The aim of this study was to investigate how three vascular plant species (Calluna vulgaris, Eriophorum angustifolium and Eriophorum vaginatum) colonising an abandoned cutover peatland affect fluxes of recent photosynthate to dissolved organic carbon (DOC), soil and plant respiration and shoot biomass. We used in situ 13CO2 pulse labelling to trace carbon (C) throughout a 65 day pulse chase period. Between 16 and 35% of the pulse of 13C remained in shoot biomass after 65 days with significant differences between C. vulgaris and E. angustifolium (P = 0.009) and between C. vulgaris and E. vaginatum (P = 0.04). A maximum of 29% was detected in DOC beneath labelled plants and losses of 13C from peat respiration never exceeded 0.16% of the original pulse, showing that little newly fixed C was allocated to this pool. There were no significant differences between the different plant species with respect to 13C recovered from DOC or via peat respiration. More C was lost via shoot respiration; although amounts varied between the three plant species, with 4.94–27.33% of the 13C pulse respired by the end of the experiment. Significant differences in 13C recovered from shoot respiration were found between C. vulgaris and E. angustifolium (P = 0.001) and between E. angustifolium and E. vaginatum (P = 0.032). Analysis of δ13C of microbial biomass indicated that recently assimilated C was allocated to this pool within 1 day of pulse labelling but there were no significant differences in the 13C enrichment of the microbial biomass associated with the different plant species. The data suggest that peat respiration represents a small flux of recent assimilate compared to other fluxes and pools and that different vascular plant species show considerable variation in the quantities and dynamics of C allocated to DOC.  相似文献

Common bean is one of the widely consumed food security crop in Africa, Asia and South America. It is a rich source of protein, minerals and micronutrients. High genotype by environment interaction is one of the main challenge in breeding for high grain micronutrient concentration. The objective of this study was to estimate SNP markers associated with grain Fe and Zn concentration using 289 common bean genotypes and 11,480 SNP markers. The study revealed that 43 quantitative trait loci (QTLs) were associated with grain Fe and Zn concentration. Five quantitative trait nucleotides (QTNs), that is, QTN Fe_1.1, QTN Fe_6.3, QTN Fe_6.5, QTN Fe_10.3 and QTN Fe_11.6 were detected both at Haramaya and Melkassa locations. Two of the markers, that is, QTN Fe_6.3 and QTN Fe_6.5, were located on chromosome 06 while QTN Fe_1.1, QTN Fe_10.3 and QTN Fe_ 11.6 were residing on chromosomes 01, 10 and 11, respectively. Among these, QTN Fe_11.6 had a large and positive consistent effect across locations. The five stable QTNs along with the potential candidate genes could be used for Fe biofortification through marker assisted selection.  相似文献

The fungal community in coniferous forest soils plays a pivotal role in ecosystem processes such as decomposition and carbon and nutrient cycling. Both saprotrophic (SP) and ectomycorrhizal (ECM) fungi occur throughout the upper soil horizons in coniferous forests and could therefore be exposed to high concentrations of monoterpenes occurring in the needle litter and roots of some tree species. Previous work has noted the differential effects of monoterpenes on the mycelial growth of a range of both SP and ECM fungi when grown in artificial nutrient media. This study used a novel experimental system to assess the effect of environmentally relevant concentrations of monoterpenes on the activity of ECM and SP fungi grown on more natural substrata. Exposure of the ECM fungus Paxillus involutus (Batsch) Fr. to vapours of either α-pinene or β-pinene resulted in a significantly greater proportion of root tips being colonised by the fungus when it was grown with seedlings of Picea abies (L.) Karst. Exposure to monoterpenes resulted in a significant decrease in respiration rate of two species of litter degrading SP fungi, Mycena galopus var. candida J. E. Lange and Collybia butyracea (Bull.) P. Kumm. There was no difference in response between the two SP species, despite the fact that previous tests in liquid nutrient media, with monoterpenes at higher concentrations, indicated that one species was sensitive and one was not. The high volatility and low solubility of monoterpenes in water make them challenging to work with. The experimental system developed here, although still artificial, provides a bridge between pure culture studies in defined media and all the complexities of forest soils in the field, by allowing the exposure of fungi to environmentally relevant monoterpene concentrations in more natural substrata.  相似文献

The use of indicators in soil monitoring schemes to detect changes in soil quality is receiving increased attention, particularly the application of soil biological methods. However, to date, the ability to compare information from different laboratories applying soil microbiological techniques in broad-scale monitoring has rarely been taken into account. This study aimed to assess the consistency and repeatability of two techniques that are being evaluated for use as microbiological indicators of soil quality: multi-enzyme activity assay and multiple substrate-induced respiration (MSIR). Data were tested for intrinsic (within-assay plate) variation, inter-laboratory repeatability (geometric mean regression and correlation coefficient) and land-use discrimination (principal components analysis). Intrinsic variation was large for both assays suggesting that high replicate numbers are required. Inter-laboratory repeatability showed diverging patterns for the enzyme assay and MSIR. Discrimination of soils was significant for both techniques with relatively consistent patterns; however, combined laboratory discrimination analyses for each technique showed inconsistent correspondence between the laboratories. These issues could be addressed through the adoption of reliable analytical standards for biological methods along with adequate replication. However, until the former is addressed, dispersed analyses are not currently advisable for monitoring schemes.  相似文献

Browsing by mammalian herbivores is a major problem in plantation forestry worldwide. This has traditionally been controlled through a reduction in herbivore numbers achieved by lethal means, namely shooting and poisoning. In recent years, however, there has been increasing social and political pressure to reduce reliance on such lethal controls, and consequently research into non-lethal controls is becoming increasingly important. In order for non-lethal methods to be efficiently incorporated into management, however, we must first identify those that are most effective under operational conditions. Different methods are rarely tested concurrently, and many have only been proven effective with captive animals or in small scale field trials. This study therefore sought to consolidate past research by combining non-lethal methods, which were known to significantly reduce browsing damage, into a single trial to enable their relative effectiveness to be compared under operational conditions.  相似文献

Interest in and initiation of farm-to-school (FTS) programs have increased in recent years, spurred on by converging public concerns about child obesity trends and risks associated with industrialization and distancing in the modern food system. A civic agriculture framework that more specifically considers civic engagement and problem solving offers insights about variations in the development and prospects for FTS programs. Drawing on comparative case studies of two emerging FTS initiatives in Pennsylvania—one in a rural setting and one in an urban setting—this article examines the role of internal and external “champions” in launching FTS programs and fostering civic engagement. Farm-to-school community stakeholders across the two cases framed FTS in broadly similar terms of (1) redressing poor food environments; (2) improving student nutrition, health and well-being; and (3) revitalizing rural community through support of local agriculture. However, specific concerns and emphases differed across the rural and urban cases, illustrating the significance of local context for such programs. The article concludes by discussing the importance of frame bridging and frame extension as strategies for expanding the FTS movement, and also ensuring programs that correspond to the specific circumstances and possibilities of their social and geographic settings.  相似文献

Surface exploration is a key step in the colonization of surfaces by sessile marine biofoulers. As many biofouling organisms can delay settlement until a suitable surface is encountered, colonization can comprise surface exploration and intermittent swimming. As such, the process is best followed in three dimensions. Here we present a low-cost transportable stereoscopic system consisting of two consumer camcorders. We apply this novel apparatus to behavioral analysis of barnacle larvae (≈800 μm length) during surface exploration and extract and analyze the three-dimensional patterns of movement. The resolution of the system and the accuracy of position determination are characterized. As a first practical result, three-dimensional swimming trajectories of the cypris larva of the barnacle Semibalanus balanoides are recorded in the vicinity of a glass surface and close to PEG2000-OH and C(11)NMe(3)(+)Cl(-) terminated self-assembled monolayers. Although less frequently used in biofouling experiments due to its short reproductive season, the selected model species [Marechal and Hellio (2011), Int Biodeterior Biodegrad, 65(1):92-101] has been used following a number of recent investigations on the settlement behavior on chemically different surfaces [Aldred et al. (2011), ACS Appl Mater Interfaces, 3(6):2085-2091]. Experiments were scheduled to match the availability of cyprids off the north east coast of England so that natural material could be used. In order to demonstrate the biological applicability of the system, analysis of parameters such as swimming direction, swimming velocity and swimming angle are performed.  相似文献

Rodents are globally important pre‐harvest pests of rice. In Southeast Asia, rodent damage to growing rice crops is commonly concentrated towards the center of rice fields, away from the field edge, resulting in a clear pattern known as the “stadium effect.” To further understand this behavior of rodent pests and to develop recommendations for future research and management, we examined the relation between giving‐up densities (GUDs) and damage patterns. In Tanay, Luzon, Philippines, GUD trays containing pieces of coconut in a matrix of sand were placed at 4 different distances from the field edge to quantify the perceived risk of predation in a rice field pest, Rattus tanezumi. GUDs were recorded during a dry and wet season crop at the reproductive and ripening stages of rice. In addition, assessments of active burrows, tracking tile activity and rodent damage to the rice crop, were conducted in the dry season. GUDs were significantly lower in the center of the rice fields than on the field edges, suggesting that rodent damage to rice is greater in the middle of rice fields due to a lower perceived predation risk. Furthermore, this perception of predation risk (or fear) increases towards the field edge and was greatest on the rice bund, where there was no vegetation cover. We discuss the implications for rodent management and rodent damage assessments in rice fields. This is the first documented use of GUDs in a rice agro‐ecosystem in Asia; thus we identify the challenges and lessons learned through this process.  相似文献
