Fasciola hepatica NP-40 released protein extract (FhNPE) exhibits potent Th1 immunosuppressive properties in vitro and in vivo. However, the protein composition of this active fraction, responsible for Th1 immune modulatory activity, has yet to be resolved. Therefore, FhNPE, a Nonidet P-40 extract, was subjected to a proteomic analysis in order to identify individual protein components. This was performed using an in house F. hepatica EST database following 2D electrophoresis combined with de novo sequencing based mass spectrometry. The identified proteins, a mixture of excretory/secretory and membrane-associated proteins, are associated with stress response and chaperoning, energy metabolism and cytoskeletal components. The immune modulatory properties of these identified protein(s) are discussed and HSP70 from F. hepatica is highlighted as a potential host immune modulator for future study.  相似文献

The degradation of organic macromolecules, including lignin, in plant-derived soil organic matter, is important to the global carbon cycle. In grasslands, saprotrophic (decomposer) fungi are major decomposers of such organic material. The aim of this study was to characterise lignin degradation, particularly with respect to lignin oxidation typical of white-rot basidiomycete fungi. Lignin breakdown products, analysed by gas chromatography–mass spectrometry (GC–MS) with TMAH thermochemolysis, in initial wheat (Triticum aestivum var. Swatham) straw samples were compared with those in samples which had been buried as a “model” resource for 46 months in a sand-dune grassland at Ainsdale National Nature Reserve, Lancashire, UK.Our results showed that lignin oxidation occurred in the straw over the 46 month period, as there were general increases in the [Ac/Al]S and [Ac/Al]G ratios and a clear decrease in the [S/G] ratio. These data provide tentative support for the theory that white-rot basidiomycete fungi are involved in the degradation of lignin in grasslands.  相似文献

Cutover peatlands are often rapidly colonised by pioneer plant species, which have the potential to affect key ecosystem processes such as carbon (C) turnover. The aim of this study was to investigate how plant cover and litter type affect fungal community structure and litter decomposition in a cutover peatland. Intact cores containing Eriophorum vaginatum, Eriophorum angustifolium, Calluna vulgaris and bare soil were removed and a mesh bag with litter from only one of each of these species or fragments of the moss Sphagnum auriculatum was added to each core in a factorial design. The presence or absence of live plants, regardless of the species, had no effect on mass loss, C, nitrogen (N) or phosphorus (P) concentrations of the litter following 12 months of incubation. However, there was a very strong effect of litter type on mass loss and concentrations of C, N and P between most combinations of litter. Similarly, plant species did not affect fungal community structure but litter type had a strong effect, with significant differences between most pairs of litter types. The data suggest that labile C inputs via rhizodeposition from a range of plant functional types that have colonised cutover bogs for 10-15 years have little direct effect on nutrient turnover from plant litter and in shaping litter fungal community structure. In contrast, the chemistry of the litter they produce has much stronger and varied effects on decomposition and fungal community composition. Thus it appears that there is distinct niche differentiation between the fungal communities involved in turnover of litter versus rhizodeposits in the early phases of plant succession on regenerating cutover peatlands.  相似文献

This paper details the evolution of activism against genetically modified organisms on the Big Island of Hawai’i. It offers an explanation for the ability of rural residents on the Big Island to pass anti-GMO legislation while other states and communities have tried and failed. I argue that the Big Island’s recent anti-GMO legislative success is due to the articulation of interests and actions between settlers to Hawai’i and Native Hawaiian community members seeking to protect Native Hawaiian rights. Tracing the history of the anti-GMO movement on Big Island highlights the unique circumstances that facilitated the passage of this bill, and is also significant for making sense of the potential future trajectories of anti-GMO-related food sovereignty movements elsewhere.  相似文献
