Amplified fragment length polymorphism for variety identification and genetic diversity assessment in oleander (Nerium oleander L.) 总被引:1,自引:0,他引:1

Oleander is a Mediterranean evergreen shrub found along watercourses, gravelly places and damp slopes. It is grown widely
as an ornamental for its abundant and long-lasting flowering as well as its moderate hardiness. Genetic relatedness among
71 accessions, including commercial varieties, different sources of the same varieties, and selections from the wild were
investigated using amplified fragment length polymorphism (AFLP). Nine primer combinations yielded a total of 603 bands of
which 241 were polymorphic. Genetic similarities among accessions were calculated according to Jaccard's Similarity Index
and used to construct a dendrogram based on the unweighted pair group method using arithmetic averages. Our results show that
the AFLP technique, which can simultaneously and assay a large number of loci randomly distributed in the genome, is much
more informative on the genetic relationship and origin of accessions than the limited number of morphological characters
conventionally used for variety discrimination. Up to about 9% molecular genetic differentiation was detected among morphologically
indistinguishable provenances of the same variety; this can be partly attributed to scoring error but mainly to somatic variation
occurring during vegetative propagation. On the other hand lower genetic distance values were sometimes found among varieties
which differ in morphological characters and are thus commercialised with different names. The possibility of considering
the amount of genetic variation within a variety as the threshold value for discrimination of initial varieties and essentially
derived varieties is discussed.

An EDTA‐anticoagulated blood sample collected from a 1.5‐year‐old, intact male, English Bulldog was submitted for a CBC. The CBC data and blood smear evaluation revealed borderline high hematocrit (54%, reference interval 37–55%), inappropriate rubricytosis, moderate leukopenia due to both mature neutropenia and lymphopenia, and mild thrombocytopenia. Numerous leukocytes showed evidence of karyolysis, pyknosis, and karyorhexis, and apoptotic bodies were frequent in the background. Many neutrophils had botryoid nuclei characterized by increased numbers of nuclear segments radially arranged with spoke‐like, delicate chromatin filaments connecting the segments centrally. The finding of botryoid nuclei and inappropriate rubricytosis was indicative of severe hyperthermia, such as heatstroke. The dog had been exercised a long time during conditions of high temperature and humidity until he collapsed. The dog was diagnosed with severe heatstroke, hypovolemic shock, disseminated intravascular coagulation, and multiorgan dysfunction syndrome. Despite aggressive treatment, the patient died of cardiopulmonary arrest. Botryoid nuclei are frequent in people with heatstroke. In the authors' experience, botryoid nuclei are seen commonly in dogs with heatstroke, but they have never been reported in veterinary medicine. The presence of petechiation with only mild thrombocytopenia and inappropriate rubricytosis also is suggestive of heatstroke and manifests ongoing life‐threatening vascular derangement. 相似文献

The aim of this work was to evaluate the analysis of DNA microsatellites for the detection of soft wheat (Triticum aestivum L.) in semolina and durum wheat bread (prepared from Triticum turgidum L. var. durum). The results enabled selection of an efficient D-genome-specific repetitive DNA sequence to detect common wheat in semolina and breads by qualitative PCR with a threshold of 3 and 5%, respectively, lowered to 2.5% by real-time PCR. This is of major importance for checking during production of some typical products recently awarded the European Protected Designation of Origin (PDO) mark such as Altamura bread, which should not contain soft wheat flour. The feasibility of quantification of common wheat adulteration in semolina using real-time PCR was also demonstrated. 相似文献

Hydrophobic polypyrrole-coated fabrics with improved electrical conductivity were produced embedding oleic acid as counter-ion. Hydrophobisation of polypyrrole was carried out by means of an ion exchange process after deposition of polypyrrole on cotton fabrics. The fabrics coated with oleic acid-doped polypyrrole showed contact angle of 111°, drop absorption time of 7 minutes and high water repellence, while electrical conductivity increased of ~2 times and heat generation improved, too. Moreover, oleic acid demonstrated a great stability as counter-ion in polypyrrole matrix being present also after washing. 相似文献

Ghrelin is involved in many biological processes, ranging from appetite regulation and the release of growth hormone to the
regulation of gastrointestinal motility and secretion processes. Ghrelin expression is not homogenously distributed throughout
the gastrointestinal tract; expression is species-specific and can also depend on the animal age. This study was performed
to investigate ghrelin immunolocalization in the gastrointestinal tract of pigs at different ages: 1 day (birth), 28 days
(weaning), 2 months, 4 months, and 7 months (pre-puberty). Tissue samples were collected along the entire gastrointestinal
tract and were examined by immunohistochemistry and double-immunofluorescence. Histometry was performed by counting the number
of endocrine ghrelin immunopositive cells in the gastrointestinal mucosa. Ghrelin was found to be present along the swine
alimentary canal from the stomach to the caecum. In all regions of the alimentary canal of the animals studied, ghrelin-immunoreactive
(IR) cells co-localized with chromogranin-A and were therefore identified as endocrine cells. In the gastric fundus, ghrelin-immunoreactivity
was partially detected in co-localization with H-K-adenosine triphosphatase and pepsinogen. Ghrelin-IR endocrine cells were
abundant in the oxyntic mucosa but less present in the small intestine and rare in the large intestine. The cell density of
the ghrelin-IR endocrine cells was lowest in the oxyntic mucosa of 1-day-old pigs. We can conclude that gastric ghrelin expression
is not related merely to age but could also potentially be influenced by food intake. 相似文献

The paper describes a low-invasive experimental apparatus designed for the accurate determination of mechanical deformation
and moisture content (MC) relationship on macroscopic wood samples. The device is particularly indicated for monitoring wooden
handwork whose mechanical deformation is especially critical in relation to its role such as, for example, works of art or
architectural works. The MC of wood is measured with a portable single-sided NMR probe and mechanical deformation by a fiber
Bragg grating optical sensor. The data obtained are of high accuracy, despite the dimensions of the sample. The methodology
provides an effective tool for investigating the dynamic relation between environmental relative humidity, MC, and shrinking–swelling
of wood. Adsorption results collected for longitudinal deformation in silver fir (Abies alba Mill.) are presented to show the sensitivity of the optical sensor. Interesting findings include the detection of two different
mechanisms of elongation and the time evolution of water mobility versus hydration and strain. 相似文献

A collection of 70 olive samples, originating from diverse areas in central-southern Italy (Abruzzo, Apulia, Calabria, and Umbria) and corresponding to 3 major cultivars denominations (‘Carolea’, ‘Coratina’ and ‘Frantoio’), was genotyped at 10 microsatellite loci. In total, 44 alleles with a mean number of 4.4 alleles per locus were detected. The molecular analysis, allowed the study to show a clear genetic diversity between the three cultivars ‘Carolea’, ‘Coratina’ and ‘Frantoio’ and to state that ‘Carolea’ is a polyclonal cultivar, while ‘Coratina’ and ‘Frantoio’, are probably monoclonal ones. The analysis of intra-varietal polymorphism, through the SSR analysis, proved to be very useful both for varietal identification and for intra-varietal ones. Our work shows that the current designations of olive cultivars fall short of describing the genetic variability among economically important plant material. A thorough investigation of the existing variability will prove of major importance for both management and economic production of olive trees. 相似文献
