of giardiasis are discussed. Air dried smears prepared from small intenstinal mucosa, intestinal contents, or freshly-voided faeces proved to be suitable for making a diagnosis, and are the recommended sample when laboratory examination will be delayed beyond a few hours.  相似文献

The environmental factors that influence infection of lettuce by ascospores of Sclerotinia sclerotiorum , and subsequent disease development, were investigated in controlled environment and field conditions. When lettuce plants were inoculated with a suspension of ascospores in water or with dry ascospores and exposed to a range of wetness durations or relative humidities at different temperatures, all plants developed disease but there was no relationship between leaf wetness duration or humidity and percentage of diseased plants. Ascospores started to germinate on lettuce leaves after 2–4 h of continuous leaf wetness at optimum temperatures of 15–25°C. The rate of development of sclerotinia disease and the final percentage of plants affected after 50 days were greatest at 16–27°C, with disease symptoms first observed 7–9 days after inoculation, and maximum final disease levels of 96%. At lower temperatures, 8–11°C, disease was first observed 20–26 days after inoculation, with maximum final disease levels of 10%. Disease symptoms were always observed first at the stem base. In field-grown lettuce in Norfolk, 2000 and 2001, inoculated with ascospore suspensions, disease occurred only in lettuce planted in May and June, with a range of 20–49% of plants with disease by 8 weeks after inoculation. In naturally infected field-grown lettuce in Cheshire, 2000, disease occurred mainly in lettuce planted throughout May, with a maximum of 31% lettuce diseased within one planting, but subsequent plantings had little (≤ 4%) or no disease. Lack of disease in the later plantings in both Norfolk and Cheshire could not be attributed to differences in weather factors.  相似文献

Fusarium proliferatum is a globally distributed fungal pathogen that affects a range of crop hosts and is one of the main producers of mycotoxins, such as fumonisins, in foods. Specific PCR primers are commonly used for detection and identification of this pathogen. The aim of this study was to validate previously published F. proliferatum-specific primers targeting the intergenic spacer (IGS) region and characterize intraspecific variation and homologous recombination events for isolates obtained from diseased garlic bulbs in Spain. Sixty-nine isolates were morphologically identified as F. proliferatum, and their identity was confirmed by sequencing of the translation elongation factor; however, specific IGS primers did not result in an amplification product for nine of these isolates. Further analysis showed that this was due to polymorphism in the IGS region and six isolates were classified as IGS type I, while the remaining isolates were type II. Sequencing of the complete IGS region revealed numerous sequence polymorphisms amongst F. proliferatum isolates, and regions of recombination. Duplication and deletion events may have occurred via unequal crossing over during mitotic or meiotic recombination. These results suggest that the IGS region may be too variable as a reliable target for F. proliferatum-specific identification.  相似文献
